Trypsin, chymotrypsin, cathepsins B and D, aminopeptidase and carboxypeptidases A and B were detected in body extracts of the storage mite Acarus farris (Oudemans) (Astigmata: Acaridae). Faeces-enriched medium exhibited higher (10-50-fold) specific protease activity rates than those measured with mite body extracts for trypsin, chymotrypsin and carboxypeptidases A and B, suggesting that they are involved in mite digestion. However, the activity of cathepsin B was only three-fold higher in faecal than in body extracts, indicating that its presence in the lumen of the digestive tract is low compared to that of serine proteases. The activity of aminopeptidases was higher in mite bodies, indicating that they might be membrane bound. Cathepsin D activity was only detected in body extracts, indicating that this enzyme is not a digestive protease in this species. Zymograms resolved three major bands of gelatinolytic activity, but at least one protease form was only present in body extracts. Protease inhibitors of different specificity were tested in vivo to establish their potential as control agents. The development of A. farris was significantly retarded when the immature stages were fed on artificial diet containing inhibitors of serine and cysteine proteases and aminopeptidases, whereas no such effect was found with inhibitors of aspartyl proteases and carboxypeptidases. Interestingly, the most significant effects on A. farris occurred when a combination of inhibitors targeting different enzyme classes was supplied mixed in the diet, suggesting a synergistic toxicity. Several plant lectins were also tested, but only wheat germ agglutinin and concanavalin-A affected development.  相似文献

The response to pirimiphos-methyl, in one strain of Acarus farris and two strains of Acarus siro, was assessed using an impregnated filter paper bioassay and by the selection of adults following exposure to pirimiphos-methyl. It was concluded that one of the strains of A. siro was resistant to pirimiphos-methyl and that a major resistance mechanism was involved. The second strain of A. siro gave a response similar to that of a laboratory strain unexposed to organophosphates and was considered to be susceptible. The A. farris strain responded to selection at the ED50 but not at the ED99, and it was concluded that a minor resistance mechanism is present in this strain. Assays of esterase activity were used to attempt to identify the biochemical mechanisms involved in the resistance detected by the bioassays. The A. farris and susceptible A. siro strains showed similar levels of esterase activity but the esterase activity of the resistant A. siro strain was significantly greater. An increase in esterase activity followed selection of both the A. farris strain and the resistant A. siro strain. An acetylcholinesterase assay showed no significant difference between the susceptible and pirimiphos-methyl selected strains of A. siro. The results suggest that esterases are involved in the resistance to pirimiphos-methyl found in A. siro and A. farris but that in A. siro, at least, other mechanisms may also be present.  相似文献

The developmental rate of immature stages and the reproduction of adults of Tyrophagus putrescentiae (Schrank), T. neiswanderi Johnston and Bruce and Acarus farris (Oudemans) were examined at 70, 80 and 90% r.h. and a constant temperature of 25°C. At 70% r.h., T. putrescentiae and A. farris immature stages failed to reach the protonymph stage as 100% of the larvae died, whereas T. neiswanderi was able to complete development. The developmental time of all immature stages for the three species was significantly increased as relative humidity was reduced. The mobile stages were particularly susceptible, as the time needed to complete their development at lower relative humidities suffered greater increases than the egg stage. At 70% r.h., T. putrescentiae and A. farris were not able to lay eggs and only 24% of T. neiswanderi pairs were fertile. The reproductive parameters of the three species at the relative humidities at which they were able to lay eggs showed significant differences, except for the percentage of fertile mating at 80 and 90% r.h. As relative humidity increased, preoviposition period was reduced and fecundity and daily fecundity was increased, whereas the oviposition period showed different patterns for the three species. The intrinsic rate of increase (r m ) of T. neiswanderi at 70% r.h. was negative indicating that, at these conditions, mite populations of this species will diminish until they disappear. As relative humidity increased from 80 to 90% r.h. this parameter was almost twofold for both Tyrophagus species. The r m obtained for A. farris at 90% r.h. was similar to that of T. neiswanderi at the same humidity while at 80% r.h. it was very small so that the population doubling time was more than 84 days. The influence of relative humidity on biology of these mites and its practical application as control measure are discussed.  相似文献

It is generally assumed that male control over mating and a lack of precopulatory female choice are prevalent in many animals and in astigmatan mites in particular. We show that several morphological structures of females of some astigmatan mites are indicative of precopulatory female choice: (1) copulatory tubes acting like intromittent organs; (2) specialized structures assisting male–female attachment and possibly allowing indirect mate selection in immature females; and (3) a unique, pad‐like terminal opisthosomal organ used to cling to the male during copulation in Glaesacarus rhombeus (= Acarus rhombeus Koch et Berendt, 1854) belonging to an extinct family, Glaesacaridae, from the Upper Eocene Baltic amber. An exceptionally well‐preserved copulating pair from amber provides insight into the function of this organ and reproductive behaviour in this mite. Female control over mating may reduce the timing of insemination, harassment by males, and damage caused by copulation. As a consequence, this can lessen male–male aggression, select against precopulatory guarding, and reduce the risk of predation. By contrast to extant taxa, males of G. rhombeus do not have any apparent specialized structures aiding clinging to the female during copulation, suggesting that this mating system is either an earlier step in the evolution of the female‐dominated mating system and/or a remarkable example of imbalanced female counteradaptations against the male's reproductive interest that may occur during an arms race between the two sexes. We offer an approach that can falsify the hypothesis assuming precopulatory female choice and discuss an alternative hypothesis suggesting that these female structures evolved in response to the need to reduce damage associated with mating or precopulatory guarding. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 102 , 661–668.  相似文献

Stored product mites are pests of serious economic and medical importance. Recently, it has been shown that the biological control of these pests based on the use of natural compounds and predators had a high potential for success. In this study, we investigated the suppression of three pest mites by combination of the predator Cheyletus malaccensis Oudemans, 1913 and bean flour (Phaseolus vulgaris), which is an antifeedant. Wheat grain (100 g) was contaminated with 100 individuals (Acarus siro (Linnaeus, 1758), Aleuroglyphus ovatus (Troupeau, 1878) or Tyrophagus putrescentiae (Schrank, 1781)). We added bean flour in concentrations of 0, 1, 5 and 10 g kg−1 (wt/wt) and/or the predator with initial predator/prey ratios 0, 1/50, 1/25 and 1/10. The experiment was carried out under abiotic conditions that are optimal for pest development. After 21 days, pest and predators numbers were recorded. Application of flour significantly reduced populations of A. siro, slightly reduced populations of T. putrescentiae, the efficiency increased with increasing concentration. A. ovatus was not affect by bean flour. Populations of all mite species were successfully reduced by the sole addition of C. malaccensis, with a higher efficiency at higher Cheyletus ratios. The additive effect of the predator and flour was mainly apparent in the reduction of T. putrescientae. The population of C. malaccensis was not affected by the presence of bean flour. It is therefore recommended to use bean flour for the reduction of A. siro, a combination of bean flour and Cheyletus for the reduction of T. putrescientae, and only Cheyletus for the reduction of A. ovatus. Traps have been used extensively to provide early warning of hidden pest infestations. To date, however, there is only one type of trap on the market in the U.K. for storage mites, namely the BT mite trap, or monitor. Laboratory studies have shown that under the test conditions (20 °C, 65% RH) the BT trap is effective at detecting mites for at least 10 days for all three species tested: Lepidoglyphus destructor, Tyrophagus longior and Acarus siro. Further tests showed that all three species reached a trap at a distance of approximately 80 cm in a 24 h period. In experiments using 100 mites of each species, and regardless of either temperature (15 or 20 °C) or relative humidity (65 or 80% RH), the most abundant species in the traps was T. longior, followed by A. siro then L. destructor. Trap catches were highest at 20 °C and 65% RH. Temperature had a greater effect on mite numbers than humidity. Tests using different densities of each mite species showed that the number of L. destructor found in/on the trap was significantly reduced when either of the other two species was dominant. Residual populations of storage mites sheltering in crevices and cracks escape conventional control treatments and are implicated in the infestation of newly harvested grain. In a series of 24 h laboratory tests, the performance of solitary adults of two predatory mite species, Cheyletus eruditus (Schrank) and Blattisocius tarsalis (Berlese), were assessed for controlling small numbers of the flour mite Acarus siro (L.). Tests were carried out in the presence or absence of prey refuges or grain debris to afford shelter to the flour mites. While C. eruditus had a significant effect on the motile stages of A. siro, in contrast B. tarsalis had a significant effect on the eggs. The maximum percentage of motile stages of A. siro eaten by C. eruditus was 82%, whereas the minimum percentage of A. siro eggs eaten by B. tarsalis was 99%. While the performance of C. eruditus in predating on motile stages of the flour mite was hindered by the presence of the prey refuge (38% eaten) and grain debris (25% eaten), the performance of B. tarsalis in predating on flour mite eggs was unaffected (100% eaten in presence of prey refuge or grain debris). In prolonged exposures (36 days) the performance of 2, 4 or 8 adult predators, either a single species or a combination of both, was assessed for their ability to control a population of the flour mite developing up to F2 from an initial inoculum of 80 females and 20 males, allowed to oviposit for 72 h in the absence of predatory mites. The maximum reduction in prey population of 80% was achieved with eight B. tarsalis. Combining the two predatory species did not enhance the reduction of A. siro population.  相似文献

Abstract.  Acarus siro is the most abundant and frequent mite to infest stored-food products, causing allergies and transmitting mycotoxin producing fungi. The predatory mite Cheyletus malaccensis is a candidate species in the biocontrol programme for this pest. In vitro , the α-amylase inhibitor acarbose is effective against the α-amylase of A. siro but not against that of C. malaccensis . In vivo , the impact of acarbose on a population of A. siro is investigated along with the interaction with the predator. Various densities of adult parthenogenetic females of C. malaccensis are reared on A. siro feeding on either a control diet or a diet containing different acarbose concentrations. The combined action of both factors significantly improved the final biocontrol efficiency with C. malaccensis , compensating for the lower energetic content of the prey on acarbose by increasing the number of prey caught. Acarbose had no negative effects on the longevity and the length of oviposition period of C. malaccensis but partially reduced its fecundity. The results are discussed in the context of the integrated control of stored-product mite pests.  相似文献
