Gymnodinium aeruginosum has the usual fine structure of a dinoflagellate but does not seem to contain a well elaborated peduncle or a microtubular basket. Naked cells are surrounded by a single large amphiesmal vesicle. It houses an endosymbiont with typical blue-green cryptophycean chloroplasts (generally only one), cryptophycean starch grains in the periplastidal cytoplasm without a nucleomorph, and two membranes separating the periplastidal cytoplasm from the cryptophycean cytoplasm which contains mitochondria, ER, vesicles and ribosomes, but no eukaryotic nucleus. The endosymbiont is surrounded by a single membrane. Possible ways of the acquisition of the endosymbiont and the problem of the existence of ribosomes within a compartment without nucleus are discussed.Devoted to Prof. Dr.L. An ammonia limited chemostat culture of Gymnodinium simplex (Lohm.) Kofoid & Swezy was perturbed with ammonia and fluctuations in the free intracellular amino acid pools were followed 80 min. The steady-state value of glutamate was 2.07 ± 10-15 mol cell-1 and of glutamine was 0.31 ± 10-15 mol cell-1. Five minutes after the perturbation, a substantial rise in glutamine was observed with a corresponding decrease in glutamate. This is considered a result of glutamine synthetase acting as the primary ammonia assimilating enzyme. The level of ammonia and the major free amino acids reached a maximum 10 min after the perturbation and then slowly decreased.  相似文献

Gymnodimium breve Davis, an unarmored marine dinoflagellate has a cell covering (theca) composed of four membranes. The inner two membranes represent a vesicular layer and in tangential section, the theca appears composed of polygonal areas. Unusual threat ridges are located in the cingular region between the epi- and hypocone. This osmotically sensitive species is extremely vesiculate with dispersed areas of cytoplasm containing typical eukaryotic organelles as well as other organelles found only in dinoflagellates. The non-vesiculated cytoplasm is continuous in serial sections. The chloroplasts can contain either quasi-radial or parallel lamellae typically consisting of three thylakoids each. The pyrenoid is multiple-stalked and lacks a starch cap. The dinophycean pusule is simple and similar to those found in several unarmored marine species. The nucleus is typically dinophycean but the chromosomes appear to lack nonfibrillar material.  相似文献

The role played by heterotrophic microplankton in the synthesis and flux of organic matter was studied in the Punta San Juan Coastal upwelling region off Peru in April and May 1977. The data from a drogue study show that the main component of the planktonic community in the freshly upwelled water is the microheterotroph component. The biomass of bacteria (49 mg C/ m3) in the newly upwelled water exceeded by two orders of magnitude the biomass of phytoplankton. Total respiration of the microheterotrophs (3.35 g C/ m2/ day) exceeded by three-fold the primary production, indicating that the heterotrophic respiration was dependent on the content of organic matter preexisting in the upwelling waters. In the upwelling at Punta San Juan the biomass of protozoa was 1 g/ m3 (wet weight) at the depth of the maximum concentration; this concentration is the highest ever observed in sea water. During transects on a section normal to the coastline an abundant population of dinoflagellates (5 to 40 × 103 /1) of the genera Gymnodinium and Prorocentrum were found in anoxic waters at 50 to 100 m. Strong red tide water coloration was observed as a result of a bloom of the autotrophic ciliate Mesodinium rubrum; the biomass of the ciliate at the surface in calm weather reached 50 to 70 g/ m3 (wet weight) and the cell density was 2 to 4 × 106/l.  相似文献

The potential of ballast water to act as a major introduction vector for toxic dinoflagellates and other phytoplankton is beyond doubt; however, evidence that links the suspected introduced species with a source population is less convincing, especially without supporting historical and biochemical data, or consideration of palaeobiogeographical scenarios that may explain current species distributions. This paper presents new molecular data based on LSU-rDNA and rDNA-ITS sequences that demonstrate an unequivocal and recent link between Temperate Asian and Australasian populations of the toxic dinoflagellates Gymnodinium catenatum and toxic strains of the Alexandriumtamarensis complex”. We integrate our data with supporting evidence from historical distribution records, sediment dating studies, toxin profiles, mating studies and previous molecular studies. We contrast the observed patterns of genetic and biochemical variation with those expected from various palaeobiogeographical scenarios explaining the evolution and natural dispersal of both species. While definitive proof is impossible, the total evidence indicates that these toxic dinoflagellates were introduced to Australasia during the past 100 years, most probably via ballast water from bulk-cargo shipping from Japan and/or south-east Asia.  相似文献

We isolated 2 virus-like agents that suppressed growth of Gymnodinium mikimotoi from coastal waters of the Uwa Sea, Japan. The agents found in the flagellate cells, named GM6 and GM7, were filterable in a 0.22-lm-pore filter with approximately 100-nm shapes. Electron microscopic observation showed the presence of virus-like particles in severely damaged G. mikimotoi cells infected by GM6. The growth-suppression activity of the agents (GM6 or GM7) was lost by heating at 50°C, with treatments of DNase and protease, and filtration through a 0.05-lm filter. Our results suggest that the agents are DNA viruses infectious to and virulent for G. mikimotoi. This is the first report of a virus-like agent specific to G. mikimotoi.  相似文献

Gyrodinium estuariale Hulburt has ultrastructural features typical for a dinoflagellate including a particularly well developed pusule and numerous prominent trichocysts. The amphiesma is relatively simple with thecal vesicles containing thecal membranes. Gymnodinium catenatum is a bloom forming dinoflagellate that has been known to cause paralytic shellfish poisoning (PSP) in humans. It is being reported with increased frequency around the world, with ballast water transport implicated as a primary vector that may have contributed to its global spread. Major limitations to monitoring and management of its spread are the inability for early, rapid, and accurate detection of G. catenatum in plankton samples. This study explored the feasibility of developing a PCR-based method for specific detection of G. catenatumin cultures and heterogeneous ballast water and environmental samples. Sequence comparison of the large sub unit (LSU) ribosomal DNA locus of several strains and species of dinoflagellates allowed the design of G. catenatum specific PCR primers that are flanked by conserved regions. Assay specificity was validated through screening a range of dinoflagellate cultures, including the morphologically similar and taxonomically closely related species G. nolleri. Amplification of the diagnostic PCR product from all the strains of G. catenatum but not from other species of dinoflagellates tested imply the species specificity of the assay. Sensitivity of the assay to detect cysts in ballast water samples was established by simulated spiked experiments. The assay could detect G. catenatum in all ‘blank’ plankton samples that were spiked with five or more cysts. The assay was used to test environmental samples collected from the Derwent river estuary, Tasmania. Based on the results we conclude that the assay may be utilized in large scale screening of environmental and ballast water samples.  相似文献

Paralytic shellfish poisoning toxin profiles of dinoflagellate cultures of Gymnodinium catenatum Graham from the Yellow and South Seas in Korea were investigated by high performance liquid chromatography fluorometric detection. Strains from the Yellow Sea had predominantly carbamate toxins, while strains from Sujeongri and Chindong in the South Sea contained the N‐sulfocarbamoyl toxins, Cl,2, as major components including the presence of GTX5 and dcSTX in some strains. Toxin profiles from St. Deukryang Bay strains (South Sea) showed both characteristics of those in the South Sea and those in the Yellow Sea. Thirty strains could be divided into three groups based on cluster analysis of toxin compositions. Group I (Yellow Sea strains) was distinguished from Group II (Sujeongri and Chindong strains) by the absence of GTX5 and dcSTX. Group III comprised Deukryang Bay strains. In conclusion, the Yellow Sea and the South Sea were found to have different dinoflagellate populations with different toxin compositions.  相似文献
