We studied the relevance of urine cues in Homarus gammarus dominance maintenance, hypothesising that urinary signals are necessary to mediate recognition of former opponents. Males in size-matched pairs interacted on two consecutive days with or without blocking urine release by adding catheters to both contestants on the second day. European lobsters established dominance in a first fight, and fight duration and aggression levels decreased strongly from first to second day in animals with free urine release, indicating the maintenance of this dominance relationship. If urine was blocked on the second day, fight durations were long in both first and second day interactions. Results demonstrate that urine signals contribute to the maintenance of dominance in H. gammarus males.  相似文献

Summary

A solid phase binding assay was developed to study the vitellogenin binding sites from solubilized Homarus americanus oocyte membrane. Different detergents (SDS, CHAPS, DOC) were tested and DOC (sodium deoxycholate) was found to be the most effective agent. The solid phase binding assay involves an adsorption of solubilized membranes in wells of microtitration plates. Enzyme labelling of the ligand was realized by coupling glutaraldehyde treated peroxidase with purified vitellin. Scatchard analysis after competition experiments in different conditions (time and temperature) revealed an apparent equilibrium dissociation constant (Kd) close to 70 nM, reached after one hour incubation at 37°C. Binding activity of oocyte membranes is maximal at the beginning of vitellogenesis and decreases in older oocytes.  相似文献

The lobster (Homarus americanus) hepato-pancreatic epithelial baso-lateral cell membrane possesses three transport proteins that transfer calcium between the cytoplasm and hemolymph: an ATP-dependent calcium ATPase, a sodium-calcium exchanger, and a verapamil-sensitive cation channel. We used standard centrifugation methods to prepare purified hepato-pancreatic baso-lateral membrane vesicles and a rapid filtration procedure to investigate whether 65Zn2+ transfer across this epithelial cell border occurs by any of these previously described transporters for calcium. Baso-lateral membrane vesicles were osmotically reactive and exhibited a time course of uptake that was linear for 10–15 s and approached equilibrium by 120 s. In the absence of sodium, 65Zn2+ influx was a hyperbolic function of external zinc concentration and followed the Michaelis-Menten equation for carrier transport. This carrier transport was stimulated by the addition of 150 M ATP (increase in Km and Jmax) and inhibited by the simultaneous presence of 150 mol l–1 ATP+250 mol l–1 vanadate (decrease in both Km and Jmax). In the absence of ATP, 65Zn2+ influx was a sigmoidal function of preloaded vesicular sodium concentration (0, 5, 10, 20, 30, 45, and 75 mmol l–1) and exhibited a Hill Coefficient of 4.03±1.14, consistent with the exchange of 3 Na+/1Zn2+. Using Dixon analysis, calcium was shown to be a competitive inhibitor of baso-lateral membrane vesicle 65Zn2+ influx by both the ATP-dependent (Ki=205 nmol l–1 Ca2+) and sodium-dependent (Ki=2.47 mol l–1 Ca2+) transport processes. These results suggest that zinc transport across the lobster hepato-pancreatic baso-lateral membrane largely occurred by the ATP-dependent calcium ATPase and sodium-calcium exchanger carrier proteins.Communicated by: I.D. The torque generated by a rotating joint comprises the useful force exerted by the joint on the external environment, and both the magnitude and distribution of torque through the step cycle during walking are important variables in understanding the mechanics of walking. The mechanics of the American lobster (Homarus americanus) and snow crab (Chionoecetes opilio) during walking were modelled to examine the relative roles of flexor versus extensor apodeme-muscle complexes, investigate which legs of these decapods likely contribute the greatest to locomotion, determine scaling effects of torque generation, and assess the relative roles of various model variables on torque production. Force generated along the length of the apodeme by the muscle was modelled based on apodeme surface area, muscle stress, and muscle fibre pinnation angle. Torque was then calculated from this estimated force and the corresponding moment arm. The flexor apodeme-muscle complex is calculated to generate consistently greater forces than the extensor, and generally this results in flexor torque being larger than extensor, though the snow crab does illustrate the opposite in two of its legs. This greater torque generation in flexion suggests that, in addition to the pushing of the trailing legs, the pulling action of the leading legs may play a significant role, at least during lateral walking. Leg 4 of both species appears to generate greater torques and thus provide the greatest forces for locomotion. Torque generation as a function of body size shows a second order response due to the increase in apodeme surface area. The pinnation angle of the muscle fibre is found to be insignificant in force generation, apodeme surface area (representing muscle cross sectional area) likely plays the most influential role in total force production, and moment arm controls the distribution of this force through the step cycle. Muscle stress remain a largely unknown quantity however, and may significantly affect both magnitude and distribution through step cycle of forces, and thus torque. Despite the uncertainty associated with the muscle stress parameter, the modelled results fit well with previously published force measurements.  相似文献

Despite being one of the most productive fisheries in the Northwest Atlantic, much remains unknown about the natural reproductive dynamics of American lobsters. Recent work in exploited crustacean populations (crabs and lobsters) suggests that there are circumstances where mature females are unable to achieve their full reproductive potential due to sperm limitation. To examine this possibility in different regions of the American lobster fishery, a reliable and noninvasive method was developed for sampling large numbers of female lobsters at sea. This method involves inserting a blunt-tipped needle into the female''s seminal receptacle to determine the presence or absence of a sperm plug and to withdraw a sample that can be examined for the presence of sperm. A series of control studies were conducted at the dock and in the laboratory to test the reliability of this technique. These efforts entailed sampling 294 female lobsters to confirm that the presence of a sperm plug was a reliable indicator of sperm within the receptacle and thus, mating. This paper details the methodology and the results obtained from a subset of the total females sampled. Of the 230 female lobsters sampled from George''s Bank and Cape Ann, MA (size range = 71-145 mm in carapace length), 90.3% were positive for sperm. Potential explanations for the absence of sperm in some females include: immaturity (lack of physiological maturity), breakdown of the sperm plug after being used to fertilize a clutch of eggs, and lack of mating activity. The surveys indicate that this technique for examining the mating success of female lobsters is a reliable proxy that can be used in the field to document reproductive activity in natural populations.  相似文献

Summary We used a polyclonal antiserum against histamine to map histaminelike immunoreactivity (HLI) in whole mounts of the segmental ganglia and stomatogastric ganglion of crayfish and lobster. Carbodiimide fixation permitted both HRP-conjugated and FITC-conjugated secondary antibodies to be used effectively to visualize HLI in these whole mounts. Two interneurons that send axons through the inferior ventricular nerve (ivn) and the stomatogastric nerve to the stomatogastric ganglion had strong HLI, both in crayfish and in lobster. These ivn interneurons were known from other evidence to be histaminergic. The neuropil of the stomatogastric ganglion in both crayfish and lobster contained brightly labeled terminals of axons that entered the ganglion from the stomatogastric nerve. No neuronal cell bodies in this ganglion had HLI. Each segmental ganglion contained at least one pair of neurons with HLI. Some neurons in the subesophageal ganglion and in each thoracic ganglion labeled very brightly. Axons of projection interneurons with strong HLI occurred in the dorsal lateral tracts of each segmental ganglion, and sent branches to the lateral neuropils and tract neuropils of each ganglion. All the labeled neurons were interneurons; no HLI was observed in peripheral nerves.  相似文献
