We developed two tests for sex identification of felids using y‐chromosome deletions in the zinc‐finger and amelogenin regions. These tests provide positive results for both males and females, while reducing the need to co‐amplify microsatellites to test for DNA quality in hair and scat samples. Furthermore, the y‐chromosome deletions are absent in a wide‐range of prey species; thus, when these tests are used on scat samples, potential contamination caused by prey DNA incidentally extracted, is minimized.  相似文献

Exposure of spinach (Spinacia oleracea L. cv. Monosa) to 0.25 μl l?1 H2S reduced the relative growth rate by 26, 47 and 60% at 15, 18 and 25°C, respectively. Shoot to root ratio decreased in plants fumigated at 18 and 25°C. Growth of spinach was not affected by a 2-week exposure to 0.10 or 0.25 μl l?1 SO2. Both H2S and SO2 fumigation increased the content of sulfhydryl compounds and sulfate. A 2-week exposure to 0.25 μl l?1 H2S resulted in an increase in sulfhydryl and sulfate content of 250 to 450% and 63 to 248% in the shoots, respectively, depending on growth temperature. Exposure to 0.15 and 0.30 μl l?1 H2S at 20°C for 2 weeks resulted in a 46% increase in sulfate content of the shoots at 0.30 μl l?1 and no detectable increase at 0.15 μl l?1 H2S; the sulfate content of the roots increased by 195 and 145% at 0.15 and 0.30 μl l?1 H2S, respectively. Fumigation with 0.25 μl l?1 SO2 at 20°C for 2 weeks resulted in an increase in sulfhydryl content and sulfate content in the shoots of 285% and 300 to 1100%. H2S fumigation during the 12 h light period or only during the dark period resulted in identical growth reduction and accumulation of sulfhydryl compounds; they were about 50 and 67% of those observed in continuously exposed plants. H2S- and SO2-exposed plants showed an increased transpiration rate, which was mainly caused by an increased dark-period transpiration. No effect of H2S and SO2 on the water uptake of the plants and the osmotic potential of the leaves was detected. Plants fumigated with 0.25 μl l?1 H2S for 2 weeks were smaller and differed morphologically from the control plants by slightly more abaxially curved leaf margins. Cross sections of the leaves showed smaller cells at the margins and smaller and fewer air spaces. The increased transpiration in the H2S-exposed plants is discussed in relation to the observed morphological changes.  相似文献

1.  Filiform hairs of various lengths on the cerci of adult crickets vibrate in a sound field. These movements were measured with a photodetector for sound frequencies from 10 Hz to 200 Hz in the species Acheta domestica, Gryllus bimaculatus and Phaeophilacris spectrum.
2.  With low air-particle velocities, the hair shafts were deflected sinusoidally from their resting position, without bending or secondary oscillations (Figs. 2 A, 3 A). At higher velocities (from ca. 80 mm/s peak velocity, depending on the properties of the individual hairs), the shaft struck the cuticular rim of the socket in which the base of the hair is seated (Fig. 2B). This contact was made at an average angular displacement from the resting position of 5.16°±1.0°.
3.  The best frequencies of the hairs were found to be between 40 Hz and 100 Hz (Fig. 5A). The slope of the amplitude curve for constant peak air-particle velocity at frequencies below the best frequencies was between 0 and 6 dB/octave. Long hairs had smaller slope values than short hairs (Fig. 5C).
4.  At its best frequency the ratio of maximal tip displacement of a hair to the displacement of the air particles in the sound field was between 0.2 and 2. Only a small number of hairs (2 out of 36) showed tip displacements exceeding twice the air-particle displacement. The values of maximal angular displacement were not correlated to hair length (Fig. 5 B).
5.  The angular displacement of the hairs was phase shifted with respect to the air-particle velocity by 0° to +45° (phase lead) at sound frequencies around 10 Hz and by -45° to -120° (phase lag) at 200 Hz (Figs. 3C, 4B). At a particular frequency long hairs tended to have larger phase lags than shorter hairs (Fig. 5D).

Summary Mechanical oscillation properties of cricket (Acheta domesticus) filiform hair sensilla were measured at different larval stages, as an indication of larval sensory capacities and for comparison with data in the literature on central nervous changes during development. The hairs were stimulated by airborne vibration over a frequency range of 10 to 220 Hz. Best frequency, angular displacement at best frequency, slope of angular-displacement tuning curve and phase of hair deflection relative to air particle velocity were tested for correlation with hair length, which is proportional to the age of a sensillum. The ranges found for the various oscillation parameters in early larval stages were similar to or larger than those in adults. Oscillation properties changed with both the developmental stage of the hair sensilla and that of the whole animal. Four individually identifiable hair sensilla were analysed separately; the sensory neurons of two of them are known to change synaptic properties during maturation. Angular displacement at a given stimulus intensity was maximal for all hairs after differentiation, and decreased during further development. The hairs did not show clear common changes for any of the other oscillation parameters. Yet particular changes were found for individual hairs.  相似文献

Summary At the end of a 4-year period of gas exchange measurements in a natural stand in the Lower Bavarian Forest, needles of an adult spruce [Picea abies (L.) Karst.] were harvested from two chambers, one with pure air and the other with ambient air. The needles were examined as to their histological properties in the stomatal apparatus and in the bundle sheath. In needles from the polluted air UV absorbance at 280 nm was decreased in the walls of the stomatal apparatus. Simultaneously, the deposition of compounds with an absorption maximum at 310 nm increased within the encrusted plate-like thickenings of the subsidiary cells. The contents of the lumina of hypodermal cells and of the bundle sheath exhibited a greater degree of autofluorescence in ambient-air material than in pure-air leaf organs. Differences between needles exposed to pure and polluted air are gradual. The damaged condition is rare in pure air, common in polluted air. The needles from outside the chambers occupied an intermediate position between pure-air and ambient-air needles. This fact is traced to an unnaturally high pollutant load in the liquid phase of the needle surfaces within the ambient-air chamber because in order to compensate pollutant losses within the system, SO2 and O3 were added even during periods of irrigation. The reduction of absorption capacity at 280 nm in the walls of the stomatal apparatus is attributed to destruction of lignin due to the high reactivity of the pollutants in the liquid phase on the damp needle surface. The importance of delignification with regard to hydroregulation is discussed.  相似文献

A beta-ray gauge system was used to study evaporation rates and drying times of liquids exposed to relatively high fluxes of air ions of either polarity produced by a corona discharge. Evaporation rates from thin layers of water, ethyl alcohol, and carbon tetrachloride increased considerably when exposed to air ions, compared to untreated liquids under constant environmental conditions. Drying times of liquid layers exposed to air ions were shortened by a factor of at least two over liquids that had not been exposed to ions. The drying time of a liquid layer was found to increase with the decrease of ion fluxes at the liquid surface. Threshold values of 1.9×1011 positive ions and 2.7×1011 negative ions cm–2 s–1 were established below which no increase in the evaporation rates were observed for ethyl alcohol and carbon tetrachloride. The evaporation rate of water at these same ion fluxes was significantly higher than that of the control. The present experiments indicate that a stream of air ions could play a major role in the observed enhancement of evaporation rates.  相似文献

In The Netherlands the decline of some phanerogamic species cannot be readily explained from obvious factors such as lowering of the groundwater table, eutrophication or land reclamation. For a number of species the hypothesis was tested that the decline is partly due to air pollution. A two-factor model was made in which decline is accounted for by (a) habitat destruction assessed from topographic maps and (b) air pollution measured as the SO2 95-percentile over the winter period 1978/1979. Effects of both factors were assumed to follow a sigmoid dose-effect curve. For a number of species decline proved to be significantly correlated with air pollution. These are notably species from the syntaxon Violion caninae. A comparison was made with results obtained for epiphytic lichens. Summary As a result of air pollution, considerable deposition of ammonium sulphate occurs on vegetation and soil in the vicinity of chicken farms and fields dressed with animal slurry. A clear relation exists between this ammonium sulphate deposition and the distance to certain agricultural activities. Field investigations and ecophysiological experiments both show that the needles ofPinus nigra var.maritima (Ait.) Melville take up ammonium and excrete potassium, magnesium and calcium. This often results in potassium and/or magnesium deficiencies and may lead to premature shedding of needles. The high levels of nitrogen in the needles are strongly correlated to fungal diseases.Whether the observed cation leaching will result in disturbed nutrient budgets depends mainly on soil conditions. Leaching of K, Mg and Ca from the soil, caused by ammonium sulphate, may further inhibit nutrient uptake.Field investigations show a clear correlation between increased ratios of NH4 to K, Mg and Ca in the soil solution and the damage to pine forests.  相似文献
