Allelopathy has been postulated as a factor in the colonisation success of Caulerpa racemosa var. cylindracea, an introduced chlorophyte alga which has become invasive in the Mediterranean Sea. In order to reveal their possible phytotoxic activity, secondary metabolites were isolated from a population growing in the Gulf of Naples (Italy) and tested on leaf tissue of the native seagrass Cymodocea nodosa, which often co-occurs with Caulerpa in nature. The seaweed metabolites were identified by 1H and 13C NMR analysis and leaf portions of C. nodosa were exposed to crude diethyl ether extract and purified compounds in the laboratory, under controlled conditions of irradiance and temperature for a period of 144 h. Eventual changes in the photosynthetic performance of the seagrass tissue following the treatment were tested by monitoring its optimal quantum yield with a pulse amplitude modulated (PAM) fluorometer. One of the identified compounds, caulerpenyne, showed a phytotoxic effect, while the other purified metabolites did not reveal any effect at the assayed concentrations. Thus, our results seem to suggest a possible allelopathic activity of caulerpenyne, which may play a role in the successful competition of the invasive C. racemosa var. cylindracea with native macrophytes, such as seagrasses.  相似文献

Caulerpenyne (Cyn) is a cytotoxic compound firstly isolated in 1978 from Caulerpa prolifera. This metabolite, constituted by a highly reactive diacetoxybutadiene moiety, exhibited a wide range of biological properties with mainly antibacterial properties and antitumoral activities. Few structure–activity relationships (SAR) are available to design more potent bioactive derivatives by pharmacomodulation. Cyn can be produced by total synthesis or extracted from natural sources in particular the green alga Caulerpa taxifolia. Since conventional chromatographic procedures to isolate Cyn from C. taxifolia are time- and solvent-consuming, it was crucial to find a more efficient process to obtain pure Cyn. In our study, Cyn has been purified from C. taxifolia with two different techniques: Centrifugal partition chromatography (CPC) and a classical chromatographic process. The comparative study showed that CPC constitutes a very simple and efficient process to access Cyn.  相似文献

The proliferation of the green marine alga Caulerpa taxifolia in the Mediterranean led us to investigate the toxic effects on marine organisms of caulerpenyne (Cyn), the major secondary metabolite synthesized by the alga. This study was performed on sea urchin eggs (Paracentrotus lividus) and isolated hepatocytes from the sea bream (Sparus aurata), in which accumulation of the toxins by metabolic processes may be of significance. Cyn provoked an acidification of seawater containing both unfertilized and fertilized eggs, as revealed by a titrable efflux of protons. The pHi in unfertilized eggs continuously increased in the presence of Cyn, whereas there was a biphasic response in both fertilized eggs and isolated hepatocytes, with a decrease of the pHi followed by recovery to the initial value. Cyn inhibited the accumulation of 14C-methylamine in acidic granules present in the cortical zone of sea urchin eggs. Dicyclocarbodiimide (DCCD), a well-known H+-ATPase inhibitor, provoked a similar inhibition. Both molecules increased pH in the acidic compartments of isolated bream hepatocytes. These results suggest that Cyn inhibits intracellular sequestration of protons and thus liberates protons into the cell cytoplasm from which they leak toward the extracellular medium. © 1997 John Wiley & Sons, Inc.  相似文献

Invasion of the Mediterranean Sea by the two world-wide famous exotic algae species, Caulerpa taxifolia and Caulerpa racemosa, is still a problem and has adverse effects on the Mediterranean sublittoral ecosystem. Biological control studies revealed that the two native Sacoglossans, Oxynoe olivacea and Lobiger serradifalci, may have an effect on the expansion of invasive Caulerpa spp. in the Mediterranean. In the framework of this study, antioxidant enzyme activities, such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px), lipid peroxidation (LPO) and oxidized glutathione (GSSG) levels, as oxidative stress markers in L. serradifalci and O. olivacea were determined at two different temperature conditions (20 and 27 °C). In both species, SOD, CAT and GSH-Px activities were found to be positively correlated with temperature. The SOD activities in L. serradifalci were higher than those in O. olivacea at both temperatures, whereas the CAT and GSH-Px activities were significantly (p<0.05) higher in O. olivacea, compared to L. serradifalci. As expected, both species showed decreased LPO levels at 27 °C compared to 20 °C. GSSG level at 27 °C in O.olivacea was significantly (p<0.05) higher than that of 20 °C. On the other hand, no statistical (p>0.05) difference in L.serradifalci existed between GSSG levels at two temperatures. But, despite the variations in the antioxidant enzyme activities, there was no significant difference in LPO levels between the species, suggesting that the oxidative consequences of a given environmental condition may vary among different species. Inasmuch as the GSSG levels were in accordance with antioxidant enzyme activities, GSH might have acted as a cofactor of GSH-Px and an individual antioxidant in these sea slugs.  相似文献

Invasive algae alter the structure and function of ecosystems. The red algae Lophocladia lallemandii grows in the West Mediterranean epiphyting the green algae Caulerpa taxifolia, another invasive alga. Our aim was to determine whether the invasive algae Lophocladia, recently introduced in the West Mediterranean, induces oxidative stress and an antioxidant response in Caulerpa taxifolia. We measured the caulerpenyne production, the activities of antioxidant enzymes and the levels of markers of lipid peroxidation in Caulerpa taxifolia. Caulerpenyne concentration was significantly higher in Caulerpa taxifolia epiphytized by Lophocladia. End-products of lipid peroxidation -malondialdehyde (MDA) and 4-hydroxinonenal (4-HNE)- were significantly increased in Caulerpa taxifolia samples from the station with Lophocladia. Antioxidant enzyme activities -catalase and superoxide dismutase (SOD)-, as well as H2O2 production significantly increased in the Lophocladia station compared to the control station. The activities of catalase and SOD determined in Lophocladia lallemandii showed no significant differences between Lophocladia living alone and Lophocladia epiphytating Caulerpa taxifolia. Caulerpa taxifolia epiphytized by Lophocladia lallemandii responded by increasing the caulerpenyne and H2O2 production and the antioxidant enzymes activities as a defensive mechanism against the new invasive algae.  相似文献
