The mutation of Scenedesmus obliquus strain C-6D is expressed in the dark only. Under these conditions, this strain synthesizes acyclic poly-cis carotenes. Cyclic carotenoids like -carotene or xanthophylls are absent. In the light the normal cyclic carotenes and xanthophylls are synthesized in the all-trans configuration. The poly-cis carotencs present in dark cultures have been analysed and quantitated. It could be shown that these poly-cis carotenes are identical with the poly-cis carotenes synthesized by the tomato mutant Lycopersicon esculentum var. Tangella. The poly-cis pathways, however, are different regulated in the two organisms. The tomato mutant accumulates prolycopene as the major carotene, whereas the mutant C-6D accumulates mainly pro--carotene. Furthermore, the mutation in Tangella is constitutive in light in contrast to Scenedesmus C-6D. Besides that, Scenedesmus C-6D synthesizes a further cis-carotene isomer of phytofluene as well as of -carotene. The configuration of these carotenes still have to be elucidated. The occurrence of this poly-cis carotene biosynthetic pathway by a mutation of only one enzyme, the phytoene desaturase which, however, is only expressed in darkness under heterotrophic conditions, is discussed.  相似文献

The membranes from the chromoplasts of Narcissus pseudonarcissus L. which are derived from the inner envelope membrane are the site of -carotene synthesis from [1-14C]isopentenyl diphosphate. The enzymes involved are partly peripheral membrane proteins (prenyltransferase, phytoene synthase) and partly integral membrane proteins (cis-trans isomerase, dehydrogenase(s), cyclase(s)). A system has been established from isolated intact chromoplasts of Narcissus pseudonarcissus flowers that synthesizes geranylgeraniol, an unknown polyprenoid alcohol, phytoene, and -carotene from [1-14C]isopentenyl pyrophosphate in a good yeild. Long chain pyrophosphates are not accumulated. San 6706 inhibits the dehydrogenation of phytoene, whereas nicotine does not lead to an accumulation of lycopene. Separation and identification of polyprenoid lipids was performed by HPLC. The study was carried out to determine the responses of two parsley (Petroselinum crispum L.) hybrids; plain and curly- leafed to water stress. Besides the control, three irrigation treatments (10, 30 and 50 % of control water amount) were applied. Plant yield (leaf area, stem length and diameter, number of leaves) and quality (fresh and dry weights and concentrations of chlorophyll, carotenes, anthocyanins and vitamin C) were measured. The responses of plant yield and quality varied according to the hybrid and water treatment. The high quality was observed in curly- leafed parsley if compared to plain- leafed parsley, which had higher yield (stem length and diameter, leaf area). Moreover, water stress improved quality but decreased yield parameters in both hybrids.  相似文献

β‐carotene, a type of terpenoid, has many metabolic and physiological functions. In particular, β‐carotene has an antitumor effect. However, the efficacy of β‐carotene against esophageal squamous cell carcinoma (ESCC) remains unclear. In our study, β‐carotene inhibited the growth of ESCC cells and downregulated expression of the Caveolin‐1 (Cav‐1) protein. Cav‐1 protein was expressed only in ESCC cells, not in Het‐1A cells. Moreover, β‐carotene triggered apoptosis, induced cell cycle G0?G1 phase arrest, and inhibited cell migration. To explore the mechanism involved in these processes, we further examined the effect of β‐carotene on the Cav‐1‐mediated AKT/NF‐κB pathway. The results showed that the level of AKT and NF‐κB phosphorylation was dramatically inhibited, which led to an increase in the Bax/Bcl‐2 ratio. Correspondingly, the activity of Caspase‐3 was also enhanced. These data suggest that β‐carotene has an antiproliferative role in ESCC cells and may be a promising chemotherapeutic agent for use against ESCC cells.  相似文献

Changes in the carotenoid content in gonads of two sea urchins species were investigated during maturation. The content of echinenones and carotenes, the two major carotenoid fractions in gonads, is highest for Strongylocentrotus intermedius at the spawning gametogenic stage of gonad maturation for both sexes. For S. nudus, the content of these pigments is highest at stages of active gametogenesis and spawning for males and at the growth stage for females. A comparison of the carotenoid content dynamics during maturation of gonads for males, females and animals at the resting (sexual inactivity) stage was also carried out.  相似文献

The Scenedesmus obliquus mutant C-6D forms untypical poly-cis carotenes and lacks cyclic carotenoids when grown heterotrophically in the dark. After transfer to light, a very fast formation of usual all-trans carotenoids is observed leading to a phenotype very similar to the wild-type. Carotene analysis and quantitative kinetics of carotenoid changes after illumination in the presence of inhibitors which inhibit the three enzymes involved in carotene conversion showed isomerization of poly-cis carotenes, especially pro--carotene, to all-trans isomers. This result and consequently photoisomerization of pro--carotene in solution demonstrated that photoisomerization is the light-dependent mechanism leading to all-trans carotenoids in illuminated cultures. Phycomyces blakesleeanus colour mutants of a new class have been isolated. The mutants form red mycelia when grown in the dark and show vegetative segregation. The affected gene, amedcarE, behaves as an extranuclear genetic factor. The red phenotype ofcarE mutants is caused by the accumulation of -carotene in a form showing spectral shift to longer wavelengths.
