Summary A chemostat was used as a model system to study competitive interactions of diazotrophic microorganisms. Enrichment experiments were carried out under microaerobic conditions (8.7 mol O2/l) with malate as the sole carbon source. The starting material was a Korean rice soil including intact root pieces. The enrichment process was governed by the dilution rate. High dilution rates resulted in the enrichment ofAzospirillum lipoferum, whereas low dilution rates led to the predominance of an unidentified organism, named Isolate R. Dilution rates were set in the range from D=0.005 to D=0.1 h–1. The growth kinetics of both organisms followed Monod's model in the enrichment culture. From the experiments, the maximum specific growth rate ofA. lipoferum and Isolate R were 0.069 h–1 and 0.025 h–1, respectively. The corresponding Ks-values were 8.4 and 0.9 (mg. 1–1). The point of theoretical coexistence of both organisms was calculated to occur at a substrate concentration of s=3.0 (mg.l–1) with a growth of rate =0.018 h–1. Hence the preset nutritional niches occupied by at least two organisms.Azospirillum lipoferum seems to represent the copiotroph microflora and Isolate R is of the oligotroph type. In addition to its high substrate affinity Isolate R liberatedca. 75% of the fixed nitrogen into the medium, which indicates its potential role for mutualistic interactions in the rhizosphere.  相似文献

A model of single-species growth in the chemostat on two non-reproducing, growth-limiting, noninhibitory, perfectly substitutable resources is considered. The medium in the growth vessel is enriched by increasing the input concentration of one of the resources. Analytical methods are used to determine the effects of enrichment on the asymptotic behaviour of the model for different dilution rates. It is shown that there exists a threshold value for the dilution rate which depends on the maximal growth rate of the species on each of the resources. Provided the dilution rate is below the threshold, enrichment is beneficial in the sense that the carrying capacity of the environment is increased, regardless of which resource is used to enrich the environment. When the dilution rate is increased beyond the threshold, it becomes important to consider which resource is used for enrichment. For one of the resources it is shown that, while moderate enrichment can be beneficial, sufficient enrichment leads to the extinction of the microbial population. For the other resource, enrichment leads from washout or initial condition dependent outcomes to survival, and is thus beneficial. There are important implications of these results to the management of natural aquatic ecosystems. For example, while enrichment may be beneficial to the microbial species during the summer months, it can lead to their decimation during spring run-off, when the natural dilution rate is higher.Research partially supported by an Ontario Graduate Scholarship. 目的:研究FXR在胃炎,胃粘膜肠化生及胃癌组织中的表达,分析其在胃癌发生中的意义。方法:采用免疫组化方法检测FXR在55例胃炎组织,61例胃黏膜肠化生组织及61例胃癌组织中的表达,利用统计学方法 SPSS17.0软件分析其在三种组织中的表达变化,结合文献回顾,分析FXR在胃癌发生中的意义。结果:FXR在胃黏膜肠化生中的表达明显高于胃炎组织(P0.05),而在胃癌组织中,FXR的表达显著低于胃粘膜肠化生组织(P0.05)。结论:FXR是一个潜在的胃癌发生生物标记物,其具体机制有待于进一步探索。  相似文献

贮食行为是动物应对食物资源的季节性和不可预测性变化的一种适应性生存策略。在群居性贮食动物中,同胎个体常表现出不同的贮食水平,而关于贮食行为与动物的个性和代谢水平之间的关系尚缺乏研究。本文以长爪沙鼠(Meriones unguiculatus)为对象,在筛选出具有高贮食和低贮食行为特征个体的基础上,比较了两组动物的个性特征(勇敢行为和探索能力)、静止代谢率、血清甲状腺激素水平、贮食期间的运动距离和贮食后的平均每日代谢率等。结果发现:高、低贮食长爪沙鼠的数量各占49%(22/45)和47%(21/45),两组动物之间的个性特征、静止代谢率和血清甲状腺激素均没有显著差异。在贮食期间高贮食个体的运动距离显著高于低贮食个体,且在停止贮食后,高贮食个体的平均每日代谢率显著低于低贮食个体。这些结果表明,在室内条件下,长爪沙鼠的贮食量高低与个性和静止代谢率无关,但高贮食个体会在停止贮食后降低其总能量消耗,以补偿贮食过程中的高能量代价。  相似文献

Objective: The traditional Chinese medicine Caulis Sargentodoxae is widely used in the treatment of ulcerative colitis (UC), but the mechanism remains unknown. The present study aims to reveal its effective components, targets and pathways through network pharmacology and bioinformatics approaches.Materials and methods: Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) was used to identify effective components. The ligand-based targets prediction was achieved through SwissTargetPrediction and TargetNet. UC-related targets were identified using Gene Expression Omnibus (GEO) data and DisGeNET. The common targets of disease and components were constructed and analyzed by PPI network. Lastly, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses are used to explain the functions of these common targets. Components-Targets-Pathways network was visualized and analyzed to further reveal the connection between the components and targets.Results: Eight active components and 102 key targets were identified to play an important role in UC. These targets were related to regulation of protein serine/threonine kinase activity, positive regulation of cell motility, response to molecule of bacterial origin, response to toxic substance, ERK1 and ERK2 cascade, peptidyl-tyrosine modification, inositol lipid-mediated signaling, cellular response to drug, regulation of inflammatory response and leukocyte migration. Moreover, HIF-1 signaling pathway and PI3K-Akt signaling pathway were the key targets involved in UC-related signaling pathways.Conclusion: The eight active components of Caulis Sargentodoxae mainly play a therapeutic role for UC through synergistic regulation of HIF-1 signaling pathway and PI3K-Akt signaling pathway.  相似文献

摘要目的:探讨CBL结合PBL及LBL的教学方法在胸外科临床实习中的应用效果。方法:选取我院胸外科临床实习学生共60人为研究对象,随机将其分为实验组及对照组,每组各30人。实验组采用CBL+PBL-4-LBL教学方法;对照组采用LBL教学方法。在学习结束时,采用闭卷考试的形式进行成绩考核;同时采用不记名问卷调查,进行两种教学效果和教学模式的评价。结果:两组学生在理论知识考试部分成绩水平持平,无统计学差异。但是实验组学生在病例分析、基本技能及总成绩等部分成绩明显高于对照组。PBL+CBL+LBL教学方法在激发学习兴趣、提高学习积极性、自学能力、建立独立思维模式等方面显著高于LBL组。结论:CBL+PBL+LBL教学方法优于传统教学,可以提高学习的积极性,有助于医学生临床思维和实践能力的培养。  相似文献

目的:腋臭是美容整形外科的常见病,目前发病机制尚不明确,已证实人体大汗腺中的载脂蛋白D(ApoD)在腋臭患者大汗腺中高表达,并且与腋臭的发生密切相关。探明ApoD在大汗腺细胞中的信号转导通路,可以进一步明确其在腋臭发病过程中的作用机制。JNK信号转导通路与多种疾病的发生有关。课题组前期已经做了JNKl对ApoD调控作用的相关研究,证明了在腋臭发病过程中JNK1是通过调控ApoD的转录来上调ApoD的表达。本实验在课题组前期研究基础上,探讨JNKl下游转录因子AP-1是否在JNKl上调ApoD通路中发挥作用。方法:取腋臭志愿者腋区皮肤组织,进行汗腺细胞培养。把汗腺细胞分为5.二氢睾酮处理组、5-二氢睾酮联合姜黄素处理组和空白对照3个组,用姜黄素抑制AP-1的活性,通过Real.timePCR实验方法检测ApoD在姜黄素抑制下的表达变化。结果:在姜黄素的抑制下,ApoD表达明显降低。在体外培养汗腺细胞加入5.二氢睾酮联合姜黄素处理后,ApoD的表达量在mRNA水平低于单独的5-二氢睾酮处理组和正常对照组(P〈0.05)。结论:姜黄素抑制了AP.1的活化导致ApoD的表达降低。在腋臭的发病过程中,JNKl的下游转录因子AP-1对ApoD有明显的上调作用。AP-1可能在JNKl上调ApoD这条通路中扮演了很重要的角色,它可能是JNK1和ApoD的中间转录因子。  相似文献

目的:研究PCI术后是否应用依诺肝素抗凝治疗对患者临床疗效的影响。方法:于2011年5月至2012年1月间,连续入选在我院行冠状动脉造影并置入了支架的患者158例,将符合标准的患者随机分为非抗凝组或抗凝组两组各79名。非抗凝组术后常规应用阿司匹林和氯吡格雷。抗凝组术后加用依诺肝素。对入选患者进行院内随访记录其主要心脏不良事件及出血事件。结果:支架植入成功率100%。术后抗凝组113处病变共置入支架135枚;非抗凝组109处病变置入支架115枚。院内随访:主要心脏不良事件和严重出血差异无统计学意义。小出血事件抗凝组多于非抗凝组(P=0.007)。结论:冠状动脉支架置入术后非抗凝治疗组缺血不良事件发生率较抗凝组无明显增加,小出血并发症明显减少。该研究结果表明,对PCI术无特殊并发症的患者术后无需常规抗凝治疗。  相似文献

利用E.coli BL21/pCDFDuet-gdh—cr-X共表达全细胞催化6-氰基-(5R)-羟基-3-羰基己酸叔丁酯不对称还原合成6-氰基-(3R,5R)-二羟基已酸叔丁酯。结果表明:在菌体用量4.85g/L、葡萄糖与底物质量浓度比为1:1、温度28℃、pH7.0条件下,80.0g/L6-氰基-(5R)-羟基-3-羰基己酸叔丁酯生物还原2h后,底物转化率可达99.0%,产物d.e.值大于99.5%。在考察范围内,NADP^+用量对催化效率无显著作用。  相似文献

为充分利用营养没有得到充分吸收的栽培过食用菌的废弃菌糠、明确在牡丹江地区再利用食用菌菌糠生产黑木耳的适宜比例,以灵芝和鲍鱼菇的废弃菌糠为试验材料,进行不同配方的对比试验。试验结果表明:黑木耳培养料中灵芝菌糠体积分数为30%时栽培效果最佳,显著优于参入鲍鱼菇菌糠的各种培养料配方(P〈0.01)。因此,可以利用灵芝菌糠废料作为替代料栽培黑木耳使用。  相似文献
