The effect of irradiation on the in vitro growth of Plasmodium falciparum was investigated. The cultured malarial parasites at selected stages of development were exposed to gamma rays and the sensitivity of each stage was determined. The stages most sensitive to irradiation were the ring forms and the early trophozoites; late trophozoites were relatively insensitive. The greatest resistance was shown when parasites were irradiated at a time of transition from the late trophozoite and schizont stages to young ring forms. The characteristics of radiosensitive variation in the parasite cycle resembled that of mammalian cells. Growth curves of parasites exposed to doses of irradiation upto 150 gray had the same slope as nonirradiated controls but parasites which were exposed to 200 gray exhibited a growth curve which was less steep than that for parasites in other groups. Less than 10 organisms survived from the 106 parasites exposed to this high dose of irradiation; the possibility exists of obtaining radiation-attenuated P. falciparum. 相似文献

Summary The influence of repair and replication on the frequency of spontaneous chromosome aberrations and of those induced by gamma-irradiation is reported.Using the technique of labelling DNA with radioactive 3H-thymidine and measuring the radioactivity of DNA isolated from embryos, the time of initiation and the duration of DNA synthesis in barley seeds was studied after the soaking of the seeds had begun. The average duration of each phase of the first DNA synthesis cycle in soaking barley seeds was found to be as follows: pre-DNA synthesis stage, 10–11 hrs; DNA synthesis stage, 8 hrs. After gamma-irradiation, the intensity of DNA synthesis decreased and the beginning of DNA synthesis was delayed.It was found that the inhibition of repair by caffeine led to an increase in the frequency of both spontaneous and induced chromosome aberrations. Caffeine enhanced several times the frequency of chromosome and chromatid aberrations at the time of the maximal activity of repair enzymes. During DNA replication, caffeine had a lower effect on the realization of premutational lesions.An inhibitor of DNA replication — hydroxyurea — had no influence on the frequency of spontaneous chromosome aberrations during the replication period, whereas after gamma-irradiation, hydroxyurea enhanced the frequency of aberrations mainly at the stage of DNA replication.The relatively small mutagenic action of both agents (caffeine and hydroxyurea) was observed during all stages of the cell cycle of germinating barley seeds. 相似文献

The incorporation of [14C]deoxycytidine, [3H]deoxyuridine, and [3H]thymidine, respectively into pyrimidine bases of DNA has been measured in rapidly proliferating P815 mouse mastocytoma cells in the presence of hydroxyurea. The incorporation of [14C]deoxycytidine-derived radioactivity into DNA cytosines is increased when compared to the incorporation into DNA thymines. The [3H]deoxyuridine-derived radioactivity is incorporated solely into DNA thymines and this incorporation is inhibited by hydroxyurea in a dose-dependent manner. This suggests an inhibitory effect of hydroxyurea on the thymidylate synthase which was proved in experiments in which the conversion of deoxyuridine monophosphate into deoxythymidine monophosphate catalysed by a crude enzyme preparation from P815 cells was inhibited in the presence of hydroxyurea. Enzymatic DNA methylation as measured by the conversion of incorporated [14C]deoxycytidine into 5-methylcytosines was not affected by hydroxyurea. 相似文献

NDP reductase activity can be inhibited either by treatment with hydroxyurea or by incubation of an nrdA
ts mutant strain at the non-permissive temperature. Both methods inhibit replication, but experiments on these two types of
inhibition yielded very different results. The chemical treatment immediately inhibited DNA synthesis but did not affect the
cell and nucleoid appearance, while the incubation of an nrdA101 mutant strain at the non-permissive temperature inhibited DNA synthesis after more than 50 min, and resulted in aberrant
chromosome segregation, long filaments, and a high frequency of anucleate cells. These phenotypes are not induced by SOS.
In view of these results, we suggest there is an indirect relationship between NDP reductase and the chromosome segregation
machinery through the maintenance of the proposed replication hyperstructure. 相似文献

The nucleolar structure and the activity of NopA100, a nucleolin-like protein, during the cell cycle in proliferating plant cells 总被引:2,自引:2,他引:0
For the purpose of gaining knowledge of the relationships between cell proliferation and ribosome biogenesis, as two fundamental
mutually interconnected cellular processes, studies were performed on cell populations synchronized in their cell-cycle progression
by treatment with hydroxyurea, followed by sampling at different times after its removal. A structural rearrangement of the
nucleolus was observed throughout the interphase, along with changes in the relative amounts of different nucleolar subcomponents.
A structural model of nucleolar organization was associated with each interphase period. Throughout interphase, the nucleolin-like
protein, NopA100, was immunodetected in the dense fibrillar component of the nucleolus, preferentially near fibrillar centers
and its levels were shown to increase from G1 to G2. A western blotting analysis of soluble nuclear protein extracts with
anti-NopA100 antibody resulted in the intense labeling of a 100-kDa band, but also of a series of proteins related to it,
suggesting that NopA100 undergoes a physiological process of proteolytic maturation, similar to that described for mammalian
nucleolin, but not reported in other biological model systems. Physiological proteolysis of NopA100, related to cell-cycle
progression, was confirmed after the nuclei extracted from synchronized cells were treated with the protease inhibitor, leupeptin,
which resulted in an increase of the 100-kDa band at the expenses of the decrease of some other bands, according to the cell-cycle
stages. We therefore conclude that there is a relationship between the increase in nucleolar activity, cell-cycle progression,
nucleolar structure, the activity of NopA100, and the proteolysis of this nucleolin-like protein. 相似文献

Anti-metabolites, such as methotrexate, 5-fluoropyrimidines or hydroxyurea, induced progressive formation of DNA lesions. 5-Fluoropyrimidines induce DNA lesions either by incorporation of the drug into DNA or by a mechanism not involving incorporation. The second mechanism, not involving incorporation, is also seen with methotrexate and hydroxyurea. The three anti-metabolites have in common their ability to reduce intracellular levels of nucleotides, resulting in reduced efficiency of repair of DNA lesions. The lesions probably appear spontaneously, independently of the drug treatment. 相似文献

Hydroxyurea, when injected intraperitoneally, exerted marked inhibition on the activity of thymidine kinase in 5 day old postnatal
cerebellum and 15 day old embryonic cerebrum. However, it failed to show any sustained inhibition on thymidine kinase activity
in 5 day old postnatal cerebrum. In this case, the marginal decrease of thymidine kinase activity noticed during early intervals
reversed back to more than normal value at a later time interval. These results along with our earlier findings are taken
to indicate the differential action of this drug on thymidine kinase activity in rapidly and slowly proliferating regions
