The relationships between egg predation of an herbivorous lady beetle Epilachna niponica (Lewis ) and its predator, the earwig Anechura harmandi (Burr ), were examined in both time and space. In spite of little annual, changes in egg densities, egg mortality due to predation varied considerably. There was no, clear relationship between the earwig density and the proportionate predation over the five years. The seasonal occurrence of earwig nymphs on thistle plants, however, was closely synchronized with that of egg predation. Predator attacks on the beetle occurred in a time-restricted manner. Thus, later cohorts mostly escape from heavy predatory pressure. No spatially density-depent egg predation was detected at the level of either thistle plants or thistle patches. Furthermore, there was no indication of aggregative behaviour of the earwig in response to local egg density. The earwig density was more likely to be associated with particular localities with sandy deposits available for its nest site.  相似文献

Summary Experiments conducted on rocky bottoms at 7–11 m depth in the San Juan Archipelago, Washington assessed effects of canopies of understory kelps on growth of benthic suspension feeders, determined the mechanisms responsible for effects, and assessed the influence of kelp canopies on survivorship of benthic fauna. Kelp canopics influenced growth rates of diverse suspension feeders. At several sites the musselMytilus edulis, the barnacleBalanus glandula, and the serpulid polychaetePseudochitinopoma occidentalis grew faster on the bottom beneath kelp canopies than on nearby exposed substrata. The cheilostome bryozoanMembranipora membranacea showed a mixed response to kelp canopies, growing faster in exposed regions at one site, but faster beneath canopies at another. There were no differences in growth of 2 other species (the cheilostome bryozoanCheilopora praelonga and the spongeMyxilla incrustans) between kelp and no-kelp treatments; however, some processes influenced by plant canopies affected their growth. Specific mechanisms responsible for kelp effects on growth were assessed in a series of field experiments usingPseudochitinopoma, Membranipora, Cheilopora andMyxilla. Particulate deposition on the bottom, which is more intense beneath canopies, negatively affected growth of all 4 species. Kelps also reduced rates of flow and prevented devented development of microalgal turfs beneath the canopy.Pseudochitinopoma grew faster in the weaker flows below canopies and bothCheilopora andMyxilla grew faster where there were no microalgal turfs. These other effects of kelp canopies were at least as important to growth (in the cases ofCheilopora andMyxilla) or more important to growth (in the case ofPseudochitinopoma) than were the general, deleterious effects of higher sedimentation beneath canopies. The lower growth rates caused by higher sedimentation beneath kelp canopies did not result in higher rates of animal mortality. Surprisingly, kelp canopies typically did not influence mortality due to predation. For 7 of 12 taxa, mortality rates did not differ between kelp-covered and exposed treatments. Significantly higher mortality occurred outside canopies for only 4 of 12 taxa, and for at least 2 of these 4 differences probably were not related to predation.Mytilus, a species rare at these depths, exhibited higher mortality beneath kelp canopies due to predation by crabs. Other macrophytes in fresh and salt water, as well as some benthic animals that create complex, 3-dimensional habitats, should influence benthic organisms and assemblages in ways analogous to the kelps acting through their effects on flow, particle transport, and shading.  相似文献

Summary Insect cell lines are not well-suited to colony formation in liquid medium following low-density cell plating. The present studies demonstrate that the time of addition of fetal bovine serum to the culture medium and the number of γ-irradiated feeder cells added to each plate are important factors in developing a useful colony formation assay. TN-368 lepidopteran and WR69-DM-1 dipteran cell lines were used for these experiments. Both cell types display increased plating efficiencies if serum is added to the medium one or more days prior to plating as compared to adding serum immediately before plating. Growth curves obtained by seeding cells at higher densities also indicate that cell growth is slightly better if serum is added one or more days before seeding. These findings are especially important for survival and toxicity studies because the results demonstrate that even seemingly minor factors involved in cell survival assays may benefit treated cells to a greater degree than untreated control cells, thus providing an erroneous assessment of cell survival. Summary We test the hypothesis that body size and population density of the deposit-feeding gastropod, Hydrobia truncata, are greater in muddy than in sandy habitats as a result of faster growth on fine- compared to coarse-grained sediments. We refute this hypothesis using a combination of field measurements and laboratory experiments. Three out of three populations tested had higher maximal growth rates and two of three populations approached their asymptotic size more quickly on sand than on silt-clay fractions of natural sediment. Growth decreased with increasing snail density and was as high or higher on sand as on silt-clay at all densities. Two populations were more fecund on sand than on silt-clay, and fecundity of the third population was not affected by sediment type. We show that the smaller body sizes observed in snails from the sandiest habitat result from late recruitment of these snails, relative to the other populations.  相似文献

The discovery of Foraminifera and Ostracodesin the dark green, sandy-glauconitic marl clay in the Joux Valley (in the Vaudois Jura) suggests to date these sediments Middle and Upper Albian. Summary The maintenance of primary cultures of adult hamster pancreatic cells on layers of irradiated C3H/10T1/2 cells was studied. Various types of pancreatic cells, acinar, islet and ductular cells could be identified in the cultures by light and electron microscopy. Morphologically the various pancreatic cells retained many differentiated characteristics of their respective in vivo cell types. Insulin production was maintained at near Day 1 levels for the 16 d in culture for which it was measured. Colonies of epithelial cells continued to grow during a 20 d culture period. It is believed that this procedure for maintaining functional and growing pancreas cells in culture may be a useful in vitro model for studying the initiation of pancreatic carcinogenesis. Preplanting treatment of soil naturally infested with Macroposthonia xenoplax with 1,2-dibromoethane (ethylene dibromide) significantly increased the growth rate of Juglans hindsii seedlings. When seedlings of J. hindsii, J. regia CV "Serf" and J. regia CV Eureka were inoculated with M. xenoplax, their growth was signilicantly less than that of nematode-free controls. This retarded growth rate was accompanied hy feeder root necrosis, longitudinal cracks in the older roots, and distinct lesions in the secondary phloem.  相似文献

Summary A cell culture system is described for the growth of rat tracheal epithelial (RTE) cells at clonal density. The system uses normal, early passage RTE cells grown on feeder layers of lethally irradiated 3T3 cells. The RTE cells have a high colony forming efficiency (5 to 10%) in culture, can be passaged up to 5 times, and are capable of more than 20 cumulative doublings per colony forming cell. The epithelial nature of the cells was confirmed by cell and colony morphology, immunoperoxidase staining of intracellular keratin, and cellular ultrastructural studies. The cytotoxic response of RTE cells to a variety of carcinogens, including a direct acting chemical carcinogen, a physical carcinogen, and a series of polycyclic aromatic hydrocarbons, was quantitated. A linear decrease in the logarithm of survival was observed with increasing doses ofN-methyl-N′-nitro-N-nitrosoguanidine (MNNG), γ-irradiation, 7,12-dimethylbenz(a)anthracene, and a diol-epoxide of benzo(a)pyrene. No toxicity was observed after treatment with benzo(a)pyrene or 3-methylcholanthrene over the concentration range examined. In contrast, phorbol ester tumor promoters stimulated cell growth markedly. Based on these and other studies, the RTE cell culture system represents a model system that will be useful for quantitative studies of epithelial cell growth, differentiation, and carcinogenesis.  相似文献

Hybridoma cells are featured by the effective utilization of both B lymphocytes and immortalized myeloma cells, allowing for the continuous generation of monoclonal antibodies specific to antigens. With regard to conventional hybridoma technology, B lymphocytes must be fused with myeloma cells using various methods to generate hybridoma cells. Nutrition plays an important role in hybridoma cell survival and amplification, which determines the fusion effect and antibody production. Here we compared the growth and survival rates of hybridoma in a commonly used peritoneal macrophage feeder layer (PMFL) nutrition supply system with a commercial hybridoma feeder additive (HFA) nutrition supply system at the post fusion stage and discussed the titer of monoclonal antibodies by enzyme linked immunosorbent assay (ELISA). Our results indicate that commercially available HFA promotes the survival and amplification of hybridoma clones and improves the titer of monoclonal antibodies indirectly.  相似文献

Abstract: The presence of bovine tuberculosis (TB) in cattle can negatively impact a state's economy and cattle industry. In Michigan, USA, wild white-tailed deer (Odocoileus virginianus) are a reservoir for reinfecting cattle herds. Although direct TB transmission between deer and cattle is rare, infected deer may contaminate cattle feed. To mitigate this risk, we designed and evaluated a deer-resistant cattle feeder (DRCF) device for deterring deer from feeders. The device delivered negative stimuli to condition deer to avoid cattle feeders. We tested the device by conducting a comparative change experiment at a high-density captive white-tailed deer operation in northeastern lower Michigan using pretreatment and treatment periods and random allocation of DRCF protection to 3 of 6 feeders during the treatment period. We used animal-activated cameras to collect data on deer use of feeders. Deer use was similar at protected and unprotected feeders during the pretreatment period but was lower at protected feeders during the treatment period. Deer-resistant cattle feeders were 100% effective during the first 2 treatment weeks, 94% during the first 5 weeks, but effectiveness then dropped to 61% during the final week. Excluding problems associated with low battery power and infrared sensors, DRCFs were 99% effective at deterring deer. Our results suggest that DRCFs can effectively limit deer use of cattle feed, potentially with minimal impact on feeding behavior of cattle, thus reducing potential transmission of bovine TB through contaminated feed. By employing DRCFs in bovine TB endemic areas, especially at times that deer are food stressed, agencies and producers can practically and economically reduce the potential for bovine TB to be transmitted from deer to cattle.  相似文献
