Abstract A β-glucosidase from centrifugated autolyzed cultures of Alternaria alternata has been purified 71 times by Sephadex G-200, CM-Biogel A and DEAE-Biogel A successively. The enzyme is a glycoprotein with 16% sugar and a M r of 160 000, formed by two subunits of 60 000 and 80 000. The enzyme has optimum pH of 5 units and optimum reaction temperature of 50°C, being stable in a pH range of 3–8 and 0 to 60°C. The enzyme hydrolyzes different substrates showing maximum affinity and maximum hydrolysis velocity on cellobiose. The β-glucosidase is inhibited by gluconolactone but not by 10 mM glucose.  相似文献

Indoleacetic acid (IAA), a factor that induces growth in epicotyls of cicer arietinum L. cv. Castellana, increases the autolytic capacity of the cell walls by 50%, suggesting that autolysis is related to the processes of cell wall loosening that accompany growth. IAA promotes an increase in the specific activities of the enzymes involved in autolysis, mainly α-galactosidase (EC 3.2.1.22). This relationship autolysis-growth. was also observed in a study of the autolytic capacity of cell walls from regions of the epicotyl with different growth capacity. The sugars released and the level of enzymatic protein were higher in the subapical region that towards the base.  相似文献

Simulation of drought by polyethylene glycol (PEG) inhibited elongation of epicotyls of Cicer arietinum L. cv. Castellana but had no effect on growth capacity since growth was restored once the inhibitory condition had been removed. The amount of proteins in the cell wall was correlated with the elongation of the epicotyls and decreased when elongation was inhibited. PEG-induced inhibition of elongation had different effects on the various glycanhydrolytic cell wall enzymes. Only α-galactosidase (EC 3. 2. 1. 22) seemed related to the lack of elongation, increasing its activity when elongation was inhibited. The β-galactosidase (EC 3. 2. 1. 23) and β-glucosidase (EC 3. 2. 1. 21) studied did not show changes in their specific activities during the inhibition of elongation. β-Galactosidase is responsible for the autolytic process in Cicer arietinum . This enzyme hydrolyzes specified linkages in the cell wall, releasing sugar constituents. Our present results show that β-galactosidase is not directly related with elongation because no changes could be observed during inhibition of elongation. The autolytic process is related with chemical processes taking place in the cell wall and preceding elongation of the epicotyls, i. e. the loosening process. Cell wall loosening is necessary for elongation to take place but elongation does not necessarily follow loosening if the osmotic conditions are unfavorable  相似文献

Abstract Production of protoplasts in different genera of filamentous fungi with their own lytic enzymes obtained from autolyzed cultures, as well as the regeneration of these protoplasts, has been studied. The results support the idea that the use of these autolytic enzymes could be a general method of production of protoplasts from filamentous fungi.  相似文献

Oryza sativa L. var. bahia coleoptile cell walls show sufficient autolytic activity for the release into the surrounding medium of amounts up to 60 μg of sugars per mg of dry weight of cell wall. The products released elute in Bio-gel P.2 as mono- and polysaccharides with glucose as the sole component. The polysaccharide component releases tri- and tetrasaccharides on treatment with a glucanase specific for β (1–3) (1–4) linkages in the same proportion as that of the mixed glucan of the cell wall. This supports the hypothesis that the polysaccharide component originates from the cell wall glucan and that autolysis is therefore related to the processes of the loss of rigidity of the cell wall. Nojirimycin (a specific glucanase inhibitor and inhibitor of auxin-induced elongation) decreases autolytic activity of the cell walls, reducing it to 30% of its normal value. Bio-gel P. 2 elution of the products released in autolysis in the presence of nojirimycin shows that only the monosaccharide fraction was affected.  相似文献

The potential of several alternative genetic engineering based strategies in order to accelerate Saccharomyces cerevisiae autolysis for wine production has been studied. Both constitutively autophagic and defective in autophagy strains have been studied. Although both alternatives lead to impaired survival under starvation conditions, only constitutively autophagic strains, carrying a multicopy plasmid with the csc1-1 allele under the control of the TDH3 promoter, undergo accelerated autolysis in the experimental conditions tested. Fermentation performance is impaired in the autolytic strains, but industrial strains carrying the above-mentioned construction are still able to complete second fermentation of a model base wine. We suggest the construction of industrial yeasts showing a constitutive autophagic phenotype as a way to obtain second fermentation yeast strains undergoing accelerated autolysis.  相似文献

Summary An electron microscopical study has been made of the notochordal cells of the tail of the larval Rana temporaria, before and after they commence to degenerate at metamorphic climax.Degeneration of the tail begins at the tip; the necrotic area proceeds to extend proximally and becomes more widespread. Simultaneously during climax the tail shortens and finally disappears as the animal acquires the froglet form.Notochordal cells autolyse independently and at random, when there is disruption and disorganisation of the surrounding notochordal sheath. The release of lysosomal enzymes intracellularly elicits organelle necrosis.Evidence is provided to support the view that necrotic notochordal tissue is phagocytosed by invading mesenchymal macrophages.  相似文献

Glycosylation is often cited as having a stabilizing effect upon proteins with respect to proteolysis, thermolysis and other forms of degradation. We present here a model study on an autolytic protease that has been chemically glycosylated to produce single glycoforms. The resulting glycosylated enzymes are more stable with respect to their own autolytic degradation and that by other proteases. Kinetic parameters for protease activity with respect to the degradation of small-molecule amide substrate reveal no significant change in inherent activity thereby suggesting that reduced autolysis and proteolysis are a consequence of stabilization, perhaps by steric blockade of cleavage points or alteration of local unfolding kinetics. Variation in glycan identity suggests that greater glycan size leads to greater stabilization.  相似文献
