An evolutionarily stable strategy (ESS) is only required to be capable of resisting invasion by rare mutant strategies. In contrast, an absolute invader strategy (AIS) is a rare mutant strategy that can invade any established strategy. We show that the predictions of the outcome of evolution made by optimization models are compatible with those made by the classical expected payoff comparisons in matrix games. We also show that if a matrix game has an AIS that AIS is unique and is also an ESS. But an ESS need not be an AIS. In pure-strategy submodels, an AIS need not be unique. An AIS of a matrix game has global asymptotic stability property in the game dynamics which involve only pure strategies including the AIS.  相似文献

Finding structural similarities between proteins often helps reveal shared functionality, which otherwise might not be detected by native sequence information alone. Such similarity is usually detected and quantified by protein structure alignment. Determining the optimal alignment between two protein structures, however, remains a hard problem. An alternative approach is to approximate each three-dimensional protein structure using a sequence of motifs derived from a structural alphabet. Using this approach, structure comparison is performed by comparing the corresponding motif sequences or structural sequences. In this article, we measure the performance of such alphabets in the context of the protein structure classification problem. We consider both local and global structural sequences. Each letter of a local structural sequence corresponds to the best matching fragment to the corresponding local segment of the protein structure. The global structural sequence is designed to generate the best possible complete chain that matches the full protein structure. We use an alphabet of 20 letters, corresponding to a library of 20 motifs or protein fragments having four residues. We show that the global structural sequences approximate well the native structures of proteins, with an average coordinate root mean square of 0.69 Å over 2225 test proteins. The approximation is best for all α-proteins, while relatively poorer for all β-proteins. We then test the performance of four different sequence representations of proteins (their native sequence, the sequence of their secondary-structure elements, and the local and global structural sequences based on our fragment library) with different classifiers in their ability to classify proteins that belong to five distinct folds of CATH. Without surprise, the primary sequence alone performs poorly as a structure classifier. We show that addition of either secondary-structure information or local information from the structural sequence considerably improves the classification accuracy. The two fragment-based sequences perform better than the secondary-structure sequence but not well enough at this stage to be a viable alternative to more computationally intensive methods based on protein structure alignment.  相似文献

Adolescent Idiopathic Scoliosis (AIS) is a 3D spinal deformity characterized by curvature and rotation of the spine. Markerless surface topography (ST) analysis has been proposed for diagnosing and monitoring AIS to reduce the X-ray radiation exposure to patients. This method captures scans of the cosmetic deformity of the torso using visible, radiation-free light. The asymmetry analysis of the torso, represented as a deviation contour map with deviation patches outlining the areas of cosmetic asymmetries, has previously been shown to predict the severity and progression of the condition in comparison with radiographs, by using classification trees. While the classification results were promising, it was reported that some mild curves were erroneously diagnosed. Furthermore, this approach is highly sensitive to threshold values selected in the decision trees. Therefore, this study aims to define a custom Neighbourhood Classifier algorithm for AIS classification to improve the accuracy, sensitivity, and specificity of predicting curve severity and curve progression in AIS. Curve severity was predicted with 80% accuracy (sensitivity = 81%; specificity = 79%) for thoracic-thoracolumbar curves and 72% (sensitivity = 93%; specificity = 53%) for lumbar curves. This represents an improvement over the previous method with curve severity accuracies of 77% and 63% for thoracic-thoracolumbar and lumbar curves, respectively. Additionally, curve progression was predicted with 93% accuracy (sensitivity = 83%; specificity = 95%) representing a substantial improvement over the previous method with an accuracy of 59%. The current method has shown the potential to further reduce radiation exposure for AIS patients by avoiding X-rays for mild and non-progressive curves identified using ST analysis.  相似文献

Highly polarized neurons need to carefully regulate the distribution of organelles and other cargoes into their two morphologically and functionally distinct domains, the somatodendritic and axonal compartments, to maintain proper neuron homeostasis. An outstanding question in the field is how organelles reach their correct destination. Long-range transport along microtubules, driven by motors, ensures a fast and controlled availability of organelles in axons and dendrites, but it remains largely unclear what rules govern their transport into the correct compartment. Here, we review the emerging concepts of polarized cargo trafficking in neurons, highlighting the role of microtubule organization, microtubule-associated proteins, and motor proteins and discuss compartment-specific inclusion and exclusion mechanisms as well as the regulation of correct coupling of cargoes to motor proteins.  相似文献

Different tracking devices are currently used to record the activity of fishing vessels in space and time and to analyse the pattern of fishing effort with respect to sea substrates, resources, or socio-economics factors. Among these devices, the Vessel Monitoring System (VMS) and the Automatic Identification System (AIS) represent the most interesting and fertile data sources for fisheries investigations but an integration of these two sources was never explored. Two large VMS and AIS datasets, covering the activity of the Italian fishing vessels with length-over-all ≥15 m for the whole year 2012, were used to perform the first assessment of the features characterizing each data source, and ultimately the properties of the estimators of ecological indicators of fishing pressure obtained using these data. In this way, a dataset related to five vessels equipped by a third high frequency device (the Fishing Observing System) was used as reference for a comparative analysis of fishing activity as reconstructed by VMS and/or AIS. Moreover, the ecological indicator 5 of fishing pressure as spatial extent of fishing activity (defined within the Data Collection Framework for Fisheries—DCF) was used as a reference to test the separate and integrated usage of VMS and AIS data. Results evidenced important differences in spatial, temporal and fleet coverage between VMS and AIS. Namely, VMS is affected by a lower frequency with respect to AIS but covers a larger portion of the fleet. Furthermore, AIS mainly operates near the harbours, so that offshore activity is underrepresented. The integration of VMS and AIS data, however, allows valorising the strengths and minimizing the limits of each source. In fact, the assessment of the total spatial extent of fishing activities (as captured by the DCF indicator 5) is significantly improved by the integrated usage of VMS and AIS data.  相似文献

Androgen insensitivity syndrome (AIS) is an X-linked recessive genetic disorder with a normal 46, XY karyotype caused by abnormality of the androgen receptor (AR) gene. One Chinese family consisting of the proband and 5 other members with complete androgen insensitivity syndrome (CAIS) was investigated. Mutation analysis by DNA sequencing on all 8 exons and flanking intron regions of the AR gene revealed a unique large deletion/insertion mutation in the family. A 287 bp deletion and 77 bp insertion (c.933_1219delins77) mutation at codon 312 resulted in a frameshift which caused a premature stop (p.Phe312Aspfs*7) of polypeptide formation. The proband's mother and grandmother were heterozygous for the mutant allele. The proband's father, uncle and grandfather have the normal allele. From the pedigree constructed from mutational analysis of the family, it is revealed that the probably pathogenic mutation comes from the maternal side.  相似文献

In Eukarya, phosphatidylinositol (PI) is biosynthesized from CDP-diacylglycerol (CDP-DAG) and inositol. In Archaea and Bacteria, on the other hand, we found a novel inositol phospholipid biosynthetic pathway. The precursors, inositol 1-phosphate, CDP-archaeol (CDP-ArOH), and CDP-DAG, form archaetidylinositol phosphate (AIP) and phosphatidylinositol phosphate (PIP) as intermediates. These intermediates are dephosphorylated to synthesize archaetidylinositol (AI) and PI. To date, the activities of the key enzymes (AIP synthase, PIP synthase) have been confirmed in only three genera (two archaeal genera, Methanothermobacter and Pyrococcus, and one bacterial genus, Mycobacterium). In the present study, we demonstrated that this novel biosynthetic pathway is universal in both Archaea and Bacteria, which contain inositol phospholipid, and elucidate the specificity of PIP synthase and AIP synthase for lipid substrates. PIP and AIP synthase activity were confirmed in all recombinant cells transformed with the respective gene constructs for four bacterial species (Streptomyces avermitilis, Propionibacterium acnes, Corynebacterium glutamicum, and Rhodococcus equi) and two archaeal species (Aeropyrum pernix and Sulfolobus solfataricus). Inositol was not incorporated. CDP-ArOH was used as the substrate for PIP synthase in Bacteria, and CDP-DAG was used as the substrate for AIP synthase in Archaea, despite their fundamentally different structures. PI synthase activity was observed in two eukaryotic species, Saccharomyces cerevisiae and Homo sapiens; however, inositol 1-phosphate was not incorporated. In Eukarya, the only pathway converts free inositol and CDP-DAG directly into PI. Phylogenic analysis of PIP synthase, AIP synthase, and PI synthase revealed that they are closely related enzymes.  相似文献

alpha-Galactosidase (EC 3.2.1.22) from ripe papaya (Carica papaya L.) fruit was fractionated by a combination of ion exchange and gel filtration chromatography into three forms, viz., alpha-galactosidase 1, 2 and 3. The predominant isoform, alpha-gal 2, was probably a tetramer with a native molecular mass of about 170 kDa and 52 kDa-sized subunits and an estimated pI of 7.3. The subunit's N-terminal amino acid sequence shared high identity (97%) with the deduced sequence of a papaya cDNA clone encoding a putative alpha-galactosidase PAG2 as well as with an Ajuga reptans L. GGT1 clone encoding a galactan: galactan galactosyltransferase (66%). During ripening, alpha-galactosidase activity increased concomitantly with firmness loss and this increase was largely ascribed to alpha-gal 2. The protein level of alpha-gal 2 as estimated by immunoblot was low in developing fruits and generally increased with ripening. alpha-Galactosidase 2 also had the ability to markedly catalyse increased pectin solubility and depolymerisation while the polymers were still structurally attached to the cell walls mimicking, in part, the changes that occur during ripening. The close correlation between texture changes, alpha-gal 2 activity and protein levels as well as capability to modify intact cell walls suggest that the enzyme might contribute to papaya fruit softening during ripening. The purported mechanism of alpha-gal 2 action as a softening enzyme was discussed in terms of its functional capacity as a glycanase or perhaps, as a transglycosylase.  相似文献
