The first and in-depth chemical investigation of the South China Sea soft coral Sinularia scabra has resulted to the isolation of a library of diverse cembrane type diterpenoids, including six new compounds, namely xiguscabrates A and B (1 and 2), xiguscabral A (3), xiguscabrols A and B (4 and 5), and 8-epi-xiguscabrol B (6), and twenty-seven known analogs (733). Their structures were elucidated by extensive spectroscopic analysis and by the comparison with literature data. In bioassay, several isolates exhibited inhibitory effects on the ConA-induced T lymphocytes and/or LPS-induced B lymphocytes proliferation. Among them, compound 24 showed considerable specific inhibition on B cell proliferation, with IC50 value of 4.4 μM and selectivity index (SI) of 10.9. The structure-activity relationship (SAR) of the tested metabolites was analyzed, and the further mechanism study of the specific B-cell targeted immunosuppressive compound 24 on purified CD19+ B cells was also performed to uncover the effects on the function and maturity of B cells, including cytokines production, abnormal activation, antigen presenting capacity and plasma cells formation.  相似文献

Seven new polyoxygenated cembranoids possessing an α‐methylene‐γ‐lactone group, crassocolides G–M ( 1 – 7 , resp.), have been isolated from the AcOEt extract of the Formosan soft coral Sarcophyton crassocaule. The structures of compounds 1 – 7 were established by detailed spectroscopic analyses, including 2D‐NMR spectroscopy (1H,1H‐COSY, HMQC, HMBC, and NOESY), while the absolute configuration of 1 was determined using a modified reaction of Mosher's method. The cytotoxicity of compounds 1 – 7 against a limited panel of cancer cell lines was also determined.  相似文献

The new cembranoid diterpene danielid ( 1 ) along with 3α‐ethoxyfuranocembranoid 2 , pukalide ( 3 ), 13α‐acetoxypukalide ( 4 ), furanocembranoid 5 , and furanosesquiterpene 6 have been isolated from the soft coral Sinularia asterolobata. The furanocembranoid diterpene 11β,12β‐epoxypukalide ( 7 ) and the sesquiterpene (?)‐bicyclogermacrene ( 8 ) have been obtained from the soft coral Litophyton arboreum. The structures were elucidated primarily by NMR spectroscopy. The furanocembranoids 2, 4 , and 5 show good antiproliferative activities against the cell lines L‐929 and K‐562, and weak cytotoxic effects on HeLa cells.  相似文献

1.Phencyclidine (PCP) is an inhibitor of the nicotinic acetylcholine receptor (AChR) with characteristics of an open-channel blocker. The location of PCP binding site on the AChR molecule is unknown.2.PCP inhibits the AChR from electric organ with a higher potency than muscle AChR. To find the molecular basis of this difference, we expressed the two native and six hybrid receptors, and two receptors containing mutated mouse subunits in Xenopus laevis oocytes. The inhibition of ACh-induced current in these receptors by PCP was studied using whole-cell voltage-clamp. All hybrid receptors generated robust ACh-induced currents, while incomplete receptors (-less or -less) did not.3.PCP potency was higher on hybrids containing Torpedo and subunits regardless of the and subunit origin. A mouse subunit containing the asparagine 6 to the serine mutation in the M2 segment conferred a high sensitivity to PCP.4.These results support the conclusion that the amino acid residues at the position 6 of the M2 segments contribute to the PCP potency difference between Torpedo and mouse receptors.5.Another noncompetitive inhibitor of the AChR, the cembranoid eupalmerin acetate (EUAC), also inhibited the electric organ receptor with a somewhat higher potency than muscle AChR. However, the IC50 values for EUAC inhibition of hybrid receptors did not follow the pattern observed for PCP. Therefore, these two inhibitors interact differently with the AChR molecule.  相似文献

The new cembrane diterpene (3E,11E)‐cembra‐3,8(19),11,15‐tetraene‐7α‐ol ( 1 ), nephthenol ( 2 ), and all‐trans‐peridinin ( 3 ) have been isolated from the soft coral Litophyton arboreum. The tetraterpene 3 , (+)‐7,8‐epoxy‐7,8‐dihydrocembrene C ( 4 ), emblide ( 5 ), sarcophytoxide ( 6 ), sarcoglaucol‐16‐one ( 7 ), guajacophine ( 8 ), and 1,4‐peroxymuurol‐5‐ene ( 9 ) have been obtained from the soft coral Sarcophyton ehrenbergi. The compounds were characterized primarily by NMR spectroscopy. Some of the terpenes were tested for their antiproliferative activity against the cell lines HUVEC and K‐562 and for cytotoxicity against the cell line HeLa, and they showed moderate activities.  相似文献
