Complete sequences of the ribosomal internal transcribed spacers (ITS1 and ITS2) and karyological characters of the monozoic (unsegmented) tapeworm Atractolytocestus huronensis Anthony, 1958 (Cestoda: Caryophyllidea) from Slovakia were analysed, revealing considerable intra-genomic variability and triploidy in all analysed specimens. Analysis of 20 sequences of each ITS1 and ITS2 spacer yielded eight and 10 different sequence types, respectively. In individual tapeworms, two to four ITS1 and three to four ITS2 sequence types were found. Divergent intra-genomic ITS copies were mostly induced by nucleotide substitutions and different numbers of short repetitive motifs within the sequence. In addition, triploidy was found to be a common feature of A. huronensis. The karyotype of Slovakian A. huronensis possesses three sets of chromosomes (3= 24, = 4m + 3st + 1 minute chromosome), similar to the previously described triploidy in conspecific tapeworms from North America. Fluorescent in situ hybridisation (FISH) with a ssrDNA probe revealed two distinct rDNA clusters for each homologue of the triplet number 2. To date, A. huronensis is the only cestode species in which intra-individual ITS sequence variants were found in parallel with its triploid nature and multiple rDNA loci. Some of these molecular and genetic features were observed in several other species of basal or nearly basal tapeworms of the orders Caryophyllidea and Diphyllobothriidea, which indicates that the phenomena may be characteristic for evolutionarily lower tapeworms and deserve more attention in future studies.  相似文献

Swiderski Z. and Mackiewicz J. S. 1976. Electron microscope study of vitellogenesis in Glaridacris catostomi (Cestoidea: Caryophyllidea). International Journal for Parasitology6: 61–73. Mature vitelline follicles consist of cells in various stages of development, progressing from immature cells of gonial type near the periphery to mature ones toward the centre. Maturation, completed before the cell leaves the follicle, is characterized by: increase in cell volume; increase in nuclear surface area restoring the N/C ratio; nucleolar transformation; extensive development of large parallel cisternae of granular endoplasmic reticulum, the shell-protein producing units; development of Golgi complexes engaged in shell globule formation; formation and storage of glycogen in the cytoplasm; simultaneous, independent formation and storage of intranuclear glycogen; progressive increase in the number and size of shell globule clusters; and disintegration of endoplasmic reticulum, degenerative changes, and accumulation of glycogen and shell globule clusters within the cytoplasm associated with a massive accumulation of glycogen in the nucleus. The functional significance of the large amount of nuclear and cytoplasmic glycogen and numerous shell globule clusters is analyzed. Vitellogenesis in G. catostomi is compared with that in other cestodes and trematodes. Some conclusions, concerning the interrelationship between the vitellogenesis pattern and the type of embryogenesis following it, are drawn and discussed.  相似文献

Mitotic and meiotic chromosomes were studied in Giemsa post-stained Feulgen squashes of testicular, ovarian, and vitelline cells. G. catostomi is 2n = 20 with eight pairs of metacentrics and two pairs of submetacentrics in its karyotype. The fundamental number of chromosome arms is FN = 40. At mitotic metaphase the mean total chromosome length (TCL) of the complement was 111·4 μm; the largest chromosomes were 8 μm (7% TCL) and the smallest 3 μm (3% TCL). Karyotype variation was not found among five diploid populations from New York State and Canada. Meiotic activity was abundant in spermatogenesis which proceeds as usual with sperm developing after two meiotic divisions. Chiasma frequency at diakinesis was 37/cell; as many as five chiasmata were observed in a single bivalent. A triploid ‘race’ (3n = 30) of G. catostomi was discovered in Bozenkill Creek near Albany, New York. Karyotype analysis confirms the triploid nature of these variants. Spermatogenesis in triploids is abnormal to the point of complete failure. The presence of an extra set of chromosomes has resulted in a breakdown of meiosis with subsequent sterility of the male system. Eggs remain unfertilized and parthenogenetic reproduction is presumed to occur. The possible origins of this unusual condition and its evolutionary implications are discussed. Ultrastructural observations using transmission and scanning electron microscopy reveal the tegument to be basically similar to that of other cestodes. The syncytial distal cytoplasm is devoid of organelles except for rod-shaped bodies, believed to be secretory vesicles, and lamellated bodies which probably contribute the raw material for new microtriches. There is evidence that these vesicles originate from the Golgi found in the sub-cuticular cells.Three types of microtriches are described: typical ones with well-developed spines, ones with short filaments instead of spines, and ones with no spines. Microtriches with spines are found only on the anterior part of the worm and may serve to anchor the worm. Microtriches on the posterior have no spines and are believed to be primarily involved in the absorption of nutrients. Between these two regions there is a transitional zone where all three types of microtriches can be found. In general the microtriches are quite uniformly distributed throughout the surface of the worm. The presence of cestodarian-like microtriches raises interesting evolutionary questions.Histochemical tests localized acid and alkaline phosphatase activity on various parts of the tegument, as well as on host intestine, while acian blue tests showed that acid mucopolysaccharide levels correspond with the concentration of the tegument vesicles.  相似文献

Kuperman B. I. & Davydov V. G. 1982. The fine structure of frontal glands in adult cestodes. International Journal for Parasitology12: 285–293. The localization and structure of frontal glands of adult cestodes of 20 species from 4 orders: Caryophyllidea, Pseudophyllidea, Proteocephalidea and Cyclophyllidea were studied by light and electron microscopy. In cestodes of various orders there was evidence of differences in the ultrastructure of secretory cells, in the character of the secretion, and in the method of its discharge. The localization of glands in most adult cestodes is in the scolex. In Cyatocephalus, Ligulidae, Bothriocephalus claviceps, they are distributed also in the anterior and middle part of the body. In the species Caryophylleus (C. laticeps) and Khawia sinensis a major gland complex previously not described was discovered. The glands of the cestodes investigated may be divided into three main groups by their structure, localization and functional role in ontogenesis: (1) penetration glands in oncospheres which develop into procercoids and plerocercoids; (2) glands of plerocercoids and adult cestodes which develop in the gut; (3) gland complexes in the representatives of the order Caryophyllidea. The secretion is usually discharged at the scolex region and less frequently on the lateral surface. Three different ways of discharging secretory material from the cestodes body are described.  相似文献
