We studied the impulse activity of neurons of the basal and lateral amygdalar nuclei generated when experimental animals (rats) performed fast stereotyped food-procuring movements by the forelimb. Within the basolateral amygdala, there are neurons whose activity is related to different stages of getting off the food, and according to the characteristics of their spiking these neurons should be divided into a number of subpopulations. Activation forestalling the movement initiation by 0.5-1.0 sec was observed in most neurons of the basolateral amygdala; this is considered a manifestation of excitation related to a motivation component of the food-procuring behavior. Activation of amygdalar neurons following movement initiation can result from generation in this structure of additional excitation necessary for successful performance of a complete food-procuring motor cycle.  相似文献

Summary An assay is described whereby Eco RI restriction fragment length polymorphisms of mitochondrial and chloroplast DNAs can definitively identify cytoplasms of interest in Brassica crop development. Restrictable mitochondrial and chloroplast DNA is extracted from as little as 2–3 g and 0.5 g leaf tissue, respectively, and the donor plants are able to continue to develop in a normal manner. An unknown cytoplasm can be identified in three days, which is a considerable saving in time and labor compared to the several years required by traditional methods. The assay is very inexpensive and should be established as a routine procedure in laboratories involved in sexual or somatic Brassica hybrid production.  相似文献

Summary When Ca2+, K+ or Cl was injected iontophoretically into the cytoplasm of intactNitella cell, only Ca2+ reversibly inhibited the cytoplasmic streaming. However, when an extremely large current was used, the cytoplasmic streaming was reversibly inhibited irrespective of the ion species. It is difficult to completely understand the life history of an intellectual excluding an understanding of his family upbringing and formative years. Family upbringing and childhood environment, often the less known part of a life history, play crucial roles in shaping the ideas and values individuals espouse in their adult life. Notwithstanding, this paper is not concerned with Don C. Ohadike’s childhood. It rather focuses on the professional career of our able historian – that is the part of his life as revealed by his most outstanding published writings. Ohadike’s published works contain a wellspring of idioms that tell much about his values, quality of mind, and his mission as an African historian. Ohadike was a humanist, an African patriot, and a nationalist crusader. His entire philosophy centered on safeguarding his African identity in an emergent world of cultural imperialism. The funds for this research were provided by a NEH-funded fellowship at the Schomburg Center, New York in the Spring of 2007. Summary A linear mitochondrial plasmid reported to be associated with cytoplasmic male sterility in the genus Brassica was analyzed. A protein was found to be associated with the 5 ends of the plasmid. The entire plasmid was cloned by the homopolymer tailing technique via free hydroxyl groups present at its 3 ends. DNA sequence analysis of the cloned plasmid revealed a perfect terminal inverted repeat of 325 base pairs. Southern hybridization and restriction enzyme mapping analysis confirmed colinearity of the native plasmid and the clone, which showed significant homology with organelle DNA but not with nuclear DNA. Under high-stringency hybridization conditions, an internal 4.6 kb fragment of the 11.5 kb plasmid hybridized to the main mitochondrial genome in several species. Although the hybridization signal was weaker, the chloroplast genome also showed homology to the mitochondrial plasmid. The plasmid was undetectable at a molar ratio of less than 1/10 000 of the main mitochondrial genome in some lines of Brassica and Raphanus that contain the Ogura male sterile cytoplasm (cms). The absence of the plasmid in these sterile lines demonstrates that the plasmid is not required for the expression and maternal inheritance of male sterility.  相似文献

By means of pH-sensitive microelectrodes, cytoplasmic pH has been monitored continuously during amino-acid transport across the plasmalemma of Riccia fluitans rhizoid cells under various experimental conditions. (i) Contrary to the general assumption that import of amino acids (or hexoses) together with protons should lead to cytoplasmic acidification, an alkalinization of 0.1–0.3 pHc units was found for all amino acids tested. Similar alkalinizations were recorded in the presence of hexoses and methylamine. No alkalinization occurred when the substrates were added in the depolarized state or in the presence of cyanide, where the electrogenic H+-pump is inhibited. (ii) After acidification of the cytoplasm by means of various concentrations of acetic acid, amino-acid transport is massively altered, although the protonmotive force remained essentially constant. It is suggested that H+-cotransport is energetically interconnected with the proton-export pump which is stimulated by the amino-acid-induced depolarization, thus causing proton depletion of the cytoplasm. It is concluded that, in order to investigate H+-dependent cotransport processes, the cytoplasmic pH must be measured and be under continuous experimental control; secondly, neither pH nor the protonmotive force across a membrane are reliable quantities for analysing a proton-dependent process.Abbreviations 3-OMG 3-oxymethylglucose - pHc cytoplasmic pH - m electrical potential difference across the respective membrane, i.e. membrane potential - H+/F (=pmf) electrochemical proton gradient  相似文献

Summary The effect of inhibition of cytoplasmic streaming on intercellular passage of carboxyfluorescein (CF) in staminal hairs ofS. purpurea was examined. Tip cells of staminal hairs were microinjected with buffered-CF. Cytoplasmic streaming was then inhibited by addition of KCN or NaN3 to the external bathing solution. In separate experiments, cytoplasmic streaming was inhibited by microinjection of cytochalasin D along with the buffered-CF. CF passage over a 5 minutes treatment period was monitored by video fluorescence microscopy and video intensity analysis. Cytoplasmic streaming ceased within 1 minute of inhibitor agent treatment, however, little change in the kinetics of intercellular passage was noted over the 5 minute experimental period. Th us, cytoplasmic streaming plays no major role in the regulation of intercellular passage of the hydrophilic, negatively charged molecule CF.The work is dedicated to professor Saal Zalik, Department of Plant Science, University of Alberta, on his 65th birthday.  相似文献

Summary Mitochondrial DNA has been isolated from paired lines of pearl millet maintainer and cytoplasmic male sterile plants. Evaluation of the DNA by agarose gel electrophoresis shows that good quality DNA of high molecular weight can be obtained from mitochondria of both maintainer and male sterile pearl millet.  相似文献

Summary The egg and central cells of Plumbago zeylanica have an average volume of 543,000 m3 and 2,560,000 m3 respectively, with surface areas of 38,600 m2 and 154,000 m2. The egg contains an average of 39,900 mitochondria and 730 plastids. The majority of the plastids are perinuclear (> 60%) with less than 40% in lateral areas or near the filiform apparatus. After fertilization, the number of maternal organelles exceeds paternal organelles by a ratio of 11,000 for mitochondria and 154 for plastids. The central cell contains an average of 178,700 mitochondria and 1,840 plastids. After fertilization, these organelles far exceed the number of sperm organelles transmitted, by a ratio of approx. 14,000 for plastids and 1820 for mitochondria. Biparental inheritance of plastids in the embryo is possible, but not favored; the only comparable data in Oenothera and Impatiens reveals that biparental inheritance is possible in up to 124 ratios. Plants lacking biparental plastid inheritance do not contain plastids in the sperm, and thus the presence of even few sperm plastids may result in expression. The number of paternal mitochondria transmitted into the central cell is greater than that transmitted into the egg as the result of preferential fertilization with the mitochondrion-rich dimorphic sperm cell, although the ratio of paternal to maternal mitochondria is 11,000 in the egg and 1820 in the central cell. The similarity in these ratios suggests that there is a critical dosage of mitochondria that is permissible within the zygotic and endospermatic lineages. This may represent either: (1) a maximum permissible value to prevent expression of paternal mitochondrial genome, (2) a minimum ratio required in order to permit recombination of maternal and paternal mitochondrial genomes, or (3) a cytoplasmic genome balance number.Abbreviations mtDNA mitochondrial DNA - Sua sperm cell unassociated with the vegetative nucleus - Svn sperm cell physically associated with the vegetative nucleus  相似文献
