Out of some 750 strains of microorganisms, a potent bacterium for lipase production was isolated from soil and was identified as Chromobacterium viscosum.

The bacterium accumulates lipase in culture fluid when grown aerobically at 26°C for 3 days in a medium composed of soluble starch, soy bean meal, lard and inorganic salts.

Chromobacterium lipase had an optimum pH of 7.0 for activity at 37°C, and an optimal temperature of 65°C at pH 7.0. The enzyme retained 80% of the activity when heated for 10 min at 70°C. This lipase was capable of hydrolyzing a variety of natural fats and oils, and it was more active on lard and butter than on olive oil. The activity was stimulated by Ca2+, Mg2+, Mn2+ and inhibited by Cu2+, Hg2+ and Sn2+. It was not diminished but rather stimulated by a high concentration of bile-salts.  相似文献

The paper reports the isolation and structural elucidation of seven diketopiperazines from the title microorganisms. Although all isolates are known, three of which were isolated from the actinomycetes for the first time. 可培养海绵共附生微生物的PKS基因筛选   总被引:1,自引:0,他引:1  
利用PCR技术对21株分离自我国南海澳大利亚厚皮海绵的放线菌及9株分离自贪婪倔海绵的芽孢杆菌进行了聚酮合酶(PKS)基因筛选。从芽孢杆菌C89中获得了一条669bp片段,BLAST比对结果表明该基因对应的氨基酸序列和枯草芽孢杆菌I型聚酮合成酶基因(PKS)KS域的相似性达96%。通过系统发育分析推测芽孢杆菌C89PKS基因属于trans-AT型。首次证明了贪婪倔海绵共附生微生物中存在PKS基因,这为海绵活性物质的微生物来源假说提供了证据;同时也为可以产生聚酮类化合物的微生物筛选以及聚酮类化合物的发酵制备奠定了基础。  相似文献

The acetylcholinesterase inhibitory and/or antitumour activities of amino-, thio- and ester-derivatives of avarol selected were evaluated for the first time at in vitro conditions. Avarol-3′,4′-dithioglycol (1) and avarol-4′-(3)mercaptopropionic acid (3) were shown to be the best inhibitors of the enzyme tested (0.50?µg and IC50 0.05?mM and 0.50?µg and IC50 0.12?mM, respectively), while 4′-tryptamine-avarone (9) and avarol-3′-(3)mercaptopropionic acid (2) exhibited the highest cytotoxicity against the human breast T-47D cancer cell line (IC50 0.66?µg/mL and 1.25?µg/mL, respectively). According to experimental data obtained, the sesquiterpenoid hydroquinone structure of bioactive avarol derivatives may inspire development of new pharmacologically useful substances to be used in the treatment of Alzheimer's disease and/or human breast tumour.  相似文献

The aim of our research is to design tank systems to culture Dysidea avara for the production of avarol. Flow information was needed to design culture tanks suitable for effective production. Water flow regimes were characterized over a 1-year period for a shallow rocky sublittoral environment in the Northwestern Mediterranean where D. avara sponges are particularly abundant. Three-dimensional Doppler current velocities at 8-10-m depths ranged from 5 to 15 cm/s over most seasons, occasionally spiking to 30-66 cm/s. A thermistor flow sensor was used to map flow fields in close proximity ( approximately 2 cm) to individual sponges at 4.5-, 8.8-, and 14.3-m depths. These "proximal flows" averaged 1.6 cm/s in calm seas and 5.9 cm/s during a storm, when the highest proximal flow (32.9 cm/s) was recorded next to a sponge at the shallowest station. Proximal flows diminished exponentially with depth, averaging 2.6 cm/s +/- 0.15 SE over the entire study. Flow visualization studies showed that oscillatory flow (0.20-0.33 Hz) was the most common regime around individual sponges. Sponges at the 4.5-m site maintained a compact morphology with large oscula year-around despite only seasonally high flows. Sponges at 8.8 m were more erect with large oscula on tall protuberances. At the lowest-flow 14.3-m site, sponges were more branched and heavily conulated, with small oscula. The relationship between sponge morphology and ambient flow regime is discussed.  相似文献

SYNOPSIS Random examination of domestic animals revealed the frequent presence of free-living amebae in their bodies. In diseased or dead cows, dogs, pigs, rabbits, pigeons, and turkeys 15 strains of amebae were found, belonging to the genera Acanthamoeba (A. polyphaga), Hartmannella (H. vermiformis) , and Vahlkampfia (V. avara, V. enterica, V. inornata). , They were usually accompanied by other infectious agents in different parts of the host bodies. Pathogenicity of 3 isolates could not be demonstrated by inoculation of laboratory animals.
Some features of the isolates differed from those previously known for members of these genera. These strains may be considered amphizoic amebae according to Page (1974).  相似文献

从我国南海陵水地区采集的一种未定种海绵Dysideasp中首次分离到7个化合物,经MS,NMR等光谱技术,确定了结构分别为胆固醇(1),过氧化麦角甾醇(2),cholesl-7-en-9a,11α-epoxy-3β,5α,6β,19-tetraol-6-monoacetate(3),甲基尿嘧啶(4),尿嘧啶(5),甲基尿嘧啶脱氧核糖核苷(6),尿嘧啶脱氧核糖核苷(7)。其中化合物3的^1H NMR和^13C NMR数据通过二维NMR实验首次得到了全归属。  相似文献

One of the most promising alternative technologies to antifouling paints based on heavy metals is the development of coatings whose active ingredients are compounds naturally occurring in marine organisms. This approach is based on the problem of epibiosis faced by all marine organisms and the fact that a great number of them cope with it successfully. The present study investigated the antifouling activity of a series of extracts and secondary metabolites from the epibiont-free Mediterranean sponges Ircinia oros, I. spinosula, Cacospongia scalaris, Dysidea sp., and Hippospongia communis. Antifouling efficacy was evaluated by the settlement inhibition of laboratory-reared Balanus amphitrite Darwin cyprids. The most promising activity was exhibited by the metabolites 2-[24-acetoxy]-octaprenyl-1-4-hydroquinone (8a), dihydrofurospongin II (10), and the alcoholic extract of Dysidea sp.  相似文献
