The post-flowering stalk rots (PFSR) are a complex disease, which are widely distributed in almost all the maize growing regions across the globe. A number of fungi are involved in causing decay of the pith resulting in pre-mature wilting of the plants. Most of the commercially grown cultivars have shown a high level of disease incidence at the grain filling stage. A systematic breeding programme on PFSR was initiated in India in collaboration with Asian Regional Maize Program of CIMMYT. Under this programme, germplasm screening was carried out at four ‘hot spot’ locations in India for different diseases: Hyderabad (Cephalosporium maydis), Udaipur (Fusarium moniliforme), Ludhiana and Delhi (Macrophomina phaseolina). Across the locations, promising maize genotypes were artificially inoculated using the toothpick method, year after year; resistant plants were selfed to derive resistant inbreds. After extensive screening, three resistant lines, namely PFSR-13-5, JCY2-2-4-1-1-1-1 and JCY3-7-1-2-1-b-1, were identified. In addition, the resistance level of five pools/populations; (PFSR (Y)-C1, PFSR (white), Extra-early (White), P-100, P-300 and P-345) was upgraded to an acceptable level. These genotypes may prove useful for utilisation in breeding cultivars for resistance to PFSR.  相似文献

The microbial transformation of the 17alpha-ethynyl-17beta-hydroxyandrost-4-en-3-one (1) (ethisterone) and 17alpha-ethyl-17beta-hydroxyandrost-4-en-3-one (2) by the fungi Cephalosporium aphidicola and Cunninghamella elegans were investigated. Incubation of compound 1 with C. aphidicola afforded oxidized derivative, 17alpha-ethynyl-17beta-hydroxyandrosta-1,4-dien-3-one (3), while with C. elegans afforded a new hydroxy derivative, 17alpha-ethynyl-11alpha,17beta-dihydroxyandrost-4-en-3-one (4). On the other hand, the incubation of compound 2 with the fungus C. aphidicola afforded 17alpha-ethyl-17beta-hydroxyandrosta-1,4-dien-3-one (5). Two new hydroxylated derivatives, 17alpha-ethyl-11alpha,17beta-dihydroxyandrost-4-en-3-one (6) and 17alpha-ethyl-6alpha,17beta-dihydroxy-5alpha-androstan-3-one (7) were obtained from the incubation of compound 2 with C. elegans. Compounds 1-6 exhibited tyrosinase inhibitory activity, with compound 6 being the most potent member (IC(50)=1.72 microM).  相似文献

Abstract Amino acid sequence alignment of the Cephalosporium acremonium isopenicillin N synthase (cIPNS) to similar non-heme Fe2+-containing enzymes from 28 different sources (bacterial, fungal, plant and animals) revealed a homologous region of high sequence conservation containing an invariant histidine residue at position 272 in cIPNS. The importance of this histidine residue in cIPNS was investigated through site-directed mutagenesis by replacing the histidine residue with leucine. The mutated gene was verified by DNA sequence analysis and expressed in Escherichia coli . When analyzed by denaturing gel electrophoresis and immunoblotting, the mutant cIPNS had identical mobility as that of the wild-type enzyme. Enzyme studies on the mutant enzyme showed loss of enzymatic activity indicating that His272 is essential for the catalytic function of cIPNS, possibly as a ligand for iron binding.  相似文献

Summary Rice oil significantly affected cephalosporin C production in a 2.5-l bioreactor culture of Cephalosporium acremonium M25. To improve cephalosporin C production, the feed conditions of rice oil were optimized. Reducing the feed rate of rice oil improved cephalosporin C production to 1.01 g/l when the consumption rate of rice oil decreased. Overall, under optimal feed conditions in the 2.5-l fed-batch culture, cephalosporin C production increased about four times compared to before optimization.  相似文献

A recent report (Patino et al., (1989) FEMS Microbiol. Lett. 58, 139-144) described the low level expression, in Escherichia coli, of the Isopenicillin N Synthase (IPNS) gene from Cephalosporium acremonium under the control of strong promoters. We report here our work on the expression of the IPNS gene. Plasmids containing the IPNS gene under the control of the trp or trc promoters directed synthesis of high levels of active IPNS in E. coli. Constitutive and inductive high level IPNS expression systems have been developed. Importantly, the expression vectors do not encode beta-lactamase so IPNS activity can be determined directly by biological assays. Analysis by nmr verified that the IPNS produced from these expression systems catalysed the conversion of delta-(L-alpha-aminoadipoyl)-L-cysteinyl-D-valine (LLD-ACV) to isopenicillin N in high yield.  相似文献

Two pregnane steroids, pregnane (1) and 3β-hydroxypregnane (2), were oxidized by fermentation with the fungus Cephalosporium aphidicola. The fermentation of pregnane (1) yielded 3β-hydroxypregnane (2) and 3β, 6β,11α-trihydroxypregnane (3), while that of 3β-hydroxypregnane (2) afforded 6β,11α-dihydroxypregn-3,20-dione (4), 3β,6β,15α-trihydroxypregn-20-one (5) and 3β,5α,11α-trihydroxypregn-20-one (6). 为了调控长链ω 3多不饱和脂肪酸的发酵 ,对用于脂肪酸发酵调控的 2个指标 18∶3(α ) /18∶2和∑C18/∑C16进行探讨。对 2个指标从化学反应平衡角度进行阐述 ,进一步证明在合适的条件下 ,头孢霉 18∶3(α ) /18∶2的变化趋势和二十二碳六烯酸变化趋势一致 ;轮梗霉∑C18/∑C16和二十碳五烯酸变化趋势一致。 18∶3(α ) /18∶2和∑C18/∑C16可以作为发酵生产二十二碳六烯酸和二十碳五烯酸的调控指标。  相似文献
