Changes in phenolic metabolism after elicitation with Colletotrichum gloeosporioides (CG) has been studied in Hypericum perforatum L. (HP) cell suspension cultures. Soluble phenolics were analysed by HPLC-DAD and HPLC-DAD-MS/MS. HP cultures elicited with the CG elicitor showed a significant increase in xanthone accumulation. Xanthone accumulation increased twelve fold when the cells were primed with methyl-jasmonate (MeJ) or salicylic acid (SA), before elicitation. HP cultures exposed only to MeJ produced a set of flavonoids, the flavones which represent a substantial part (approx. 40%) of the total flavonoids accumulated in these cells. The possible importance of xanthones as a component of defence mechanism of HP against biotic stress is discussed.  相似文献

Spontaneous forms of hemp (Cannabis sativa L., often reported as Cannabis sativa var. spontanea Vavilov ) with a low content of psychoactive cannabinoids can be considered as a valuable source of other phytoconstituents to be used in nutraceuticals or for their health promoting properties. Chemical data on this hemp variety are rather scarce. In this article, we report a comprehensive phytochemical characterization of leaves, male and female inflorescences of C. sativa growing wild in Hungary. For the purpose, the essential oil along with polar extracts were analyzed using GC/MS, NMR and LC‐DAD‐MS techniques, respectively. The results indicated that female inflorescence essential oil contains high amounts of the CB2 agonists, (E)‐caryophyllene (28.3 %) and cannabidiol (CBD; 24.9 %), whereas leaves and male inflorescence essential oils contained lower amounts of both compounds. HPLC/MS allowed to quantify cannabidiol (CBD) and cannabidiolic acid (CBD?A) in the ethyl acetate extracts from leaves, male and female inflorescences; they were 0.3, 0.8 and 0.9 %, and 0.2, 0.3 and 0.4 %, respectively. Flavonoids of this spontaneous form of hemp were formed by C‐glycosides and glucuronic acids of kaempferol and apigenin with a total content of 3.8, 6.1 and 7.8 mg/g in methanolic extracts from leaves, male and female inflorescences, respectively. Based on these results, spontaneous C. sativa may represent an important source of CB2 agonists and bioflavonoids to be used in nutraceuticals, cosmetics and pharmaceuticals.  相似文献

Galinsoga ciliata Raf. Blake like Galinsoga parviflora Cav., comes from the Andes region. The chemical composition, activity and use are similar for both species. Galinsoga species are used in folk medicine as anti-inflammatory agents and accelerators for wound healing. Extracts are applied topically onto the skin to treat dermatological diseases, eczemas, lichens and hard-healing-wounds, and also to treat snakebites. Orally they used to cure flu and colds.In the studies using HPTLC method, different stationary phases, including unmodified silica gel, silica gels modified with CN, NH2, DIOL and RP18 groups were tried. The best separation of the tested compounds was achieved on silica gel plates, when as mobile phases mixtures – ethyl acetate–acetic acid–formic acid–water (100:11:11:26, v/v/v/v), ethyl acetate–methanol–formic acid–water (50:3:4:6, v/v/v/v) and ethyl acetate–methyl ethyl ketone–formic acid–water (30:9:3:3, v/v/v/v) – were used. Using reference substances, in the examined extracts the presence of flavonoids: patulitrin, quercimeritrin, quercitagetrin, and phenolic acids – caffeic and chlorogenic acids was found.HPLC analyses of extracts were carried out on a reversed-phase Zorbax SB column (150 mm × 2.1 mm, 1.9 μm). The mobile phase (A) was water/acetonitrile/formic acid (95:5:0.1, v/v/v) and the mobile phase (B) was acetonitrile/formic acid (100:0.1, v/v). A linear gradient system was used: 0–30 min, 1–30% B. Application of HPLC-DAD-MS method confirmed the results obtained by HPTLC method. Moreover, in the tested extracts the presence of caffeoylglucaric acids as dominating compounds was detected.  相似文献

Urtica dentata Hand (UDH) is traditionally used in the Alpine region as a herbal medicine. Immunotherapy using total coumarins (TC) of UDH has been proposed, yet the cellular and molecular mechanisms remain incompletely characterized. Additionally, there is no method available for the quantification of the main coumarins in UDH. We describe maturation-resistant, TC-conditioned dendritic cell (DC), which expressed much lower MHC class II (I-Ak) and CD86, showed reduced capacity to stimulate effector T cell responses and upregulated PD-Ll (programmed death ligand-1). TC-DC-stimulated regulatory cells (Treg) were superior alloantigen-specific suppressor of the T effector response as compared to those stimulated by control (CTR)-DC. Furthermore, TC-conditioned DC increased the levels of Foxp3 and CTLA-4 in the CD25 T cell population. TC-DC downregulated toll like receptor 4 (TLR4) protein expression in response to LPS. This indicates that down-regulation of TLR4 in response to TC on DC is a critical signaling pathway that regulates the phenotype and function of DC. We also established a sensitive and specific high-performance liquid chromatography-diodearray detection-mass spectrometry (HPLC-DAD-MS) method for simultaneous identification of its main coumarins, 6,6',7,7'-tetramethoxyl-8,8'-biscoumarin (1), 7,7'-dihydroxy-6,6'-dimethoxy-8,8'-biscoumarin (2), 7,7'-dimethoxy-6,6'-biscoumarin (3), and scoparone (4). A demonstration of this mechanism and the method for identification and quantification of TC in UDH endorsed their potential as a tolerance-promoting herbal medicine to prevent or treat transplantation rejection and autoimmune diseases.  相似文献

普洱熟茶以其特殊的香气、口感和保健功效,近年来受到愈来愈多消费者的喜爱,是最受欢迎的茶叶品种之一.普洱熟茶由普洱生茶经特殊的微生物后发酵过程生产而成,而普洱生茶则以大叶茶的鲜叶为原料,是一种不发酵的绿茶.本文采用HPLC-DAD-MS技术,对普洱熟茶的化学成分进行了分析,从中共检测出21个峰;通过[M+H]+、[M-H]和碎片离子峰,以及与标准品对比,其中18个峰的化学结构分别得到了鉴定.研究结果显示,去除了大量存在的咖啡因后,HPLC-DAD-MS技术可以用于快速有效地检测普洱熟茶中的化学成分.  相似文献
