Echinacoside is an important bioactive compound extracted from Cistanche tubulosa which was endangered by overexploitation. It is imperative to find an alternative source. Echinacoside was isolated from Penstemon barbatus (Can.) Roth for the first time. The peak contents of echinacoside are 9.09 ± 0.32 mg/g and 7.25 ± 0.36 mg/g respectively in the leaves and roots annually. The methanolic extracts from 20 g of dried powder of the roots of P. barbatus were pre-purified by AB-8 resin and the fraction containing echinacoside was further purified by conventional high-speed counter-current chromatography (HSCCC) and recycling HSCCC with the solvent system n-butanol–water (1:1, v/v). Totally 42.0 mg echinacoside with a purity of 96.3% was recovered. The recovery rate of echinacoside by recycling HSCCC reached 91.0%. The structure of our echinacoside confirmed by IR, 1H NMR and 13C NMR is identical to the standard sample. This indicates that P. barbatus might be ideal source for preparation of large scale of echinacoside.  相似文献

The dog lipocalin allergen Can f 2 is an important cause of allergic sensitization in humans worldwide. Here, the first crystal structure of recombinant rCan f 2 at 1.45 Å resolution displays a classical lipocalin fold with a conserved Gly-Xaa-Trp motif, in which Trp19 stabilizes the overall topology of the monomeric rCan f 2. Phe38 and Tyr84 localized on the L1 and L5 loops, respectively, control access to the highly hydrophobic calyx. Although the rCan f 2 calyx is nearly identical with the aero-allergens MUP1, Equ c 1 and A2U from mouse, horse and rat, respectively, no IgE cross-reactivity was found using sera from five mono-sensitized subjects. However, clear IgE cross-reactivity was demonstrated between Can f 2 and the cat allergen Fel d 4, although they share less than 22% sequence identity. This suggests a role for these allergens in co-sensitization between cat- and dog-allergic patients.  相似文献

The author presents reviews that identify success factors in music and arts education partnerships between cultural institutions and K-12 schools. She incorporates the evaluation of one Massachusetts partnership, Arts Can Teach (ACT), to examine the connection between partnerships and K-12 arts-program policy decisions. ACT is a collaborative effort among Boston's Wang Center for the Performing Arts, the Lynn Public Schools, and LynnArts, which matched music specialists and teachers in other disciplines with practicing artists for oneyear partnerships. Success factors of the ACT partnership are considered in terms of their similarity to success factors from the literature on music education partnerships. The author discusses implications for increasing and sustaining music and arts education programming and local arts education policy development.  相似文献

Summary Field experiments were conducted during 1979 and 1980 summer seasons on sandy loam soils of low moisture retentive capacity to study the effect of high frequency irrigation at different levels of N on groundnut yield and nutrient uptake (NPK). Four irrigation frequencies (irrigation at 2, 4, 6 and 8 cm cumulative can evaporation, corresponding to irrigation once in 3, 5, 7 and 10 days respectively) and four levels of nitrogen (0, 20, 40 and 60 kg N/ha) were tested in a factorial randomized block design with three replications. Pod yield of groundnut was maximum (3,293 kg/ha) when irrigations were scheduled at 4 cm cumulative can evaporation (once in 5 days). Addition of N did not increase the pod yield. N and P uptake by the crop was maximum (180 kg N and 18 kg P/ha) with high frequency irrigation of scheduling irrigation at 4 cm cumulative can evaporation. Highest uptake of N (183 kg/ha) and P (19 kg/ha) was with a combination of 20 kg N/ha and high frequency irrigation (4 cm CCE). K uptake was low with low irrigation frequency, while it was highest (67 kg K/ha) at 20 kg N/ha.  相似文献

Plantations released from management are vulnerable to transient oscillations until cohort dynamics are broken and the vertical and horizontal structures of the plantation are transformed to those of more natural forests. Cohort-desynchronizing factors such as canopy disturbances are expected to accelerate this process. Using well-established mangrove plantations in Can Gio (Viet Nam) as an example, we tested whether lightning gaps can affect transition dynamics of plantations to more natural forests by damping the amplitude or by shortening the period of oscillations in tree densities. This was done by applying point pattern analyses to remotely sensed data, and by further combining statistical and individual-based modelling. The occurrence of lightning gaps was biased by the forest matrix, which presented a challenge for the point pattern analysis. This problem was solved by using the scattered forest area as a binary mask. A Matern cluster process model was found to be suitable for describing the lightning regime. This statistical model was incorporated into the individual-based mangrove model KiWi, and simulation experiments revealed that: (i) the evenly spaced distribution of the tree cohorts in the plantation supports non-linear transition behaviour, i.e. oscillation of tree density, and (ii) the lightning regime in Can Gio damps the oscillation amplitude but is not sufficient to prevent the latter nor to decrease the length of the period of oscillations.  相似文献

Why and when the immune system skews to Th2 mediated allergic immune responses is still poorly characterized. With two homologous lipocalins, the major respiratory dog allergen Can f 1 and the human endogenous, non‐allergenic Lipocalin‐1, we investigated their impact on human monocyte‐derived dendritic cells (DC). The two lipocalins had differential effects on DC according to their allergenic potential. Compared to Lipocalin‐1, Can f 1 persistently induced lower levels of the Th1 skewing maturation marker expression, tryptophan breakdown and interleukin (IL)‐12 production in DC. As a consequence, T cells stimulated by DC treated with Can f 1 produced more of the Th2 signature cytokine IL‐13 and lower levels of the Th1 signature cytokine interferon‐γ than T cells stimulated by Lipocalin‐1 treated DC. These data were partially verified by a second pair of homologous lipocalins, the cat allergen Fel d 4 and its putative human homologue major urinary protein. Our data indicate that the crosstalk of DC with lipocalins alone has the potential to direct the type of immune response to these particular antigens. A global gene expression analysis further supported these results and indicated significant differences in intracellular trafficking, sorting and antigen presentation pathways when comparing Can f 1 and Lipocalin‐1 stimulated DC. With this study we contribute to a better understanding of the induction phase of a Th2 immune response.  相似文献

ABCA1 belongs to the A class of ABC transporter, which is absent in yeast. ABCA1 elicits lipid translocation at the plasma membrane through yet elusive processes. We successfully expressed the mouse Abca1 gene in Saccharomyces cerevisiae. The cloned ABCA1 distributed at the yeast plasma membrane in stable discrete domains that we name MCA (membrane cluster containing ABCA1) and that do not overlap with the previously identified punctate structures MCC (membrane cluster containing Can1p) and MCP (membrane cluster containing Pma1p). By comparison with a nonfunctional mutant, we demonstrated that ABCA1 elicits specific phenotypes in response to compounds known to interact with membrane lipids, such as papuamide B, amphotericin B and pimaricin. The sensitivity of these novel phenotypes to the genetic modification of the membrane lipid composition was studied by the introduction of the cho1 and lcb1-100 mutations involved respectively in phosphatidylserine or sphingolipid biosynthesis in yeast cells. The results, corroborated by the analysis of equivalent mammalian mutant cell lines, demonstrate that membrane composition, in particular its phosphatidylserine content, influences the function of the transporter. We thus have reconstituted in yeast the essential functions associated to the expression of ABCA1 in mammals and characterized new physiological phenotypes prone to genetic analysis. New compounds able to counteract staphylococcal biofilm formation are needed. In this study we investigate the mechanism of action of pyrrolomycins, whose potential as antimicrobial agents has been demonstrated. We performed a new efficient and easy method to use microwave organic synthesis suitable for obtaining pyrrolomycins in good yields and in suitable amount for their in vitro in-depth investigation. We evaluate the inhibitory activity towards Sortase A (SrtA), a transpeptidase responsible for covalent anchoring in Gram-positive peptidoglycan of many surface proteins involved in adhesion and in biofilm formation. All compounds show a good inhibitory activity toward SrtA, having IC50 values ranging from 130 to 300?µM comparable to berberine hydrochloride. Of note compound 1d shows a good affinity in docking experiment to SrtA and exhibits the highest capability to interfere with biofilm formation of S. aureus showing an IC50 of 3.4?nM. This compound is also effective in altering S. aureus murein hydrolase activity that is known to be responsible for degradation, turnover, and maturation of bacterial peptidoglycan and involved in the initial stages of S. aureus biofilm formation.  相似文献
