Summary The relative hydraulic conductivities of major and minor longitudinal veins, and the apoplastic permeability of the bundle sheaths surrounding all longitudinal and transverse veins were investigated in representatives of the C3, C4/NAD-ME, C4/NAD-ME/PCK intermediate, C4/PCK and C4/NADP-ME photosynthetic types. Using the Hagen-Poiseuille equation and measurements of tracheary element diameters, the number of elements in each vein type and the numbers of each vein type, we calculated that 87–99% of the water flow in a longitudinal direction would be expected to occur in the major veins. The permeability of the mestome sheaths and parenchymatous bundle sheaths surrounding the veins was tested using the negatively-charged, fluorescent dye, trisodium 3-hydroxy-5,8,10-pyrenetrisulfonate (PTS). This dye proved nontoxic to plant tissue at a concentration of 0.5%, according to a deplasmolysis test with onion epidermal strips. The PTS concentration achieved in the tested grass leaves was about 0.035%, well below the toxic limit. When a solution of PTS was fed to the leaves by means of a basal cut, the dye moved into the veins of all orders. From there, it moved outward into the surrounding tissues, indicating that the sheaths surrounding the veins of all orders in all species tested were permeable. Therefore, contrary to previous predictions based on structural observations and some tracer studies, bundle sheaths with suberized cell walls do not function as endodermal layers.  相似文献

The leaf ultrastructure of NADP-malic enzyme type C4 species possessing different anatomical features in the Cyperaceae was examined: types were the Rhynchosporoid type, a normal Kranz type in which mesophyll cells are adjacent to Kranz cells, and Fimbristyloid and Chlorocyperoid types, unusual Kranz types in which nonchlorophyllous mestome sheath intervenes between the two types of green cells. They show structural characteristics basically similar to the NADP-malic enzyme group of C4 grasses, that is, centrifugally located chloroplasts with reduced grana and no increase of mitochondrial frequency in the Kranz cells. However, the Kranz cell chloroplasts of the Fimbristyloid and Chlorocyperoid types exhibit convoluted thylakoid systems and a trend of extensive development of peripheral reticulum, although those of the Rhynchosporoid type do not possess such particular membrane systems. The suberized lamella, probably a barrier for CO2 diffusion, is present in the Kranz cell walls of the Rhynchosporoid type and in the mestome sheath cell walls of the other two types, and tightly surrounds the Kranz cells (sheaths) that are the sites of the decarboxylation of C4 acids. These ultrastructural features are discussed in relation to C4 photosynthetic function.  相似文献

Summary At the end of a 4-year period of gas exchange measurements in a natural stand in the Lower Bavarian Forest, needles of an adult spruce [Picea abies (L.) Karst.] were harvested from two chambers, one with pure air and the other with ambient air. The needles were examined as to their histological properties in the stomatal apparatus and in the bundle sheath. In needles from the polluted air UV absorbance at 280 nm was decreased in the walls of the stomatal apparatus. Simultaneously, the deposition of compounds with an absorption maximum at 310 nm increased within the encrusted plate-like thickenings of the subsidiary cells. The contents of the lumina of hypodermal cells and of the bundle sheath exhibited a greater degree of autofluorescence in ambient-air material than in pure-air leaf organs. Differences between needles exposed to pure and polluted air are gradual. The damaged condition is rare in pure air, common in polluted air. The needles from outside the chambers occupied an intermediate position between pure-air and ambient-air needles. This fact is traced to an unnaturally high pollutant load in the liquid phase of the needle surfaces within the ambient-air chamber because in order to compensate pollutant losses within the system, SO2 and O3 were added even during periods of irrigation. The reduction of absorption capacity at 280 nm in the walls of the stomatal apparatus is attributed to destruction of lignin due to the high reactivity of the pollutants in the liquid phase on the damp needle surface. The importance of delignification with regard to hydroregulation is discussed.  相似文献

Monoclonal antibodies were prepared against two species of Methanomicrobiaceae. Antibody 1A is specific for Methanospirillum hungatei strain JF1 and the determinant it recognizes is expressed on the surface of JF1 cells, where it is exposed and accessible to antibody. The determinant is found in a polypeptide (MW<12,000) in the sheath that covers the bacterial cell; it is not present in Methanospirillum hungatei strain GP1; and it is not expressed on the surface of whole cells of the other 24 methanogenic bacteria tested. It is therefore a marker of strain JF1, consequently, antibody 1A is potentially useful for tracking JF1 and fragments thereof in a variety of samples. Antibody 7A is specific for Methanogenium cariaci JR1c. It did not react with any other methanogen tested, not even with Mg. marisnigri or Ms. hungatei JF1, although these cross-react with Mg. cariaci if tested with polyclonal antisera. In C4 grasses belonging to the NADP-malic enzyme-type subgroup, malate is considered to be the predominant C4 acid metabolized during C4 photosynthesis, and the bundle sheath cell chloroplasts contain very little photosystem-II (PSII) activity. The present studies showed that Flaveria bidentis (L.), an NADP-malic enzyme-type C4 dicotyledon, had substantial PSII activity in bundle sheath cells and that malate and aspartate apparently contributed about equally to the transfer of CO2 to bundle sheath cells. Preparations of bundle sheath cells and chloroplasts isolated from these cells evolved O2 at rates between 1.5 and 2 mol · min–1 · mg–1 chlorophyll (Chl) in the light in response to adding either 3-phosphoglycerate plus HCO 3 or aspartate plus 2-oxoglutarate. Rates of more than 2 mol O2 · min–1 · mg–1 Chl were recorded for cells provided with both sets of these substrates. With bundle sheath cell preparations the maximum rates of light-dependent CO2 fixation and malate decarboxylation to pyruvate recorded were about 1.7 mol · min–1 · mg–1 Chl. Compared with NADP-malic enzyme-type grass species, F. bidentis bundle sheath cells contained much higher activities of NADP-malate dehydrogenase and of aspartate and alanine aminotransferases. Time-course and pulse-chase studies following the kinetics of radiolabelling of the C-4 carboxyl of C4 acids from 14CO2 indicated that the photosynthetically active pool of malate was about twice the size of the aspartate pool. However, there was strong evidence for a rapid flux of carbon through both these pools. Some behavioral and physical defenses of Lymantria dispar (L.) pupae are described. It was found that the layer of webbing surrounding pupae significantly reduced oviposition rates in the pupal parasitoid Brachymeria intermedia (Nees) (Hymenoptera, Chalcididae). The reasons for this reduction and consequent parasitoid responses were investigated. The role of these behaviors in this host parasitoid relationship are discussed.
Résumé Le comportement défensif des chrysalides de Lymantria dispar L. consiste en cambrage et en tournoiement. Les chrysalides encoconées étaient plus sensibles aux stimuli tactiles que celles qui ont été extraites des cocons. Brachymeria intermedia Nees avait moint de succès dans l'insertion complète de la tarière dans des chrysalides encoconées qu'extraites, car elles devenaient empêtrées dans le cocon quand la chrysalide se défendait. Il y avait différentes réponses du parasitoïde à l'empêtrement. Elles comprenaient l'abandon de l'attaque par un départ, la remise en selle sur l'hôte, la morsure à travers les fils du cocon, l'attaque d'un hôte voisin, le repos ou la toilette. Les taux d'insertion de la tarière pouvaient être augmentés par l'élimination artificielle de quelques fils. La durée des opérations était plus courte sur les chrysalides plus petites. La discussion a porté sur l'importance de ces comportements sur les relations de ces parasitoïdes avec leurs hôtes.

Abstract. Two anatomical variants of Panicum maximum Jacq. were observed to accumulate an unusually large number of starch grains in the bundle sheath chloroplasts when grown under controlled environmental conditions in a nutrient medium containing a low level of nitrate nitrogen (20 mg N dm−3 as KNO3). When these plants were placed under dark conditions the chloroplasts were destarched, but exhibited a marked distortion of the thylakoid membranes. Under a higher level of nitrate nitrogen supply (200 mg N dm−3 as KNO3) the number of starch grains was markedly reduced compared to that observed above in both plant variants. When the nitrogen was supplied as ammonium nitrogen (200 mg N dm−3 as NH4Cl) there was again a high level of starch in the bundle sheath chloroplasts, the level being only slightly lower than that observed at the low KNO3 supply. An unusually large number of starch grains accumulated in the bundle sheath chloroplasts in the absence of added phosphorus in the nutrient medium, in the presence of the higher nitrate nitrogen level. It is suggested that the increased starch accumulation results from a reduced trans-location of Calvin cycle intermediates out of the chloroplasts into the cytoplasm and that both nitrate nitrogen and phosphorus may play an important role in this process. A good correlation between high net photosynthetic activity and low bundle sheath starch content was observed. Nutrient medium requirements favouring low starch content in chloroplasts also favoured high net photosynthetic rates.  相似文献

The carbon isotope composition of terrestrial C4 plants depends on the primary carboxylation of phosphoenolpyruvate (PEP) and on the diffusion of CO2 to the carboxylation sites, but is also influenced by the final carboxylation of ribulose-1,5-bisphosphate (RuBP). Several models have been used for reproducing this complex situation. In the present review, a particular model is applied as a means to interpret the effects of environmental and genetically determined factors on carbon isotope discrimination during C4 photosynthesis. As a new feature, the model considers four types of limitation of the overall CO2 assimilation rate. Both carboxylation reactions are assumed to be limited by either maximum enzyme activity or maximum substrate regeneration rate. The model is applied to experimental data on the effects of CO2, irradiance and water stress on short-term discrimination by leaves of several C4 species measured simultaneously with CO2 gas exchange characteristics. In particular, different patterns of the influence of low irradiances on carbon isotope discrimination are interpreted as due to variations in that irradiance at which a transition from limitation by PEP regeneration rate and RuBP carboxylase activity to limitation by the regeneration rates of both substrates occurs. After discussing literature data on the effects of environmental conditions on carbon isotope discrimination by C4 plants seasonal and developmental changes in carbon isotope composition, studies on the systematic and geographic distribution of C4 plants, evolutionary and genetical aspects, and some ecological implications are reviewed.  相似文献
