滇紫草愈伤组织培养与紫草素产生   总被引:9,自引:1,他引:8  
浓度为10~(-5)Smol/1和10~(-6)mol/l的2,4-D和NAA分别与10~(-5)mol/l的KT组合,能明显抑制滇紫草(Onosma paniculatum Bur. et Fr.)愈伤组织中紫草素的产生,但几乎不受天然生长素IAA和KT组合的影响。葡萄糖较蔗糖能更有效地促进紫草素的产生,它们的最适浓度均为6%。LH和CH能抑制紫草素的产生,CH浓度大于0.02%时能抑制愈伤组织的生长,LH对生长无明显影响。椰乳浓度为10%时,能明显地促进紫草素的产生,紫草素的含量是对照的24倍。  相似文献

为探讨UV-B辐射对鲜土贝母有效成分和抗氧化活性的影响,对土贝母(Bolbostemma paniculatum)新鲜块茎的有效成分含量和抗氧化活性进行了研究。结果表明,100 k J m–2的高剂量UV-B辐射使鲜土贝母的还原糖含量升高60.17%,大黄素含量升高209.60%。辐射后还原能力及清除DPPH·自由基能力均显著高于对照(P0.01)。这说明UV-B辐射可显著提高土贝母鲜品的有效成分含量和抗氧化活性,因此鲜土贝母加工前进行UV-B辐射可望提高药材质量。  相似文献

Trioza eugeniae oviposited significantly more eggs on Syzygium paniculatum leaves free of eggs compared to leaves with pre-existing eggs in a choice experiment, suggesting that females modify oviposition based on cues associated with the presence of eggs. In separate experiments, females avoided ovipositing on parts of leaves where eggs were present, but readily oviposited on areas of the same leaf without eggs. Epideictic pheromones are apparently not used by this species because females readily oviposited on areas of a leaf from which eggs had previously been removed. Females laid 57% of all their eggs along the margins of normal leaves, but also oviposited on artificial margins produced by cutting the leaves. The preference for natural margins over artificial margins was eliminated when the entire margin was coated with an antitranspirant compound to mask the damaged edge. These studies show that ovipositing psyllids respond both to the presence of eggs and to the suitability of the substrate.  相似文献

Experiments were conducted to determine potential cues used by nymphs when they select the preferred abaxial side of a leaf. Nymphs most frequently settle and develop on the abaxial side of leaves. The degree of preference for the abaxial side was found to be reduced when a high density of nymphs were present, however. The orientation of leaves in a completely darkened chamber did not influence settling preference for the abaxial side, even when leaves were oriented abaxial side up. Coating both sides of a leaf with an antitranspirant compound did not prevent nymphs from preferring the abaxial side for settling. Similar results were obtained when leaves were dipped in pentane. These studies show that nymphs do not use gravity, color, light, or contact chemoreception when selecting suitable settling sites on a leaf.  相似文献

以徐长卿子叶为材料,进行愈伤组织诱导、愈伤组织和不定芽分化、试管苗生根、移栽及移植等研究。结果表明,MS+6-BA 0.3 mg/L+NAA 0.2 mg/L+2,4-D 0.4 mg/L是子叶愈伤组织诱导和继代培养的理想培养基;MS+6-BA 0.8 mg/L+NAA 0.2 mg/L是愈伤组织分化培养的理想培养基;MS+6-BA 0.6 mg/L+NAA 0.2 mg/L是不定芽分化继代培养的理想培养基;炉灰渣是试管苗移栽的理想基质,移栽成活率可达97%。试管苗移植后长势旺盛,根系发达,当年开花。根据本试验结果可建立徐长卿子叶诱导再生体系。  相似文献

应用500~2500伦琴(R)的60钴γ射线照射滇紫草(OnosmapaniculatumBur.etFranch.)细胞系,确定了应用γ射线处理促进紫草素合成的最适辐照剂量为1500R,处理后的细胞系在生产培养基上培养21d后紫草素含量(以干重计)达到94.79mg/g,较对照提高了1446%,通过小团块选种法从中筛选到了色素含量(以干重计)高达103.42mg/g的高产细胞系Mut1,其营养生长与对照相比没有差别。  相似文献

Endogenously occurring nitric oxide (NO) is involved in theregulation of shikonin formation in Onosma paniculatum cells.NO generated after cells were inoculated into shikonin productionmedium reached the highest level after 2 d of culture, whichwas 16 times that at the beginning of the experiment, and maintaineda high level for 6 d. A nitric oxide synthase (NOS) inhibitor,N-nitro-L-arginine (L-NNA), and a nitrate reductase (NR) inhibitor,sodium azide (SoA), consistent with their inhibition of NO biosynthesis,decreased shikonin formation significantly. This reduction couldbe alleviated or even abolished by exogenous NO supplied bysodium nitroprusside (SNP), suggesting that the inhibition ofNO biosynthesis resulted in decreased shikonin formation. However,when endogenous NO biosynthesis was up-regulated by the elicitorfrom Rhizoctonia cerealis, shikonin production was enhancedfurther, showing a dependence on the elicitor-induced NO burst.Real-time PCR analysis showed that NO could significantly up-regulatethe expression of PAL, PGT and HMGR, which encode key enzymesinvolved in shikonin biosynthesis. These results demonstratedthat NO plays a critical role in shikonin formation in O. paniculatumcells.  相似文献

The Aspergillus oryzae elicitor was extracted from mycelia. The concentrated crude preparation of which was treated through DEAE-Cellulose, Sepharose-4B. Bio-Gel p- 4 and 732 columns. Elicitor activity was associated with fraction F Ⅰ b2-H, which had no affinity for DEAE-Cellulose and 732 resin. Its molecular weight was 1200~2200 D and its carbohydrate content was 6.7% of that of the crude. The elicitor activity was 120 times higher than that of crude preparation. There were also fractions F Ⅱ of nucleic acids and F Ⅰ b2-Na of nucleotides, amino acids in crude elicitor preparation. They did not affect shikonin derivative formation at low concentration, but inhibited shikonin derivative formation at high concentration. Fraction FIa of polysaccharid nature in the crude preparation which strongly inhibited shikonin derivative formation was another kind of elicitor of a new metabolite yellow pigment.  相似文献
