In this study, the presence of actin in cultured trypanosomatids was investigated using polyclonal antibodies to heterologous actin. Polyclonal antisera to rabbit muscle actin and a monospecific anti-actin antibody react with a 43-kDa polypeptide in extracts of Trypanosoma cruzi, Herpetomonas samuelpessoai and Leishmania mexicana amazonensis on protein immunoblots. The 43-kDa polypeptide co-migrates with skeletal muscle actin and is retained within trypanosomatid cytoskeletons. Attempts to isolate H. samuelpessoai actin through DNase I affinity chromatography showed that the 43-kDa polypeptide did not bind to the column. Instead, low yields of a 47-kDa polypeptide were obtained indicating that the trypanosomatid actin displays unusual DNase I binding behavior when compared to actins from higher eukaryotes. Immunofluorescence studies confirmed that cytoskeletons retain the actin-like protein. In H. samuelpessoai , actin is localized in the region close to the flagellum, whereas in T. cruzi it is more homogeneously distributed. The data presented here show that trypanosomatid actin displays biochemical characteristics similar to actins of other protozoa.  相似文献

SYNOPSIS Previous surveys of feral Drosophila melanogaster in Clarke County, Georgia, indicated 10 to 30% of the flies were parasitized with Herpetomonas ampelophilae. In the present study, herpetomoniasis in D. melanogaster reared in population cages rose from 0.01% to 90% in 14 days. The infection fluctuated between 5% and 100% during the 250 days in which the ageledeme was maintained.
Herpetomonas were found endotrophically, peritrophically, and in the Malpighian tubules. In population cages, although there were sporadic rises in percentages, peritrophic and Malpighian tube infections usually remained low. Endotrophic infections often reached 100%, with individual flies being heavily parasitized. Although Chatton felt that several species of Herpetomonas inhabit a single species of Drosophila , we suggest that all the Herpetomonas in D. melanogaster be referred to as H. ampelophilae.  相似文献

Abstract

A trypanosomatid flagellate, Herpetomonas sp., is recorded from an adult Mythimna separata (Walker) (Lepidoptera: Noctuidae). Promastigote, paramastigote, and opisthomastigote stages measured 3.4–12 × 1.3–4.3 μm, Giemsastained, with flagella measuring 13.8–18.7 μm. A few amastigotes (3.9–8.3 × 3.7–6.7 μm) were also observed. Fourteen larvae fed with a suspension of flagellates were found later to be infected with the protozoan. No flagellate infections were found in the hymenopteran parasitoid, Apanteles ruficus (Haliday), or the hyperparasitoid, Trichomalopsis sp.  相似文献

In this work, we present the sequences and a comparison of the glycosomal GAPDHs from a number of Kinetoplastida. The complete gene sequences have been determined for some species (Crithidia fasciculata, Herpetomonas samuelpessoai, Leptomonas seymouri, and Phytomonas sp), whereas for other species (Trypanosoma brucei gambiense, Trypanosoma congolense, Trypanosoma vivax, and Leishmania major), only partial sequences have been obtained by PCR amplification. The structure of all available glycosomal GAPDH genes was analyzed in detail. Considerable variations were observed in both their nucleotide composition and their codon usage. The GC content varies between 64.4% in L. seymouri and 49.5% in the previously sequenced GAPDH gene from Trypanoplasma borreli. A highly biased codon usage was found in C. fasciculata, with only 34 triplets used, whereas in T. borreli 57 codons were employed. No obvious correlation could be observed between the codon usage and either the nucleotide composition or the level of gene expression. The glycosomal GAPDH is a very well-conserved enzyme. The maximal overall difference observed in the amino acid sequences is only 25%. Specific insertions and extensions are retained in all sequences. The residues involved in catalysis, substrate, and inorganic phosphate binding are fully conserved, whereas some variability is observed in the cofactor-binding pocket. The implications of these data for the design of new trypanocidal drugs targeted against GAPDH are discussed. All available gene and amino acid sequences of glycosomal GAPDHs were used for a phylogenetic analysis. The division of the Kinetoplastida into two suborders, Bodonina and Trypanosomatina, was well supported. The literature shows that the effects of direct electric currents on biological material are numerous, including bactericidal, fungicidal, parasiticidal, and anti‐tumoral, among others. Non‐pathogenic trypanosomatids, such as Herpetomonas samuelpessoai, have emerged as important models for the study of basic biological processes performed by a eukaryotic cell. The present study reports a dose‐dependent anti‐protozoan effect of direct electric treatment with both cathodic and anodic current flows on H. samuelpessoai cells. The damaging effects can be attributable to the electrolysis products generated during electric stimulation. The pH of the cell suspension was progressively augmented from 7.4 to 10.5 after the cathodic treatment. In contrast, the anodic treatment caused a pH decrease varying from 7.4 to 6.5. Transmission electron microscopy analyses revealed profound alterations in vital cellular structures (e.g., mitochondrion, kinetoplast, flagellum, flagellar pocket, nucleus, and plasma membrane) after exposure to both cathodic and anodic current flows. Specifically, cathodic current flow treatment induced the appearance of autophagic‐like structures on parasite cells, while those submitted to an anodic current flow presented marked disorganization of plasma membrane and necrotic appearance. However, parasites treated in the intermediary chamber (without contact with the electrodes) did not present significant changes in viability or morphology, and no pH variation was detected in this system. The use of H. samuelpessoai as a biological model and the direct electric current experimental approach used in our study provide important information for understanding the mechanisms involved in the cytotoxic effects of this physical agent. SYNOPSIS Herpetomonas sp. was found repeatedly in the Malpighian tubules of laboratory-reared male and female Aedes aegypti and Aedes albopictus in Malaysia. The flagellates occurred irregularly, in batches, and were absent during long periods. The data suggest an exogenous source of infection for the mosquitoes, presumably from another insect, probably of another genus. Thirty to 40% of the flagellates of Aedes contained intracytoplasmic rod-shaped structures strongly resembling bacteria. These were found often in groups suggesting intracellular multiplication. They were passed to the Herpetomonas daughter cells during division. Parasitism of Aedes by Herpetomonas is extremely unusual, only one previous record, an inconclusive one, having been found. Parasitism by Herpetomonas containing bacterium-like rods has apparently never been reported.  相似文献

SYNOPSIS. The kinetoplast nucleoid of Herpetomonas muscarum muscarum is a disk-shaped mass of filaments with sharply delimited anterior and posterior margins. The kinetoplast nucleoid of Herpetomonas muscarum ingenoplastis is elongate, tapered, and made up of long, helically arranged filaments. Possible explanations of these structures on the basis of work by others on isolated kinetoplast DNA are given. The double flagellum of Herpetomonas is seen by electron microscope to be two complete flagella in close contact but with no visible connection to each other.  相似文献

Ergosterol was the only sterol detected in Herpetomonas samuelpessoai grown in a defined, lipid-free medium. When cultivated in a complex medium, this flagellate was found to contain 6 additional sterols. As measured by incorporation of L-[Me-14C]methionine, in the absence of acetate, the sterol synthesis was greater at 28 C than at 37 C; in the presence of acetate, however, this synthesis was greater at 37 C. When [2-14C]acetate was used as the sterol precursor, the synthesis level at 37 C exceeded that at 28 C.  相似文献

Herpetomonas roitmani, a non-pathogenic trypanosomatid was grown in chemically defined media either containing proline or glucose as carbon source. Using transmission electron microscopy we observed that cells grown in the presence of proline present more lipid inclusions, and a larger mitochondrion with more cristae and higher activity of succinate cytochrome c reductase. On the other hand, cells grown with glucose as carbon source had more glycosomes, which were preferentially located close to the bacterium endosymbiont, and a much higher activity of hexokinase, a typical glycosome marker. Three-dimensional reconstruction and morphometrical analysis confirm these observations. The number of promastigotes of H. roitmani increased in the presence of proline. Taken together these results indicate that the growth conditions markedly influenced the ultrastructure and the metabolism of H. roitmani.  相似文献
