Biodegradation of phenol has been investigated using a bacterial consortium consisting of two bacterial isolates; one of them used for the first time in phenol biodegradation. This consortium was isolated from activated sludge and identified as Providencia stuartii PL4 and Pseudomonas aeruginosa PDM (accession numbers KY848366 and MF445102, respectively). The degradation of phenol by this consortium was optimal at pH 7 with using 1500?mg?l?1 ammonium chloride as a nitrogen source. Interestingly, after optimizing the biodegradation conditions, this consortium was able to degrade phenol completely up to 1500?mg?l?1 within 58?h. The immobilization of this consortium on various supporting materials indicated that polyvinyl alcohol (PVA)-alginate beads and polyurethane foam (PUF) were more suitable for biodegradation process. The freely suspended cells could degrade only 6% (150?mg?l?1) of 2500?mg?l?1 phenol, whereas, the immobilized PVA-alginate beads and the immobilized PUF degraded this concentration completely within 120?h of incubation with degradation rates (q) 0.4839 and 0.5368 (1/h) respectively. Thus, the immobilized consortium of P. stuartii PL4 and P. aeruginosa PDM can be considered very promising in the treatment of effluents containing phenol.  相似文献

Epacris stuartii (Epacridaceae) is an endangered heathland shrub in which seedling recruitment occurs almost exclusively after fire. Seed viability and multiple seed dormancy mechanisms were examined to explore why levels of seedling recruitment were low after some fires, despite high rates of pre-fire seed production. The individual and combined effects of heat shock, smoke derivatives and darkness on germination were tested in the laboratory using an orthogonal logit-linear model, a form of analysis ideally suited to hypotheses concerning multiple germination cues. Seed viability (56%) was found not to be limiting. After 89 days of incubation, germination was significantly enhanced by heat shock, smoke treatment and continuous darkness but there were no significant interactions. These effects were equal in magnitude and additive such that maximum germination (42% of viable seeds) was stimulated when all three treatments were combined, significantly less germination occurred in response to any two treatments combined (22–23%), any single treatment stimulated germination at levels (10–11%) significantly less than two treatments and the lowest levels of germination occurred among untreated seeds (4%). Relative to the untreated control, germination was stimulated by smoke derivatives in high concentrations but not in low concentrations. The effect of darkness diminished with time so that after 270 days of incubation darkness had no significant effect on germination, while heat and smoke still enhanced germination significantly, additively and interchangeably. More prolonged germination of seeds exposed to light on the soil surface than buried seed may spread the risk of desiccation of emerging seedlings over multiple rainfall events. The role of heat and smoke derivates suggests that fire management is a crucial tool for influencing seedling recruitment and hence the survival of E. stuartii at its only known location. Mating in the marsupial genus Antechinus is a synchronous annual event that is characterized by monoestry in females and abrupt postmating mortality in males. Male semelparity (multiple copulations during a single breeding season per lifetime) is often assumed to occur as a consequence of the intense mating effort expended by males in the rut, but the forces selecting for this remain elusive. Here, we investigate selection in male brown antechinus, Antechinus stuartii, and test two hypotheses for the evolution of semelparity: intermale competition and sperm competition. If intermale competition drives semelparity, we predicted that males would be under strong selection for large body size. If sperm competition is important, we predicted that selection would be strongest on scrotal size, a surrogate for testes volume. Using microsatellite markers, we found that 92% of females in free-living conditions mated with multiple males, producing litters of eight that had up to four fathers. These observations confirm the potential for sperm competition. Using selection analysis, we then found paternity success in 119 males to be related most strongly to body mass and scrotal size, thus providing support for both hypotheses. Large males presumably experience increased paternity success by gaining more matings or prolonged copulations via mate guarding, while large testes may allow increased sperm investment per copulation. Increased levels of free corticosteroid hormones in males facilitate the extreme mating effort during the short period of rut, but lead to immune suppression and consequently to the phenomenon of postmating mortality.  相似文献

The black rat, Rattus rattus, is an alien rodent in Australian ecosystems where niche overlap with native small mammals may lead to competition for resources and displacement of native species. In coastal habitats surrounding Jervis Bay in south‐eastern Australia, R. rattus co‐occurs with the native bush rat, Rattus fuscipes, and brown antechinus, Antechinus stuartii. Relative distributions and abundances, and fine‐scale space use suggest invasive and native rodents compete for use of space and habitat. Such competitive interactions were not evident between R. rattus and native A. stuartii, which was negatively influenced more by disturbance to habitat. Differences in rodent communities between spatially separate forests forming the northern and southern peninsulas of Jervis Bay potentially reflect symmetrical competition and differences in competitive outcomes. In southern forests, R. rattus was largely restricted to patches of disturbed forest associated with campgrounds. Competitive interference by native rodent populations inhabiting surrounding intact forests may have so far limited R. rattus colonization of these areas. In northern forests, R. rattus was the predominant rodent irrespective of disturbance, while populations of R. fuscipes were unusually low seemingly due to poor juvenile recruitment. Native individuals avoided areas frequented by adult R. rattus and given that species did not partition use of microhabitats, R. rattus most likely precluded R. fuscipes from suitable habitat and in doing so limited native populations. We discuss how natural disturbance of habitat and human activity have potentially facilitated successful invasion by R. rattus of the northern forests. Studies that manipulate rodent populations are required to support these interpretations of observed patterns.  相似文献

A study was conducted to test the effect of inoculation with soil containing mycorrhizae propagules and other soil micro organisms on survival of stem cuttings of Epacris impressa. With non-sterile soil present, the cuttings were mycorrhizal, more survived and the survivors were healthier, with more flowers and roots, although root area was not increased. Soil inoculum thus increased the success of propagation of Epacris impressa for conservation purposes and in commercial horticulture.  相似文献

The O-polysaccharide of Providencia stuartii O33 was obtained by mild acid degradation of the lipopolysaccharide and the following structure of the tetrasaccharide repeating unit was established: -->6)-alpha-D-GlcpNAc-(1-->4)-alpha-D-GalpA-(1-->3)-alpha-D-GlcpNAc-(1-->3)-beta-D-Quip4N(Ac-D-Asp)-(1-->, where d-Qui4N(Ac-D-Asp) is 4-(N-acetyl-D-aspart-4-yl)amino-4,6-dideoxy-D-glucose. Structural studies were performed using sugar and methylation analyses and NMR spectroscopy, including conventional 2D 1H, 1H COSY, TOCSY, NOESY and 1H, 13C HSQC experiments as well as COSY and NOESY experiments in an H2O-D2O mixture to reveal correlations for NH protons. The O-polysaccharide of P. stuartii O33 shares an alpha-D-GlcpNAc-(1-->3)-beta-D-Quip4N(Ac-D-Asp) epitope with that of Proteus mirabilis O38, which seems to be responsible for a marked serological cross-reactivity of anti-P. stuartii O33 serum with the lipopolysaccharide of the latter bacterium. P. stuartii O33 is serologically related also to P. stuartii O4, whose O-polysaccharide contains a lateral beta-D-Qui4N(Ac-L-Asp) residue.  相似文献

Abstract A Citrobacter sp. was reported previously to accumulate heavy metals as cell-bound heavy metal phosphates. Metal uptake is mediated by the activity of a periplasmic acid-type phosphatase that liberates inorganic phosphate to provide the precipitant ligand for heavy metals presented to the cells. Amino acid sequencing of peptide fragments of the purified enzyme revealed significant homology to the phoN product (acid phosphatase) of some other enterobacteria. These organisms, together with Klebsiella pneumoniae , previously reported to produce acid phosphatase, were tested for their ability to remove uranium and lanthanum from challenge solutions supplemented with phosphatase substrate. The coupling of phosphate liberation to metal bioaccumulation was limited to the metal accumulating Citrobacter sp.; therefore the participation of species-specific additional factors in metal bioaccumulation was suggested.  相似文献
