Insulin-like growth factors induce apoptosis as well as proliferation in LIM 1215 colon cancer cells 总被引:2,自引:0,他引:2
The insulin-like growth factor (IGF) system plays an important role in cell proliferation and survival. However, more recently, a small number of studies have shown that IGFs induce apoptosis in some cells. Our initial studies showed this occurred in LIM 1215 colon cancer cells but not RD rhabdomyosarcoma cells. IGFs induced both proliferation and apoptosis in LIM 1215 cells, and the induction of apoptosis was dose-dependent. [R54, R55]IGF-II, which binds to the IGF-I receptor with normal affinity but does not bind to the IGF-II receptor, induced apoptosis to the same extent as IGF-II, whereas [L27]IGF-II, which binds to the IGF-I receptor with 1000-fold reduced affinity, had no effect on apoptosis. These results suggest that the IGF-I receptor is involved in induction of apoptosis. Western blot analyses demonstrated that Akt and Erk1/2 were constitutively activated in RD cells. In contrast, phosphorylation of Akt and Erk1/2 were transient and basal expression of Akt protein was lower in LIM 1215 cells. Analysis of apoptosis-related proteins showed that IGFs decreased pro-caspase-3 levels and increased expression of pro-apoptotic Bad in LIM 1215 cells. IGFs co-activate proliferative and apoptotic pathways in LIM 1215 cells, which may contribute to increased cell turnover. Since high turnover correlates with poor prognosis in colorectal cancer, this study provides further evidence for the role of the IGF system in its progression. 相似文献

In fast growing organisms, like fish larvae, fatty acids provided through live prey are essential to satisfy high energy demand and are required to promote growth. Therefore, in recent decades, a great amount of research has been directed towards the development of lipid enrichment in order to improve larval fish survival and growth. However, in fish, the biochemical and molecular processes related to highly unsaturated fatty acid (HUFA) administration are still poorly understood. In the current study, the false percula clownfish, a short larval phase marine species, was used as an experimental model and the effects of a standard and a HUFAs-enriched diet were tested through a molecular, biochemical, ultrastructural and morphometric approach. Our results support the hypothesis that HUFA administration may improve larval development through the presence of better structured mitochondria, a higher synthesis of energy compounds and coenzymes with a central position in the metabolism, with respect to controls. This higher energy status was confirmed by better growth performance and a shorter larval phase in larvae fed with an enriched diet with respect to the control. This strategy of rapid growth and early energy storage may be considered positively adaptive and beneficial to the survival of this species. 相似文献

Myostatin (MSTN) is a well-known negative regulator of skeletal muscle development. Reduced expression due to natural mutations in the coding region and knockout as well as knockdown of MSTN results in an increase in the muscle mass. In the present study, we demonstrated as high as 60 and 52% downregulation (p?0.01) of MSTN mRNA and protein in the primary fetal myoblast cells of goats using synthetic shRNAs (n?=?3), without any interferon response. We, for the first time, evaluated the effect of MSTN knockdown on the expression of MRFs (namely, MyoD, Myf5), follistatin (FST), and IGFs (IGF-1 &; IGF-2) in goat myoblast cells. MSTN knockdown caused an upregulation (p?0.05) of MyoD and downregulation (p?0.01) of MYf5 and FST expression. Moreover, we report up to ~four fold (p?0.001) enhanced proliferation in myoblasts after four days of culture. The anti-MSTN shRNA demonstrated in the present study could be used for the production of transgenic goats to increase the muscle mass. 相似文献
