Morphological variation for the NOR chromosome was studied for four half-siblings of a sexual outbreedingTaraxacum, for three siblings of the obligate agamospermT. pseudohamatum, and for two individuals of the agamospermT. brachyglossum. No rearrangement was detected for the 113 chromosomes of sexuals, or for 41 chromosomes of two agamospermous individuals. In the other three agamospermous individuals, 3/16, 5/50, and 5/20 chromosomes showed evidence of chromosomal rearrangement. The majority of rearrangement events (10/13) occurred to the satellite rather than to the body of the NOR-chromosome. It is considered that such high levels of somatic chromosomal rearrangement in agamospermousTaraxacum may be the result of activity by transposable genetic elements. This recombination may be of selective advantage to asexual plants which cannot generate genetic variability through the sexual process.  相似文献

RICHARDS, A. J., 1990. Studies in Garcinia , dioecious tropical forest trees: the phenology, pollination biology and fertilization of G. hombroniana Pierre . Garcinia hombroniana is a facultative agamosperm which is pollinated by Trigona bees. Nectar is restricted to the large discoid stigma (or pistillode in male flowers), which also captures and hydrates pollen. The 'wet' stigma and binucleate pollen suggest that Garcinia arose from hermaphrodite plants with a gametophytic self-incompatibility system.
On stigmas, nectar is secreted early on three or four successive days. On male pistillodes, nectar is secreted when anthers dehisce, on the second morning after anthesis. Pollen is most viable when freshly collected, but some viability remains four days after collection. Pollen germinates within 24 h of hydration. Similar results to pollinations are obtained by germinating pollen in 1 % sucrose.
Garcinia hombroniana flowers principally from January to June. Cultivated females are considered as 'big bang' strategists. Male flowers are considered as 'steady state' strategists.  相似文献

Two molecular markers, a RAPD (randomly amplified polymorphic DNA) and a RFLP/STS (restriction fragment length polymorphism/sequence-tagged site), previously were found associated with apomictic reproductive behavior in a backcross population produced to transfer apomixis from Pennisetum squamulatum to pearl millet. The occurrence of these molecular markers in a range of 29 accessions of Pennisetum comprising 11 apomictic and 8 sexual species was investigated. Both markers were specific for apomictic species in Pennisetum. The RFLP/STS marker, UGT 197, was found to be associated with all taxa that displayed apomictic reproductive behavior except those in section Brevivalvula. Neither UGT197 nor the cloned RAPD fragment OPC-04600 hybridized with any sexually reproducing representatives of the genus. The cloned C04600 was associated with 3 of the 11 apomictic species, P. ciliare, P. massaicum, and P. squamulatum. UGT197 was more consistently associated with apomictic reproductive behavior than OPC04600 or cloned C04600, thus it could be inferred that UGT197 is more closely linked to the gene(s) for apomixis than the cloned C04600. The successful use of these probes to survey other Pennisetum species indicates that apomixis is a trait that can be followed across species by using molecular means. This technique of surveying species within a genus will be useful in determining the relative importance of newly isolated markers and may facilitate the identification of the apomixis gene(s).  相似文献

This study investigated the occurrence of the rust fungus Puccinia chondrillina in clonal populations of its host plant, Chondrilla juncea. In 16 populations spread across eastern Turkey, 48 different multilocus isozyme phenotypes were identified in the host. Of these clones, 88% were restricted to single localities, while the remaining 12% were found in 2–11 populations. For 13 of the 16 plant populations the commonest host clone was always infected. Indeed, at ten sites this clone was the only one found to carry disease. In the remaining three populations the rusted plants were all of the second commonest isozyme type. The possibility of such a tight association of rust incidence with host clone frequency simultaneously across a wide geographic area is very low (P0.023), supporting the contention that the pathogen P. chondrillina may be imposing negative frequency-dependent selection on these C. juncea populations.  相似文献

Apomixis is defined as asexual reproduction through seeds, although this outcome can be achieved by multiple pathways. Since little is known about the molecular control of these pathways, how they might intersect is also a mystery. Two of these pathways in the grass family, diplospory and apospory, are receiving attention from molecular biologists. Apospory in Pennisetum/Cenchrus, two genera of panicoid grasses, results in the formation of four-nucleate embryo sacs that lack antipodals. Sexual reproduction frequently aborts so that the resulting seed is composed of (1) a parthenogenetically derived embryo that is genetically identical to the mother and (2) endosperm formed through pseudogamy. The transmission of apomixis is associated with the transfer of a linkage block on a single chromosome. This linkage block contains repetitive sequences as well as hemizygous, low-copy DNA sequences. Fluorescence in situ hybridization has demonstrated that these DNA regions occur on only a single chromosome, but not its homologs, in the polyploid apomicts studied. Features of the apomixis-associated region resemble those of other chromosomal segments isolated from recombination and replete with "selfish" DNAs.  相似文献

Dandelions (Taraxacum officinale Weber(sensu lato); Asteraceae) have been introduced to NorthandSouth America with human migration from Europe. While potential sourcepopulations have both sexually and obligate asexually (agamospermous)reproducing lineages, apparently only the latter have successfully colonizedtheAmericas. The consequences of obligate agamospermy on dandelion populationgenetic diversity in North America remain little explored. Here we use fourdifferent synthetic DNA probes that reveal genetic markers at multiplevariable-number-tandem-repeat (VNTR) loci to examine patterns of geneticvariation among plants collected along three different central North Americantransects with plants (21 to 22 individuals per transect) separated by: 1) >2 m and < 60 m (short transect); 2) > 5km and < 30 km (medium transect); and 3) > 30km and < 340 km (long transect). The mean numberofVNTR markers revealed per plant was 59.3. Co-clonal individuals (proportion ofbands shared exceeding 90%) were found in each transect, with the indexof clonality (the percent of co-clonal individuals detected in a transect)ranging from 34.12% for the short transect to 18.65% for the longtransect. Co-clonal individuals were separated by up to 200 km.With redundant examples of co-clonal individuals removed, mean similarity(proportion of band sharing) of distinct genotypes within transects was 0.426,and no statistical differences in level of similarity between transects, norindication of genetic differentiation between transects, was detected (meanFst between transect levels with all individuals included =0.05). These results indicate: 1) that dandelion genetic diversity ofcolonizinglineages in central North America is moderately high and does not reflectextreme bottleneck effects shown by some colonizing species; and 2) thatdandelion seed dispersal can be very effective in maintaining similar levels ofgenetic diversity at the different scales of sampling in this study, withcertain clones maintaining numerous, widespread individuals. Evidence that VNTRmutation is detectable within dandelion clonal lineages is presented,demonstrating that clonal families with lines increasinglydifferentiated from one another will continually evolve, and that Muller'srachet is, in all likelihood, turning for asexual lines.  相似文献

Natural hybridization in Taraxacum between native sexual diploids and introduced agamospermous triploids occurring in Japan was studied by means of chloroplast DNA (cpDNA) marker. We first determined the nucleotide sequences between trnT (UGU) and trnF (GAA) of cpDNA for 22 plants obtained from Japan and Europe. The sequences analyzed were about 1,574 base pairs long. Among all accessions, the total numbers of polymorphic characters were 56 nucleotide substitutions, three insertions/deletions (ins/dels), and one repeat number polymorphism of mononucleotide motif. Of these polymorphic characters, four nucleotides and one ins/del were applicable in the discrimination between Japanese and European taxa of dandelions. We selected the ins/del in an intergenic region between trnL (UAA) 3′ exon and trnF (GAA) as a cpDNA marker. Using a newly developed cpDNA marker, 225 plants of putative Taraxacum officinale collected from 11 populations in Niigata City were investigated. Eighty-two percent of them showed a Japanese haplotype of cpDNA, and they were regarded as hybrids. Compared with the previous studies, it is likely that the prevalence of the hybrid plants is a general phenomena at least in urban areas in Japan. Summary Apomixis, asexual reproduction through seed, is an obligate mode of reproduction in several species from the genus Pennisetum. Transfer of apomixis to sexual, cultivated pearl millet (P. glaucum) from a wild species P. squamulatum has resulted in an obligate apomictic backcross line with a low, but unknown number, of chromosomes from the wild species. Molecular markers (restriction fragment length polymorphisms and random amplified polymorphic DNAs) have been identified that unequivocally demonstrate the presence of P. squamulatum DNA in BC3. Three of the informative RFLP clones have been sequenced and converted to sequence-tagged sites that can be amplified by the polymerase chain reaction. Molecular genetic analysis of more advanced back-cross individuals, using the two types of polymerase chain reaction-based molecular markers, has demonstrated co-inheritance of apomictic reproduction and two of the molecular markers. The remaining five molecular markers generally co-segregate with each other but are not linked with the mode of reproduction. These results suggest that genes for apomixis apparently can be transmitted by a single chromosome. Chromosome-specific markers will provide a starting point for the mapping of this genetically intractable reproductive trait.  相似文献

Analysis of 265 plants derived from 13 sites in the Rokko Mountains shows that the widely distributed eastern AsianEupatorium chinense var.simplicifolium consists of seven cytotypes. These polymorphic karyotypes comprise four levels of ploidy based on X=10 and partial deficiency, occurring either alone or in combination. The polyploid cytotypes exhibit anomalous meiosis and highly variable pollen stainability in contrast to the nearly-normal behaviour of the diploid. The polyploid cytotypes were revealed as being agamospermous. They are readily distinguishable from the diploid cytotypes by several morphological characters and by growth habits closely related to their respective niches. Two or more cytotypes occurred in all sites examined, the most common being the coexistence of triploid and tetraploid cytotypes. The frequencies of occurrence of the pentaploid plants and the cytotypes involving a deficient chromosome were relatively low. While the diploid cytotype is restricted to fragile gravitational slopes and rocky areas which are poor in species and lack tall competitors, the polyploids occur widely in the grasslands, the roadsides or the forest-margins, closely associated with tall grasses and forbs such asMiscanthus sinensis. Based on these data, theE. chinense var.simplicifolium complex is divided into two distinct groups: the diploid cytotype and the polyploid cytotypes.  相似文献

 Apomixis has never been reported in natural populations of pearl millet [Pennisetum glaucum (L.) R.Br.], although many wild relatives of pearl millet are obligate or facultative aposporous apomicts. Four-nucleate aposporous embryo sacs are formed from somatic cells of the nucellus that do not undergo meiosis. Two mutants of pearl millet, female sterile (fs) and stubby head, have two developmental characteristics in common: a significant reduction in head length compared with the wild-type and the formation of aposporous embryo sacs. Reproductive development in fs and stubby head mutants was examined in depth because of the potential for illuminating basic cellular or developmental factors that may function to alter embryo sac development. Genetic analysis of stubby head showed that this phenotype is conferred by genes at two loci linked in coupling within 29 cM. Crosses between fs and stubby head mutants showed that, despite the similarities in phenotypes, the mutations are at different loci. The mutants differ from wild-type in their inflorescence structure from the time of initiation of spikelet primordia through terminal differentiation of the ovule. Both mutations could be categorized as meristic, since a change in inflorescence branch or organ number was common and gynoecium development varied. We speculate that heterochronic development of the floral meristem and organ initiation/specification programs may be the underlying mechanism for phenotypic changes in these mutants throughout the floral phase.
