TTUR 2-2, an alkalophilic Bacillus strain isolated from soil, grew well in media containing cholic acid (CA) at 5% or higher and efficiently converted 7α- and 12α-hydroxyl groups of CA to keto groups, with the conversion rate for both hydroxyl groups reaching 100% by 72 hours of cultivation. The strain also converted a 3α-hydroxyl group to a keto group, but the conversion rate was about 5% at 72 hours. The strain neither affected any other part of the CA molecule, nor oxidized 7β- or 12 β -hydroxyl groups.

By NTG mutagenesis, the following mutants were acquired; (1) converting only the 7α- and 12α-hydroxyl groups, (2) converting only the 12α-hydroxyl group, and (3) converting only the 7α-hydroxyl group. These mutants selectively produce 12-ketochenodeoxycholic acid (12KCDCA), 7-ketodeoxycholic acid (7KDOCA), and 7,12-diketolithocholic acid (7,12DKLCA), from CA; and 7-ketolithocholic acid (7KLCA) from cheno-deoxycholic acid (CDCA), respectively, at high yields, close to 100%.  相似文献

本文用正交实验设计法探讨了杂色曲霉(Aspergillus versicolr)在加有两类不同性质营养物的人胃液中产生杂色曲霉素(Sterigmatocystin,简称ST)的条件。发现在26℃斜面静置培养12天后可产生ST。加入半合成物质的最佳配伍是:蔗糖1,000.0mg;蛋白胨50.0mg;KH_2PO_4 7.5mg;MgSO_4·7H_2O_2.5mg;人胃液10.0ml,称之为SPKM人胃液培养基。加入天然物质的最佳配伍是:玉米粉0.5g;豆腐粉0.25g,人胃液10.0ml,称之为CS人胃液培养基。还进一步研究了pH值和培养时间对杂色曲霉产毒菌株在SPKM和CS人胃液培养基中生长及产生ST的影响。根据临床胃酸缺乏程度分级标准,分为pH 1.0,3.0,6.5,8.0四个组。发现在两种人胃液培养基中,无论是杂色曲霉生长,还是产生ST,pH3.0到6.5是发生质变的范围。在两种人胃液培养基pH为6.5时,37℃静置培养8天有痕量ST产生,10天后就明显增加。所以杂色曲霉产生的ST可能是慢性萎缩性胃炎易癌变的原因之一。  相似文献

Callus was induced from juice vesicles of satsuma mandarin on Murashige & Skoog medium supplemented with -naphthaleneacetic acid (NAA), kinetin (K) and gibberellin (GA). Adventitious embryoids arose from the callus tissue on the medium containing 1 mgl–1 NAA alone. The process of digestion of captured feeds in a pitcher, an insect-trapping organ, ofNepenthes was studied. Changes in bacterial population, pH and NH4 + concentrations in pitcher juice were examined. Strong activities of both acid- and alkaline phosphatase, phosphoamidase, esterase C4 and esterase C8 were found in the pitcher juice. Optimum pH of proteases in the juice and those secreted from bacteria showed pH 3.0 and pH 8.0–9.0, respectively. Twenty six strains of bacteria were isolated from 4 pitchers: 10 strains were gram positive, 16 strains were gram negative (10 strains had casein hydrolase activity). A proton excretion was induced by NH4 + released from the added solutions, and accordingly, the pH of the solutions fell. As a simulation model of the digestion process of feeds in pitcher juice and polypeptone solution was added into the washed pitcher. A good correlation was found among the NH4 + concentration, pH and bacterial cell titer.  相似文献

采用正交试验和中心组合设计相结合的数理统计方法,在2L发酵罐中对紫云英汁液培养单细胞蛋白发酵培养基进行筛选、优化,试验结果表明较优培养基为:初糖2.3%,酵母粉0.16%,KHPO0.2%,MgSO0.05%。酵母浓度最高达10.46g/L,基质生长得率为0.55g菌体/g基质,基质转化率为83%。研究结果同时说明采用数理统计方法设计实验,工作量小,效率高,结果准确。  相似文献

Maintenance of high cell viability was the main characteristic of our new strains of thermotolerant Saccharomyces. Total sugar conversion to ethanol was observed for sugarcane juice fermentation at 38-40 degrees C in less than 10 h and without continuous aeration of the culture. Invertase activity differed among the selected strains and increased during fermentation but was not dependent on cell viability. Invertase activity of the cells and optimum temperature for growth, as well as velocity of ethanol formation, were dependent on medium composition and the type of strain used. At high sugarcane syrup concentrations, the best temperature for ethanol formation by strain 781 was 35 degrees C. Distinct differences among the velocities of ethanol production using selected strains were also observed in sugarcane syrup at 35-38 degrees C.  相似文献

Occluded virions of the Bombyx mori nuclear polyhedrosis virus were efficiently liberated from polyhedra by dissolution with the silkworm gut juice. The liberated virions were purified by sucrose density gradient centrifugation and the bands of enveloped virions were observed in the gradients. There was no functional difference between the gut juice-liberated and the carbonate-liberated virions. Disruption of enveloped virions by the gut juice was observed, but the formation of nucleocapsids from the degradation of the occluded virions was not detected. High yields of the enveloped virions from the polyhedra dissolved by the gut juice was obtained by separating the virions through sucrose density gradient centrifugation immediately after the dissolution of the polyhedra. Many factors, e.g., rearing seasons, silkworm strains, and rearing conditions, affect the polyhedra-dissolving property of the larval gut juice.  相似文献

The general pathology induced by δ-endotoxin in terms of larval behavior and hemolymph chemistry has been widely studied in the so-called Type I insect, Bombyx mori. The succession of symptoms is divided into four arbitrary stages: Stage 0, appearance and locomotion normal, no feeding; Stage 1, slightly sluggish; Stage 2, extremely sluggish; and Stage 3, complete paralysis. The action of δ-endotoxin is highly specific to the midgut since contractile movement of both foregut and hindgut continues long after all locomotor activity and heartbeat have stopped. Immediately after the silkworm stops feeding and blood pH sharply rises, there is an associated abrupt rise in the K+ concentration of hemolymph. Thereafter, the rise in K+ is linear while the rise in pH is not. In vivo measurements have not yielded the same simple linear dependence of pH on K+ concentrations that is found in in vitro mixtures of hemolymph and midgut juice. Ligation experiments showed that the same pathological sequence (rise of pH and K+ concentration, and general paralysis) follows whether the toxin has unrestricted access to the entire midgut or only part of it (anterior or posterior). From the results of injections of midgut juice or various salt solutions into hemocoel, we came to the conclusions that the blood pH and the symptoms are not necessarily parallel and the intact midgut and Malpighian tubules have strong functions for ion regulation.  相似文献
