In this minireview, the earlier researches that led to the discovery of the period-four oscillations of the flash-induced oxygen formation are presented. It also includes the background of the classical model proposed by Bessel Kok, in which the formation of oxygen requires the sequential accumulation of four positive charges on the donor side of the same reaction center. Among his many contributions to photosynthesis, William Arnold made critical suggestions about the mechanism of the initial stages of excitation energy transfer and its measurement. Thus he helped found not only the general concept of the photosynthetic unit but also the key idea behind the detailed functional aspects of its chlorophyll antenna. The detection of hydrogen metabolism in green algae more than 60 years ago by Hans Gaffron dispelled the widely accepted dogma at that time that this feature was unique to prokaryotic organisms. Research on this unexpected aspect of algal physiology has continued until today because of its evolutionary implications and possible practical significance. This minireview focuses on the work of Gaffron and his collaborators, whose experiments provided most of the information about the mechanism of hydrogen metabolism in algae during the 35 years following its discovery. It is shown that the emergence of our present mechanistic concepts was closely linked to the changing perception of the process of photosynthetic water oxidation. Whereas the mechanism of `photoreduction,' i.e., the photoassimilation of carbon dioxide with hydrogen as the electron donor, was well understood already by Gaffron's group as being a reaction mediated by Photosystem I only, a clear concept of the mechanism of light-dependent hydrogen production has been more difficult to establish. Gaffron and his collaborators provided ample evidence, however, that `photohydrogen' evolution can be fueled by reducing equivalents derived from a photolysis of water as well as by an oxidation of internal and external organic molecules. The presently prevailing view embraces this concept of multiple pathways, but the relative contribution of each of them, and the regulatory mechanisms determining it, remain a matter of debate. This account is focused upon the early part of my career in order to illuminate the logistics and the culture of our science in the period 1936 to 1949. A roundabout path took me from a farm in Pennsylvania to a PhD under George Burr at Minnesota in 1939. In studying the photosynthetic competence of chlorophyll formed by the green alga Chlorella in darkness, I stumbled upon the phenomenon of photoinhibition. In a two-year postdoctorate at the Smithsonian Institution, I worked under E.D. McAlister. Our major accomplishment was in making simultaneous recordings of fluorescence and CO2 uptake during the induction period. Variations in photosynthetic behavior of Chlorella led to a study of culture conditions and a recognition of the changing conditions which occur in batch cultures. A continuous culture apparatus (turbidostat) was developed as a means of attaining steady-state growth and production of uniform experimental material. I exploited the device in work at my first (and only) position at The University of Texas in 1941 and subsequent years. Study of the CO2/O2 gas exchange ratio led to the recognition of the important role of nitrate in the photosynthetic metabolism of algae. A fragment of cauliflower mosaic virus (CaMV) DNA, containing delta 3, one of the three discontinuity sequences, was cloned in various ways into CaMV DNA deleted for the delta 3 sequence. The series of constructions was monitored for the appearance of the typical single-strand (ss) discontinuity after hybrid CaMV replication in plants. The delta 3 discontinuity was observed only if the orientation of inserted DNA sequence was the same as in the wild-type virus. Long polylinker sequences used for insertion of the fragment into cloned viral DNA, affected the stability of the insert in progeny viral DNA in plants by acting as recombination targets.  相似文献

Charles Stacy French, one of the great men of photosynthesis research, died on 13 October 1995. He received his PhD at Harvard University where he associated with William Arnold, Caryl Haskins, later president of the Carnegie Institution of Washington, and Pei-Sung Tang. He did early work on the photosynthesis of photosynthetic bacteria with Robert Emerson and later with Otto Warburg. French worked for three years with James Franck in Chicago. His associates there included Hans Gaffron, Robert Livingston, Warren Butler and Roderick Clayton. After spending three years at the University of Minnesota he became the director of the Department of Plant Biology of the Carnegie Institution of Washington and remained there until he retired in 1973. French's research career at the Carnegie Institution was marked by the development of novel and ingenious pieces of equipment such as the French pressure cell used to prepare chloroplast particles to measure partial reactions of photosynthesis. He developed the first recording fluorescence spectrophotometer and demonstrated efficient energy transfer between certain photosynthetically-active pigments, a spectrophotometer that measured the first derivative of absorbance, as well as a novel analog computer to show that complex absorption curves in living plants are produced by a number of distinct forms of chlorophyll occurring in vivo. French used the Blinks rate-measuring oxygen electrode to measure action spectra of oxygen evolution by photosynthesis automatically. He and Jack Myers did some of the pioneering work on the Emerson effect showing the necessary cooperation of two photosystems in photosynthesis. French used the Carnegie Institution's fellowship program to bring large numbers of scientists from around the world to his laboratory. When Stacy French died in 1995, the field of photosynthesis lost one of its great and early pioneers.This is CIW/DPB publication No. 1314.  相似文献
