The aim of this work was the evaluation of fluorescence ITS-PCR (f-ITS) as a molecular tool to analyze the microbial community involved in the biodeterioration of cultural heritage surfaces. As a case study we analyzed by f-ITS ninety-two bacterial strains isolated from a medieval fresco and the surrounding air environment. The internal transcribed spacer between the 16S and 23S rRNA genes was amplified, and then the fluorescently labeled PCR products were separated by capillary electrophoresis. Bacterial strains were identified by 16S rDNA sequencing. The f-ITS electropherograms showed different profiles coherent with the affiliation of the strains at the genus and species levels. Among the isolates obtained from the fresco surface, those belonging to the genus Bacillus were the most prevailing exhibiting 8 different f-ITS profiles. The airborne bacilli exhibited only 2 of these 8 profiles. Staphylococcus were mostly isolated from air and produced 4 different profiles. Pseudomonas isolates presented 3 different profiles, and one of them was typical of Pseudomonas putida. Members of the other genera produced their distinctive profiles. Summary In gene-for-gene systems, specificity of hostparasite interactions is most often estimated qualitatively using the symbols +, –, (i.e. susceptibility and/or resistance). In large sets (interaction patterns) it becomes impossible to analyze numerous data by mere comparison. This is overcome by application of cluster analysis. In our experiments the methods in question were used to estimate the data obtained in a study on interactions between more than 220 Lactuca sativa cultivars and 12 Bremia lactucae physiological races (isolates) of Czechoslovak origin. The matrix of similarity coefficients was analyzed by hierarchical clustering. Similarity and/or dissimilarity of host R-genotypes was graphically expressed using the method of two principal components. The results obtained are related to genetic constitution of race specific resistance of the host and the possibility of predicting effective resistance sources. 相似文献

按月采样,分析测定虫草蝠蛾幼虫体微量元素的组成。应用Q模式系统聚类方法,分析了幼虫受环境影响引起的元素代谢变化。结果表明虫体所含元素与环境温度的变化及自身的生理活动密切相关。5、10月份幼虫组的元素含量相近,前者正当幼虫结束休眠后恢复活动的时期,后者是幼虫处于准备进入越冬的前期,两组幼虫此时均处于取食高峰期。3、4月份组的亦较接近,幼虫正渐恢复活动但不取食。8月份幼虫蜕皮前后,消耗较大,需摄取大量食物。计算结果表明7、9月份的元素含量接近,这与上述现象有一定联系;应用对应因子分析法得到的结果是:元素Fe、P对10、11月份幼虫组的贡献值显著;Na、Ca、Mg对8、9月份的贡献值显著;Cu、Zn、Co、Cd、Si对4、5月份的贡献值显著。 相似文献

This paper examines the pattern(s) of sexual dimorphism within the upper dentition ofProconsul specimens from the early Miocene of east Africa. These fossils are compared against the corresponding dentition ofPan troglodytes andGorilla gorilla using principal components and cluster analyses. This paper demonstrates that both sexes ofPan andGorilla are characterized by their own distinctive shape patterns. It is also demonstrated that someProconsul specimens examined here display a pattern that is dissimilar from otherProconsul specimens also examined. This suggests that at least two species ofProconsul may have to be recognized as having lived in this region during the early Miocene. The identification of distinct patterns

Xylans, the major portion of the hemicellulose of plant cell walls and grasses, are heteropolymers consisting principally of xylose and arabinose. Microbial xylanases with different multiplicities and properties are reported. Most studies on the mode of action of these xylanases have been carried out with fungi and there is very little information available on bacterial xylanases. Fungal xylanases have three or more substrate binding sites: for exampleAspergillus niger, Ceratocytis paradoxa, Cryptococcus albidus andChainia sp. endoxylanases have four to seven subsites with the catalytic site located at the centre of these sub-sites. The analysis of these sub-sites is either by kinetic or end-product analysis studies. Kinetic studies are used for exo-type enzymes while the end-product analysis studies are more convenient for endo-type enzymes. The vegetation of a dune slack at Mason Bay, Stewart Island, New Zealand was found to comprise a mosaic of communities. Although the broad vegetational patterns could be correlated with the depth of the water table, the patterns were far from simple. The gel-to-fluid first-order melting transition of lipid bilayers is simulated by the use of a microscopic interaction model which includes a variable number of lipid-chain conformational states. The development of numerical syntaxonomy during its first 20 yr is reviewed. The use of methods of numerical classification and ordination is the dominating feature of the development. National and local phytosociological data banks were established, large data sets handled and many important vegetation monographs were methodically based on multivariate data analysis. Particularly the development in Italy, the Netherlands, Czechoslovakia, and Sweden contributed to new theoretical elements of numerical syntaxonomy. Ordination became a common tool of searching for reticulate synsystematic relations between community types. The most popular ordination techniques have been Principal Components Analysis and Detrended Correspondence Analysis. Hierarchical agglomerative techniques of clustering still prevail in classification, although the divisive strategy of TWINSPAN has also become an effective tool for phytosociological clustering and table sorting.
