30 Aeromonas hydrophila water isolates were tested for bacteriocin-like substance (BLS) production using a target panel of closely related microorganisms and other Gram-positive and Gram-negative bacteria, including food-borne pathogens. A. hydrophila showed antibacterial activity against one or more indicator microorganisms, but the activity emerged only with non-phylogenetically related genera or species. In particular all A. hydrophila showed antibacterial activity against one or more of the tested Staphylococcus strains, five against Listeria spp. (Listeria seeligeri, Listeria welshimeri and Listeria ivanovii), and eight presented a weak antagonistic activity towards Streptococcus agalactiae and Lactobacillus spp. Inhibitory activity was not observed against the other Gram-positive (Listeria monocytogenes, Listeria innocua and Enterococcus spp.) and Gram-negative tested strains, including Aeromonas sobria, Aeromonas caviae and the same A. hydrophila, when used as indicator. Anti-staphylococcal activity was observed with a gradual increase of the inhibition zone during incubation and seemed to be influenced by A. hydrophila hemolytic expression. Extrachromosomal analysis showed the presence, in 70% of the strains, of one to five plasmids with molecular masses ranging from 2.1 to 41.5 MDa, but it was not possible to relate this result with BLS production.  相似文献

目的获取高产抑制鸡白痢沙门菌生长的类细菌素乳酸乳球菌的突变菌株。方法在紫外线照射致死率为82.7%的剂量下,对处于对数生长后期的乳酸乳球菌V528进行紫外线二次复合诱变处理。分别随机对第一次和第二次复合处理后生长的117株和310株菌进行效价测定分析。结果在实验所用的照射剂量下,抑菌效价提高的正突变菌株占有一定优势,负突变菌株较少,其中突变菌株Lact.174的抑菌效价较V528的效价提高了8.48倍,并具有一定的遗传稳定性。结论运用紫外诱变育种技术可有效地获得高产类细菌素的突变菌株。  相似文献

Bacillus licheniformis ZJU12, which was isolated from soil, could produce bacteriocin-like peptides that exhibited a broad spectrum of antagonistic activity against various species of Gram-positive bacteria and fungal pathogens, but not against Gram-negative bacteria tested except Xanthomonas oryzae pv. oryzae, a rice pathogen. The bacteriocin-like peptides were sensitive to proteinase K and trypsin. The activity was stable during temperature exposure up to 100 °C for 30 min, but lost completely at 121 °C for 15 min. The cell-free supernatant of B. licheniformis ZJU12 was shown to retain the activity within the pH range of 2–9, and the optimum pH for the activity was about 6.5. No adverse effect of the antagonistic compound to mice was observed in acute toxicity tests with the dose of 0.8 mg/20 g.  相似文献

动物乳杆菌的分离鉴定及其抑菌蛋白的特性分析   总被引:2,自引:0,他引:2  
通过滤纸片法从健康肉猪猪大肠、小肠中分离得到212株抗生物质产生菌, 以杯碟法复筛, 得到1株对溶壁微球菌(Micrococcus lysodeikticus)、金黄色葡萄球菌(Staphylococcus aureus)、枯草芽孢杆菌(Bacillus subtilis)、蜡状芽孢杆菌(Bacillus cereus)等革兰氏阳性菌和大肠杆菌(Escherichia coli)、鼠伤寒沙门氏菌(Salmonella typhimurium)等革兰氏阴性菌以及部分真菌如禾谷镰刀霉(Fusarium graminearum)均有强烈抑制作用的乳酸菌。经形态学、生理生化特征及16S rDNA序列同源性分析等手段鉴定该菌株为动物乳杆菌(Lactobacillus animalis)。排除酸和过氧化氢的干扰后, 该菌株的发酵上清液对指示菌仍有明显抑菌活性; 用蛋白酶处理该菌株的发酵上清液后, 抑菌活性丧失; 发酵液粗提物具有较好的热稳定性(经121°C处理20 min仍有较强抑菌活性)以及较宽的抑菌活性pH值范围(3.5~5.5), 因此初步认为该菌株产生一类具有广谱抑菌活性的类细菌素物质。  相似文献

Twenty-five Campylobacter isolates were screened for production of antimicrobial substances using a deferred antagonism assay. Sixteen isolates showed activity against either Staphylococcus aureus, Salmonella enterica serovar Enteritidis or Candida albicans. The inhibitory activity was sensitive to treatment with pronase E, trypsin and pepsin, suggesting that the antimicrobial compound(s) are proteinaceous. Activity spectra of isolates included S. aureus, Micrococcus luteus, Streptococcus sp., Bacillus subtilis, a drug-resistant clinical isolate of S. aureus and one isolate of C. albicans. Producing isolates showed cross-immunity and inhibitory activity was only observed on solid media. The findings of this study suggest that Campylobacter produces proteinaceous inhibitory substances.  相似文献

本研究通过正交试验获得嗜酸乳杆菌sR-1分泌类细菌素的最适培养条件,即葡萄搪2%,大豆蛋白胨2.5%,酵母粉0.4%,血浆蛋白2.5%。并首次利用流加培养的方式,改进了类细菌素产生菌的发酵条件,抑菌直径从19mm提高到25mm,同时对类细菌素生物学特性进行了初步的探索。结果表明:嗜酸乳杆菌产生的类细菌素对多种蛋白酶不敏感,在低pH下能抑制革兰阴性、阳性的致病菌,在pH6.5下吸附菌体作用最强。  相似文献

从分离自内蒙古传统乳制品的67株乳酸菌中筛选得到一株产生类细菌素的布氏乳杆菌KLDS1.0364,对其所产类细菌素进行初步分离纯化,同时研究其所产类细菌素的生物学特性.KLDS1.0364无细胞发酵上清液经阳离子交换树脂纯化后,采用tricine-SDS-PAGE测定类细菌素分子量,并测定了类细菌素的部分特性.KLDS1.0364产生的类细菌素分子量约为21.6kD,对热和pH值稳定,可被多种蛋白酶失活,不能被过氧化氢酶和α-淀粉酶失活.KLDS1.0364产生的类细菌素的作用方式是杀菌,且抑菌谱广,可抑制多种革兰氏阳性菌、革兰氏阴性菌和真菌.  相似文献

目的对产类细菌素嗜酸乳杆菌P302的发酵条件进行研究。方法采用琼脂扩散法测定发酵上清液对大肠埃希菌O139的抑菌活性。结果通过正交试验确定P302产类细菌素的最佳培养基组分为牛肉膏4%、胰蛋白胨0.2%、酵母粉0.8%、碳酸钙0.4%和蔗糖1%,0.4%复合维生素,0.3%Tween-80有利于类细菌素的产生。发酵工艺条件为最适初始pH7.0,最适温度37℃,最佳接种量0.3%,种龄14h,厌氧培养22h。结论通过优化发酵条件和发酵工艺,类细菌素的产量提高了35%。  相似文献
