Objective
Proteoglycan 4 (Prg4) has emerged from human association studies as a possible factor contributing to weight gain, dyslipidemia and insulin resistance. In the current study, we investigated the causal role of Prg4 in controlling lipid and glucose metabolism in mice.Methods
Prg4 knockout (KO) mice and wild-type (WT) littermates were challenged with an obesogenic high-fat diet (45% of total calories as fat) for 16?weeks. To further stimulate the development of metabolic alterations, 10% fructose water was provided starting from week 13.Results
Prg4 deficiency only tended to reduce diet-induced body weight gain, but significantly improved glucose handling (AUC: ?29%; p?<?0.05), which was also reflected by a tendency towards a reduced HOMA-IR score (?49%; p?=?0.06 as compared to WT mice). This coincided with lower hepatic expression of glycolysis (Gck: ?30%; p?<?0.05) and lipogenesis (Acc: ?21%; p?<?0.05 and Scd1: ?38%; p?<?0.001) genes, which translated in significantly lower hepatic triglyceride levels (?56%; p?<?0.001) in Prg4 KO mice as compared to WT mice. Prg4 KO mice likely had lower glucose utilization by skeletal muscle as compared to WT mice, judged by a significant reduction in the genes Glut4 (?29%; p?<?0.01), Pfkm (?21%; p?<?0.05) and Hk2 (?39%; p?<?0.001). Moreover, Prg4 KO mice showed a favorable white adipose tissue phenotype with lower uptake of triglyceride-derived fatty acids (?46%; p?<?0.05) and lower gene expression of inflammatory markers Cd68, Mcp1 and Tnfα (?65%, ?81% and ?63%, respectively; p?<?0.01) than WT mice.Conclusion
Prg4 KO mice are protected from high-fat diet-induced glucose intolerance and fatty liver disease. 相似文献4.

In Cephalotascus sinensis (Rehd. et Wils. ) Li the somatic chromosome number was found to be of 2n=24. Eleven pairs of chromosomes possessed their centromeres at median or median-submedian regions. The shortest pair of chromosomes was the SAT chromosome which possessed their centromeres at the submedian regions. The sex chromosomes were demonstrated by the Giemsa C-banding technique. The sexual determination mechanism of female was WZ type (2n=24=22A+WZ), and that of male was ZZ type (2n=24=22A +ZZ). 相似文献

Cephalotaxus oliveri Mast. which is native to China (Hunan, Hupei, Sichuan, Yunnan and Guangdong, etc.) belongs to family Cephalotaxaceae. Recently, a series of research work on alkaloids from Cephalotaxus genus has been reported because of their anticancer activities. However, other major active components-biflavones are neglected. In order to discuss the relationship of gymnosperm and study the active components we isolated three biflavones from the le- aves of Cephalotaxus oliveri Mast. Two of them co-3, co-4, are known compounds ie amentoflavone-4',4"',7,7"-tetramethyl ether and sciadopitysin, and the third one co-2, is a new compound named oliveriflavone. It's structure was elucidated as amentoflavone-4',4'",7,7"-ether by UV, IR, 1HNMR, MS and chemical reactions. 相似文献

Three species of evergreen Cephalotaxus (C. fortunei Hook. f., C. sinensis Li and
C. harringtonia cv. Fastigiata) were used as materials to study some functional properties of
chloroplasts. It is found that the oxygenevolving capacity of the chloroplasts from these plants
is inhibited but partial reaction of PS-II and effect of Mg2+ on energy distribution between
two photosystems are detectable during the winter. Seasonal effects on the functional properties
of chloroplasts from evergreen Cephalotaxus are similar to that of conifer chloroplasts.
The ratio between F685, F695 and F735 of fluorescence emission spectra at 77°K of chloroplasts is different among these three species. It is found by using SDS-PAGE that the number of polypeptide resolved from thylakoid membrane of C. harringtonia cv. Fastigiata substantially differs from that of C. fortunei Hook.f. and C. sinensis Li. The result shows that the
fluorescence emission spectrum feature and polypeptide composition of thylakoid membrane may
be used as a tool for systematics of the genus Cephalotaxus. 相似文献

Glioblastoma, an aggressive brain tumor, has a poor prognosis and a high risk of recurrence. An improved chemotherapeutic approach is required to complement radiation therapy. Gold(I) complexes bearing phosphole ligands are promising agents in the treatment of cancer and disturb the redox balance and proliferation of cancer cells by inhibiting disulfide reductases. Here, we report on the antitumor properties of the gold(I) complex 1-phenyl-bis(2-pyridyl)phosphole gold chloride thio-β-d-glucose tetraacetate (GoPI-sugar), which exhibits antiproliferative effects on human (NCH82, NCH89) and rat (C6) glioma cell lines. Compared to carmustine (BCNU), an established nitrosourea compound for the treatment of glioblastomas that inhibits the proliferation of these glioma cell lines with an IC50 of 430 μM, GoPI-sugar is more effective by two orders of magnitude. Moreover, GoPI-sugar inhibits malignant glioma growth in vivo in a C6 glioma rat model and significantly reduces tumor volume while being well tolerated. Both the gold(I) chloro- and thiosugar-substituted phospholes interact with DNA albeit more weakly for the latter. Furthermore, GoPI-sugar irreversibly and potently inhibits thioredoxin reductase (IC50 4.3 nM) and human glutathione reductase (IC50 88.5 nM). However, treatment with GoPI-sugar did not significantly alter redox parameters in the brain tissue of treated animals. This might be due to compensatory upregulation of redox-related enzymes but might also indicate that the antiproliferative effects of GoPI-sugar in vivo are rather based on DNA interaction and inhibition of topoisomerase I than on the disturbance of redox equilibrium. Since GoPI-sugar is highly effective against glioblastomas and well tolerated, it represents a most promising lead for drug development.
