The behaviors of nuclei and microtubules (MT) in Acrosiphonia duriuscula (Ruprecht) Collins were observed in detail using fluorescence and electron microscopy. Numerous nuclei exist in cells of A. duriuscula (multinucleate cells). Cortical MT radiate from the apex of the tip cell and run parallel to its long axis. Between 30 and 40% of nuclei in the upper part of cytoplasm migrate downward to the region where cytokinesis will take place, and these numerous nuclei form a ‘nuclear ring’ before mitosis. The parallel array of the cortical MT changes to a transverse orientation at the region where cytokinesis will take place, and finally forms a characteristic circumferential band. Mitosis starts from the nuclei in the ring. Cortical MT disappear in the region of the nuclear ring and many mitotic spindles form. The band-shaped array of MT remains. Mitosis spreads in an apparent wave to the other nuclei. After mitosis, daughter nuclei that formed a nuclear ring migrate apically and repopulate the apical daughter cell. When the numerous daughter nuclei have relocated, a rearrangement of the cortical MT occurs. They are randomly arranged at first, but finally become parallel to the long axis of the cell. Cytokinesis occurs by furrowing of the cell, and the band-shaped array of MT could be detected at the leading edge of the furrow.  相似文献

The internal transcribed spacer regions of the nuclear ribosomal DNA cistron of green algal endophytes, previously identified as Chlorochytrium inclusum Kjellman and Codiolum petrocelidis Kuckuck, were sequenced. Culture studies have implicated marine Chlorochytrium and Codiolum -like cells in the life histories of a number of macrophytic green algae. The identity of the endophytes was resolved by comparing DNA sequences from unicells endophytic in the foliose red alga Mazzaella splendens (Setchell et Gardner) Hommersand and in the crustose phase of Mastocarpus papillatus (J. Agardh) Kylin, with sequences from species of Acrosiphonia, Urospora, Ulothrix, and Monostroma. All endophyte isolates were more closely related to species of Acrosiphonia than to those of Urospora, Ulothrix, or Monostroma. The results support previous culture studies, and conclusively identify the endophytes obtained in the field as alternate life history phases of one or more Acrosiphonia species in southern British Columbia, Canada.  相似文献

Abstract. The β-dimethylsulphoniopropionate (DMSP) concentrations of 5 Antarctic green algae grown at 2, 30 and 55 μmol photons m?2 s?1 were determined during a period of one year, cultivated under fluctuating daylengths mimicking the conditions of the natural habitat at King George Island. Antarctica. The intracellular DMSP content of all species decreased simultaneously with decreasing daylengths and vice versa. Additionally, the DMSP level was affected by the light intensity: the higher the photon fluence rate the greater the algal DMSP concentration. Under conditions of darkness, there was a degradation of the DMSP pool in members of the Acrosiphoniales, while the DMSP content in members of the Ulvales did not change. The results indicate a light-dependent DMSP accumulation in algae. Therefore, they may help to explain the seasonal variability of DMSP and its cleavage product dimethylsulphide (DMS) in coastal waters.  相似文献

Cyclin‐dependent kinases, the most notable of which is cdc2, are key regulators of the cell cycle and are highly conserved in evolution. We have cloned and analyzed one cDNA containing an open reading frame of 337 amino acids from a multinucleate, multicellular green alga, Acrosiphonia duriuscula (Ruprecht) Collins. The deduced protein, named Adcdc2‐1, showed 51% and 54% amino acid sequence identity to yeast cdc2/cdc28 and human cdc2, respectively. Several domains that characterize the cdc2‐related kinases were identified from this sequence, although the PSTAIRE motif was replaced with PPTTIRE. Furthermore, this protein has conserved Tyr and Thr residues that are sites of phosphorylation in cdc2‐related kinases and are important for regulating kinase activity. The present results suggest that the universal cdc2 is conserved in algae with unique structural characteristics.  相似文献

Phylogenetic relationships in the Arctic-Antarctic disjunct seaweed species Acrosiphonia arcta (Dillwyn) J. G. Agardh (Acrosiphoniales, Chlorophyta) were examined using restriction fragment-length polymorphism analysis of the fast-evolving nuclear ribosomal intergenic spacer (IGS) region and random amplified polymorphic DNA (RAPD) markers. Twenty-two isolates collected from 10 different locations in both hemispheres were compared. Five IGS length classes were identified among the 10 locations. Throughout the North Atlantic, IGS regions were found to be extremely homogeneous whereas RAPD patterns revealed subdivided populations that suggest founder effects. Acrosiphonia arcta populations found in the Arctic and North Atlantic oceans are hypothesized to be of Pacific origin. Extensive differences found between Arctic Greenland populations and those in the North Atlantic suggest that colonization of Arctic Greenland occurred as an independent event. Recolonization of the Antarctic peninsula from Southern Chile is favored, whereas the directionality of transequatorial passage along the western coast of the Americas could be in either direction.  相似文献
