Eighteen strains of xylariaceous fungi have been screened for higher activities of cellulolytic enzymes,Trichoderma reesei QM 9414 was also examined for comparison. Strains ofXylaria anisopleura andX. regalis had higher endocellulase (CMCase) and exocellulase (Avicelase) activities after 2 weeks' incubation.Hypoxylon stygium produced the highest activity of -glucosidase 3 days after inoculation. The optimum pH for these cellulolytic enzymes was approx. 5.0 and the optimum temperatures ranged from 37 to 50°C. A mixed culture process usingT. reesei QM 9414 andH. stygium was developed to obtain enhanced synthesis of cellulase. -Glucosidase activities in the mixed culture increased within 48h whenH. stygium was introduced after 24h. 相似文献

The non-catalytic region of the Clostridium stercorarium cellulase CelZ (Avicelase I) comprises two protein segments (C and C′) grouped into different subfamilies of cellulose-binding domain (CBD) family III. The C-terminally located family IIIb domain C was identified as a true cellulose-binding domain responsible for anchoring the CelZ enzyme to cellulose. The family IIIc domain C′ immediately adjacent to the catalytic domain was unable to mediate binding to cellulose. A deletion study revealed a lack of independence of this pair of domains: almost the entire C′ domain was required to maintain the catalytic activity and the thermostability of the enzyme. 相似文献

The formation of cellulases and -glucosidase and their location in the fungus Phialophora malorum was studied on some different carbon sources. The cellulases were found to be partly cell-free and partly cell-bound during growth on cellulose and carboxymethyl-cellulose. Glucose and cellobiose repressed the cellulase formation but a low carboxymethylcellulase activity was measurable on the glucose-grown mycelium. The unicellular stage did not appear to grow on carboxymethyl-cellulose or cellulose, but mycelium was formed on these carbon sources. 相似文献

A Streptomyces sp. was isolated that produced novel thermoalkalotolerant cellulase activity after growth on crystalline cellulose at 50°C.
Three major components of the cellulases (CMCase, Avicelase and cellobiase) were produced with maximal activities (11.8, 7.8
and 3.9 IU/ml) and maximum specific activities 357, 276 and 118 IU/mg protein, respectively, after 120 h growth. Maximum CMCase
activity was between 50 and 60°C measured over 3 h. The enzyme also retained 88% of its maximum activity at 70°C and pH 5,
and 80% of the activity at pH 10 and 50°C when assayed after 1 h. After incubation at 40°C for 1 h with commercial detergent
(Tide) at pH 11, 95% activity was retained. The enzyme mixture produced glucose from crystalline cellulose. 相似文献

The bacterium Streptomyces reticuli produces an unusual mycelia-associated cellulase (Avicelase, Cell) which is solely sufficient to degrade crystalline cellulose to cellobiose. The enzyme consists of a binding domain, one adjoining region with unknown function, and a catalytic domain belonging to the cellulase family E. During cultivation, the strain produces a specific protease which processes the Avicelase to a truncated enzyme lacking the binding domain. The cellulase synthesis is regulated by induction (Avicel) and repression (metabolizable sugars and glycerol). 相似文献

Termitomyces clypeatus secreted a 24-kDa xylanase constitutively in xylan medium, but required a gluconeogenic amino acid or Krebs cycle acid for the secretion of a 56-kDa amyloglucosidase in dextrin medium. Aspartate, glutamate, succinate and fumarate all increased secretion of amyloglucosidase from 50% to >90% and enzyme production by 10-fold with little effect on xylanase production. Glutamate or succinate stimulated in vitro release of intracellular amyloglucosidase from washed mycelia in the presence of cycloheximide. Amyloglucosidase accumulated in the absence of glutamate was a high-molecular-mass protein that did not migrate in PAGE. Cellular regulation by the fungus of the secretion of amyloglucosidase is indicated. 相似文献

Seven anaerobic fungal isolates from Cervus dama (domesticated and free living) were grown on carboxymethyl cellulose (CMC) and avicel, and monitored over a five day period
for substrate utilization and cellulase activities. All fungal isolates showed monocentric growth patterns; four of them had
polyflagellated zoospores and morphologically resembled members of the genus Neocallimastix; the other three had monoflagellated zoospores and resembled members of the genus Piromyces.
