Collagen is an attractive marker for tissue remodeling in a variety of common disease processes. Here we report the preparation of protein dendrimers as multivalent collagen targeting ligands by native chemical ligation of the collagen binding protein CNA35 to cysteine-functionalized dendritic divalent (AB2) and tetravalent (AB4) wedges. The binding of these multivalent protein constructs was studied on collagen-immobilized chip surfaces as well as to native collagen in rat intestinal tissues. To understand the importance of target density we also created collagen-mimicking surfaces by immobilizing synthetic collagen triple helical peptides at various densities on a chip surface. Multivalent display of a weak-binding variant (CNA35-Y175K) resulted in a large increase in collagen affinity, effectively restoring the collagen imaging capacities for the AB4 system. In addition, dissociation of these multivalent CNA35 dendrimers from collagen surfaces was found to be strongly attenuated.  相似文献

Starburst dendrimers are highly branched oligomers. A rigid dendritic hydrocarbon, C1134H1146, has recently been synthesized. It consists of 94 phenylacetylene units displayed in a self-similar two-dimensional skeleton isomorphous to the three-coordinated Bethe lattice. The three-dimensional representation of phenylacetylene dendrimer shows a globular architecture with large voids and niches in its interior, characteristic of hyperbolic surfaces. This work investigates the geometrical scaling behavior of this starburst dendrimer using the symmetry properties of a Bethe lattice embedded in the hyperbolic plane. The results for C1134H1146 provide its density profile and an upper bound for its macromolecular size. Generation 4 polyamidoamine (PAMAM) and, for the first time, hyperbranched poly(ethylene imine) or polyglycerol dendrimers have been loaded with Gd3+ chelates, and the macromolecular adducts have been studied in vitro and in vivo with regard to MRI contrast agent applications. The Gd3+ chelator was either a tetraazatetracarboxylate DOTA-pBn4− or a tetraazatricarboxylate monoamide DO3A-MA3− unit. The water exchange rate was determined from a 17O NMR and 1H Nuclear Magnetic Relaxation Dispersion study for the corresponding monomer analogues [Gd(DO3A-AEM)(H2O)] and [Gd(DOTA-pBn-NH2)(H2O)] (k ex298 = 3.4 and 6.6 × 106 s−1, respectively), where H3DO3A-AEM is {4-[(2-acetylaminoethylcarbamoyl)methyl]-7,10-bis(carboxymethyl-1,4,7,10-tetraazacyclododec-1-yl)}-acetic acid and H4DOTA-pBn-NH2 is 2-(4-aminobenzyl)-1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid. For the macromolecular complexes, variable-field proton relaxivities have been measured and analyzed in terms of local and global motional dynamics by using the Lipari–Szabo approach. At frequencies below 100 MHz, the proton relaxivities are twice as high for the dendrimers loaded with the negatively charged Gd(DOTA-pBn) in comparison with the analogous molecule bearing the neutral Gd(DO3A-MA). We explained this difference by the different rotational dynamics: the much slower motion of Gd(DOTA-pBn)-loaded dendrimers is likely related to the negative charge of the chelate which creates more rigidity and increases the overall size of the macromolecule compared with dendrimers loaded with the neutral Gd(DO3A-MA). Attachment of poly(ethylene glycol) chains to the dendrimers does not influence relaxivity. Both hyperbranched structures were found to be as good scaffolds as regular PAMAM dendrimers in terms of the proton relaxivity of the Gd3+ complexes. The in vivo MRI studies on tumor-bearing mice at 4.7 T proved that all dendrimeric complexes are suitable for angiography and for the study of vasculature parameters like blood volume and permeability of tumor vessels. A branched amino acid was synthesized from methyl glucopyranoside; this amino acid presents three amino groups protected by Fmoc and one acid group and can be used in classic peptide synthesis. In parallel, similar azido terminated blocks were synthesized.Successive coupling reaction and deprotection afforded dendrimers with up to 27 azido functional groups. As an example of application, d-mannose and l-fucose residues were linked through CuAAC coupling and resulting glycodendrimers were evaluated in their interaction with DC-SIGN using SPR competition assay.  相似文献

A series of ferrocenyl ended dendrons containing π-conjugated systems were obtained using Wittig and Heck reactions. The dendrons were attached to eight functionalized resorcinarenes via Williamson reaction obtaining high molecular weight dendrimers. No electronic communication between metal centers was observed by cyclic voltammetry. All the dendrimers were characterized by 1H, 13C NMR, FTIR, UV-Vis, MALDI-TOF, elemental analyses, and electrochemical studies.  相似文献

Nowadays, the researchers make a big effort to find new alternatives to overcome bacterial drug resistance. One option is the application of bacteriophage endolysins enable to degrade peptidoglycan (PG) what in consequence leads to bacterial cell lysis. In this study we examine phage KP27 endolysin mixed with poly(propyleneimine) dendrimers to evaluate an antimicrobial effect against Pseudomonas aeruginosa. Polycationic compounds destabilize bacterial outer membrane (OM) helping endolysins to gain access to PG. We found out that not only bacterial lipopolysaccharide (LPS) is the main hindrance for highly charged cationic dendrimers to disrupt OM and make endolysin reaching the target but also the dendrimer surface modification. The reduction of a positive charge and concentration in maltose poly(propyleneimine) dendrimers significantly increased an antibacterial effect of endolysin. The application of recombinant lysins against Gram-negative bacteria is one of the future therapy options, thus OM permeabilizers such as cationic dendrimers may be of high interest to be combined with PG-degrading enzymes.  相似文献

The aggregation behavior of 3,5-dihydroxybenzyl alcohol based dendritic polymers through intermolecular hydrogen bonding between periphery situated variable number of OH groups was studied using dynamic light scattering and transmission electron microscopy. Size of the aggregates was found to be dependent on the generation number of dendrimers: generations 1-3 form aggregates above critical aggregation concentration (cac), while higher generations (4-5) aggregate even at concentrations much lower than cac. Encapsulation of the dye disperse red 1 (DR1) into dendrimer aggregates was accompanied by a blue shift in the UV-Vis absorption spectrum of DR1.  相似文献

Abstract

Dendritic macromolecules are hyperbranched polymers that emanate from a central core, have a defined number of generations and functional end groups, and are synthesized in a stepwise process by a repetitive reaction sequence. This hyperbranched topology results in a unique series of physical and chemical properties exhibited by these molecules which, in turn, could be exploited in a number of diverse possible applications, such as nanoscale catalysis, micelle mimics, immuno-diagnostic and NMR imaging agents, chemical sensors, molecular antennae, just to name a few.

Nonetheless, if on one hand the synthesis procedure allows for control over parameters such as size, shape and reactivity (and hence, on final properties), on the other hand it really hampers the production of large quantities of these materials. Accordingly, their cost is still quite high and, therefore, the materials available for characterization is still rather limited.

Since, however, their full application potentials (particularly in material science and engineering) will not be realized before the understanding of their physical properties is considerably more advanced, in this work we report the results obtained on structural details and related properties of several amine- and nitrile-terminated poly(propylene)imine dendrimer generations by computer simulation studies and discuss them in the light of (scarce) available experimental data.  相似文献

An efficient synthesis of a series of carbosilane dendrimers uniformly functionalized with sialyl α(2 → 3) lactose (Neu5Acα(2 → 3)Galβ(1 → 4)Glcβ1→) moieties was accomplished. The results of a preliminary study on biological responses against influenza virus hemagglutinin, using the sialyl lactose clusters showed unique biological activities on the basis of the structure–activity relationship according to the carbosilane scaffolds.  相似文献

Dendrimeric-T and dendrimeric-A with 36 nucleotides were synthesized using phosphoramidite reagents. These dendrimers contain the nucleosides dA and dT, which are the components of the dA-dT base pair. Branching was obtained using a bifurcated and trifurcated reagent. The dendrimers were purified by ion-exchange chromatography. No specific interactions between these dendrimers were observed using atomic force microscopy. However, the dendrimeric-T is able to disrupt monolayers of fatty acids, resulting in a reorientation of the lipids (from head-to-tail to head-to-head) as demonstrated by scanning tunneling microscopy (STM).  相似文献
