The effects of light quality and irradiance, and supply of organic carbon and vitamins on the growth of two forms of Ecklonia radiata in tissue culture were examined. A callus of unpigmented cells developed over the cut surface of newly excised explants of stipe. This growth was best in the dark but stopped after 10 weeks. Pigmented, mainly filamentous clumps of cells developed from explants after several weeks in culture. These required light for growth, with growth being enhanced by increasing photon flux density up to 30 μmol photon m-2 s-1, with the active spectral component being red light (> 600 nm). The addition to the medium of a range of organic carbon sources or vitamins did not stimulate growth of either culture type in the dark. author for correspondence  相似文献

The effect of various macroalgal diets on the growth of grow-out (>20 mm shell length) South African abalone Haliotis midae was investigated on a commercial abalone farm. The experiment consisted of four treatments: fresh kelp blades (Ecklonia maxima (Osbeck) Papenfuss) (c. 10% protein); farmed, protein-enriched Ulva lactuca Linnaeus (c. 26% protein) grown in aquaculture effluent; wild U. lactuca (c. 20% protein); and a combination diet of kelp blades + farmed U. lactuca. Abalone grew best on the combination diet (0.423 ± 0.02% weight d?1 SGR [specific growth rate]; 59.593 ± 0.02 ?m d?1 DISL [daily increment in shell length]; 1.093 final CF [condition factor]) followed by the kelp only diet (0.367 ± 0.02% weight d?1 SGR; 53.148 ± 0.02 ?m d?1 DISL; 1.047 final CF), then the farmed, protein-enriched U. lactuca only diet (0.290 ± 0.02% weight d?1 SGR; 42.988 ± 0.03 um d”1 DISL; 1.013 final CF) that in turn outperformed the wild U. lactuca only diet (-0.079 ± 0.01% weight d?1 SGR; 3.745 ± 0.02 ?m d”?1 DISL; 0.812 final CF). The results suggest that protein alone could not have accounted for the differences produced by the varieties of U. lactuca and that the gross energy content is probably important.  相似文献

Local and chemical distribution of phlorotannins in brown algae   总被引:4,自引:2,他引:2  
The local and chemical distribution of phlorotannins among the Japanese Laminariaceae, Eisenia bicyclis, Ecklonia cava and Ecklonia kurome, was investigated. As a result of light microscopy observations with vanillin-HCl staining, phlorotannins were found to be accumulated within the vegetative cells of the outer cortical layer of the thalli, regardless of the species, stage of growth or organ. Crude phlorotannins comprised about 3.0% of the algal powder for each of the algae. High-performance liquid chromatography (HPLC) showed that the phlorotannins of E. bicyclis were composed of phloroglucinol (0.9%), phloroglucinol tetramer (4.4%), eckol (7.5%), phlorofucofuroeckol A (21.9%), dieckol (23.4%), and 8,8'-bieckol (24.6%), plus some other unknown phenolic compounds (17.3%). The composition of the phlorotannins differed little among the Laminariaceae, except for a significantly larger amount of the tetramer, MW 478, in E. bicyclis.  相似文献

The focus of this study was to measure natural spatial variability in the biodiversity of fauna inhabiting kelp holdfasts in northeastern New Zealand at several spatial scales: from meters up to hundreds of kilometers. We wished to test the hypothesis that multivariate variation and biodiversity would vary significantly at different spatial scales in different ways for the major phyla in the holdfast community (Arthropoda, Annelida, Mollusca and Bryozoa). Biodiversity was considered in terms of richness, total abundance, structural composition (as measured by the Bray-Curtis dissimilarity measure) and taxonomic breadth for each major phylum and for the assemblage as a whole. We also examined the effect of taxonomic resolution on multivariate patterns. Species richness and total abundance increased with increases in holdfast volume. Multivariate variation was greatest at the smallest spatial scale for all phyla, but different phyla showed different patterns of multivariate variation at different spatial scales. Variations among locations at the largest spatial scale were primarily due to differences in the composition and richness of bryozoans and molluscs. Location effects became less and less distinct with decreases in taxonomic resolution. There were very few significant differences in richness or abundance for holdfasts of a given volume, taxonomic breadth did not vary significantly across locations, nor did the proportional abundances of phyla. These consistencies across large spatial scales in the absence of environmental impacts and results from other studies suggest that holdfast communities in New Zealand systems would provide a useful model assemblage against which future impacts may be detected as changes in proportions of component phyla. In addition, high variability detected at small and large scales at the species level, especially for bryozoans and molluscs, suggest that these communities may also provide unique opportunities for studying and understanding sources and functions of marine biodiversity.  相似文献

We tested the ability of sporophytes of a small kelp, Ecklonia radiata (C. Agardh) J. Agardh, to adjust their photosynthesis, respiration, and cellular processes to increasingly warm ocean climates along a latitudinal gradient in ocean temperature (~4°C). Tissue concentrations of pigment and nutrients decreased with increasing ocean temperature. Concurrently, a number of gradual changes in the metabolic balance of E. radiata took place along the latitudinal gradient. Warm‐acclimatized kelps had 50% lower photosynthetic rates and 90% lower respiration rates at the optimum temperature than did cool‐acclimatized kelps. A reduction in temperature sensitivity was also observed as a reduction in Q10‐values from cool‐ to warm‐acclimatized kelps for gross photosynthesis (Q10: 3.35 to 1.45) and respiration (Q10: 3.82 to 1.65). Respiration rates were more sensitive to increasing experimental temperatures (10% higher Q10‐values) than photosynthesis and had a higher optimum temperature, irrespective of sampling location. To maintain a positive carbon balance, E. radiata increased the critical light demand (Ec) exponentially with increasing experimental temperature. The temperature dependency of Ec was, however, weakened with increasing ocean temperature, such that the critical light demand was relaxed in kelp acclimated to higher ocean temperatures. Nevertheless, calculations of critical depth limits suggested that direct effects of future temperature increases are unlikely to be as strong as effects of reduced water clarity, another globally increasing problem in coastal areas.  相似文献

Fragments of Ecklonia cava Kjellman were cultured under controlled laboratory conditions of light irradiance, water temperature, and photoperiod. To clarify the relationship between the maturation of E. cava and the photosynthetic products, laminaran, the content in the fragments was measured with the progress of maturation. The culture conditions ranged from 12.5 to 100 µmol m?2 s?1, 10–25°C, and 14 : 10 h LD (light : dark) to 10 : 14 h LD. In the case of low light conditions, despite an optimum temperature for maturation, the fragments did not form sori and laminaran was not accumulated during the culture period. In the case of sufficient light and non‐optimum temperature conditions, the fragments did not form sori, but laminaran was accumulated. When the fragments were cultured under optimum light and temperature conditions for maturation, laminaran was accumulated in the early stage of maturation, just before or after cortex of the bladelets thickened, and decreased with the progress of maturation, and all fragments matured regardless of the length of the photoperiod. So, these results support the idea that laminaran is used as the main respiratory substrate in the maturation of E. cava.  相似文献

Matrix metalloproteinase (MMP) inhibitors have been identified as potential therapeutic candidates for metastasis, arthritis, chronic inflammation and wrinkle formation. For the first time here we report a detailed study on the inhibitory effects of phlorotannins in brown algae, Ecklonia cava (EC) on MMP activities in cultured human cell lines. A novel gelatin digestion assay could visualize complete inhibition of bacterial collagenase-1 activity at 20 microg/ml of EC extract during preliminary screening studies. Sensitive fluorometric assay revealed that EC extract can specifically inhibit both MMP-2 and MMP-9 activities significantly (P < 0.001) at 10 microg/ml. In addition, artificially induced activities of MMP-2 and MMP-9 in human dermal fibroblasts and HT1080 cells were inhibited by EC extract in a more or less similar manner to the positive control doxycycline. Even though the expression levels of MMPs differ from one cell type to the other, gelatin zymography clearly revealed that both MMP expression and activity in cells can be inhibited by EC extract. More interestingly, EC extract did not exert any cytotoxic effect even at 100 microg/ml anticipating its potential use as a safe MMP inhibitor.  相似文献

Kelp forests dominated by species of Laminariales are globally recognized as key habitats on subtidal temperate rocky reefs. Forests characterized by fucalean seaweed, in contrast, receive relatively less attention despite being abundant, ubiquitous, and ecologically important. Here, we review information on subtidal fucalean taxa of Australia's Great Southern Reef, with a focus on the three most abundant and widely distributed genera (Phyllospora, Scytothalia, and Sargassum) to reveal the functionally unique role of fucoids in temperate reef ecology. Fucalean species span the entire temperate coastline of Australia (~71,000 km2) and play an important role in supporting subtidal temperate biodiversity and economic values on rocky reefs as well as in adjacent habitats. Climatic and anthropogenic stressors have precipitated significant range retractions and declines in many fucoids, with critical implications for associated assemblages. Such losses are persistent and unlikely to be reversed naturally due to the life history of these species and colonization of competitors and grazers following loss. Active restoration is proving successful in bringing back some fucoid species (Phyllospora comosa) lost from urban shores and will complement other passive and active forms of conservation. Fucalean forests play a unique role on subtidal temperate reefs globally, especially in Australia, but are comparatively understudied. Addressing this knowledge gap will be critical for understanding, predicting, and mitigating extant and future loss of these underwater forests and the valuable ecosystem services they support.  相似文献

Photosynthetic rates were compared between Ecklonia cava bladelets with and without zoosporangial sori sampled from the subtidal zone (about 5 m deep) in Nabeta Bay, Shimoda, Japan. Photosynthetic rates of bladelets were lower in the sorus portion than in the non-sorus portion on the basis of area, dry weight and chlorophyll a. Respiration rates were higher in the sorus portion than in the non-sorus portion on the basis of area and chlorophyll a, whereas they were almost the same on a dry weight basis. The differences were mainly due to a large difference in dry weight per unit bladelet area between the sorus and the non-sorus portion. Light compensation points were higher in the sorus portion than in the non-sorus portion.  相似文献
