L-Arabinose has been described as a gratuitous inducer of the yeast -galactosidase. This has been found to be an artefact resulting from galactose contamination of commercial samples of L-arabinose. The inactivation produced on UDP-glucose 4-epimerase by the pentose does not amplify the inducer activity of contaminating D-galactose.  相似文献

MicroRNAs (miRNAs) have been established to regulate skeletal muscle development in mammals. However, few studies have been conducted on the regulation of proliferation and differentiation of bovine myoblast cells by miRNAs. The aim of our study was to explore the function of miR-483 in cell proliferation and differentiation of bovine myoblast. Here, we found that miR-483 declined in both proliferation and differentiation stages of bovine myoblast cells. During the proliferation phase, the overexpression of miR-483 downregulated the cell cycle–associated genes cyclin-dependent kinase 2 (CDK2), proliferating cell nuclear antigen (PCNA) messenger RNA (mRNA), and the protein levels. At the cellular level, cell cycle, cell counting kit-8, and 5-ethynyl-2´-deoxyuridine results indicated that the overexpression of miR-483 block cell proliferation. During differentiation, the overexpression of miR-483 led to a decrease in the levels of the myogenic marker genes MyoD1 and MyoG mRNA and protein. Furthermore, the immunofluorescence analysis results showed that the number of MyHC-positive myotubes was reduced. In contrast, the opposite experimental results were obtained concerning both proliferation and differentiation after the inhibition of miR-483. Mechanistically, we demonstrated that miR-483 target insulin-like growth factor 1 (IGF1) and downregulated the expression of key proteins in the PI3K/AKT signaling pathway. Altogether, our findings indicate that miR-483 acts as a negative regulator of bovine myoblast cell proliferation and differentiation.  相似文献

The main purpose of the International Atomic Energy Agency (IAEA) Public Awareness Strategies for Tissue Banks is to provide guidance on organizing and running awareness campaigns, in order to consolidate tissue banking activities. Within the IAEA Public Awareness Strategies for Tissue Banks, there are two important topics, which need to be singled out due to their importance for a successful public and professional awareness campaign. These are the selection of the audiences and the organization of media events within a Communication Strategy. The experience in the field of tissue banking in several countries has shown that interaction between the public, the professional health care staff, the media and the tissue bank personnel is essential if the activities of the banks are to be successful. It must be emphasized however, that any public and professional awareness strategy will not be successful, unless it is considered as part of an integrated system that is adopted by the concerned Government.  相似文献

The National University Hospital (NUH) Tissue Bank was established in October 1988. The National University of Singapore (NUS) was officially appointed by IAEA to be the IAEA/NUS Regional Training Centre (RTC) for RCA Member States for training of tissue bank operators on September 18, 1996. In the first five years since its establishment the National University Hospital Tissue Bank concentrated its work on the sterile procurement and production of deep frozen femoral heads and were used in patients for bone reconstruction. The cost of producing these tissues were about SGD$ 250 per femoral head although cost fees were initially charged at SGD$ 50 per femoral head. The most important activity carried out by Singapore within the IAEA was training. Between November 1997 and April 2007, a total of nine courses were conducted by RTC with a total of 180 tissue bank operators registered, 133 from Asia and the Pacific region (13 countries, including 2 from Iran), 14 from Africa (Zambia, Libya, Egypt, Algeria, and South Africa), 6 from Latin America (Brazil, Chile, Cuba, Peru, and Uruguay), 9 from Europe (Greece, Slovakia, Poland, and Ukraine), and 2 from Australia. The last batch (ninth batch) involved 20 students registered in April 2007 and will be due to sit for the terminal examination in April 2008.  相似文献

The first multi-tissue bank was founded at Havana in 1958. At that time, freeze-drying was used at the bank as a method of preserving, as well as Cobalt 60 irradiation to sterilise bone tissue, heart valves and others. The impact of the IAEA program in tissue banking activities in Cuba can be summarised as follows: (a) Increase in the production of sterilised tissues using ionising radiation (bone, pig skin and amnion) for medical treatment in the tissue bank of the Hospital Frank Pais; (b) increase of the quality of the productions of bone tissues, pig skin and amnion; (c) reduction in the import of tissues by increasing the local production of tissues; (d) sustainability in the number of donors through the implementation of a public and professional awareness campaign; (e) training of six persons in the Regional Training Centre of Buenos Aires; (f) qualification of one person in the administration of a tissue bank and in the implementation of a Quality System. The amount of tissues produced and sterilised using the ionising radiation techniques in the established banks was 25,510 units. The amount of patients treated with sterilised tissues produced by the established banks was 2,448.  相似文献

The hyperactivation of the mechanistic target of rapamycin complex 1 (mTORC1) in chondrocytes has been shown to accelerate the severity of destabilization of the medial meniscus-induced and age-related osteoarthritis (OA) phenotypes with aberrant chondrocyte hypertrophy and angiogenesis. Meanwhile, we previously reported that miR-483-5p is essential for the initiation and development of OA by stimulating chondrocyte hypertrophy and angiogenesis. The connection between mTORC1 and miR-483-5p activation in OA progression, however, remains unclear. In this study, we elucidated their relationship and identified the underlying mechanisms. The expression of miR-483-5p in the articular cartilage of cartilage-specific TSC1 knockout mice was assessed compared with control mice using the Agilent Mouse miRNA (8*60K) V19.0 array and real-time polymerase chain reaction (RT-PCR). The functional effects of the stimulation of miR-483-5p via histone deacetylase 4 (HDAC4) by mTORC1 in OA development, subsequently modulating its downstream targets matrilin 3 and tissue inhibitor of metalloproteinase 2, were examined by immunostaining, western blotting, and real-time PCR. This study revealed that miR-483-5p was responsible for mTORC1 activation-stimulated OA. Mechanistically, mTORC1 controls the HDAC4-dependent expression of miR-483-5p to stimulate chondrocyte hypertrophy, extracellular matrix degradation, and subchondral bone angiogenesis, and it consequently initiates and accelerates the development of OA. Our findings revealed a novel mTORC1-HDAC4-miR-483-5p pathway that is critical for OA development.  相似文献

Tissue banking started in Thailand in 1979. Five years after this, the Bangkok Biomaterial Centre (BBC) was established in the Faculty of Medicine, Siriraj Hospital, with the support of the IAEA program. The objective of the Centre was to provide sterile bones and tissues for clinical use. Through the passage of time, the Bangkok Biomaterial Centre has gained confidence from the end user and by 2007 has processed 33,872 allografts from 491 deceased donors and 4,035 live donors were used in medical treatment in 3,596 patients in more than 79 different hospitals. More than 305 surgeons from Thailand used the tissue produced in the BBC. At the beginning of its work the BBC concentrate its activities on the production of the following tissues: freeze dried bone, freeze dried dura mater and freeze dried fascia lata. All of these tissues were sterilised using ethylene oxide gas until the end of year 1984. Since 1985 the BBC sterilise tissue using ionising radiation. The BBC is now producing deep-frozen; bone tendon, cartilage, trachea and soft tissue; freeze-dried; bone, fascia lata, dura mater, amniotic membrane, bone hydroxyapatite, bone tablet and fresh preserved amniotic membrane Yongyudh Vajaradul is a Founder of Bangkok Biomaterial Centre and also a President of TATB, Bangkok, Thailand. BackgroundThis work aims to provide a simulated method to be used by designers of medical accelerators and in clinical centers to manage and minimize particles’ interaction in the patient-dependent part of a 6 MV X-Ray Beam generated by the Elekta linear accelerator system, based on the latest GATE software version 9.0 Monte Carlo simulation, IAEA phase space data, and the last version of “Slurm” computing cluster.Materials and methodsThe experimental results are obtained using the Elekta 6 MV accelerator. The simulation MC developed includes the majority of the patient-dependent segments, such as Multi-Leaf Collimator (MLC), Tongue and Groove T&G, Rounded leaf Part, including the Jaws (XY). This model is used, with a simulated Iba Blue Phantom 2 homogeneous water phantom with dimensions 480 × 480 × 410 mm3, positioned at a Source-to-Surface-Distance (SSD) of 100 cm, all of the interactions of the mega voltage 6 MV radiations in water are simulated. The IAEA phase space (PS) provided by the International Atomic Energy Agency database and cluster computing (Slurm HPC-MARWAN, CNRST, Morocco) are employed to reduce our simulation time.ResultsThe results confirm that there are many interactions in all areas and the patient-dependent part’s internal structures. Thus, electrons and positrons participation appear in the generated field previously designed to be an X-ray beam. Besides, to validate our implementation geometry, the PDD’s and transverse profiles, at a depth ranging from 1.5 to 20 cm, for a field size of 10 × 10 cm2, the beam quality such as D10%, dmax (cm), d80 (cm), TPR(20/10), the two relative differences in dose were derived on σi and σi,max are calculated, respectively. Additionally, gamma index formalism for 2%/2 mm criteria is used. Once and for all, we typically take a good agreement between simulation MC GATE 9.0 and the experiment data with an error less than 2%/2 mm.ConclusionsIn the field of X-ray photons, a significant contribution of electrons and positrons has been found. This contribution could be enough to be essential or affect the delivered dose. A good agreement of 98% between this new approach of simulation MC GATE 9.0 software based on IAEA phase space and experimental dose distributions is observed regarding the validation tests used in this task.  相似文献
