Two electroejaculators were used to collect semen from 40 adult male coyotes. The most effective apparatus used a two-ring rectal probe and an AC voltage of 18 (Vrms) at 1000 Hz. With this ejaculator, 11 of 15 coyotes produced a satisfactory semen sample, which averaged 0.9 ml in volume and 70 million spermatozoa per ml.  相似文献

A new procedure for the analyses of tryptophan and the total amino acid composition of proteins was based on the observations that pyridine borane reduces tryptophan in trifluoroacetic acid, while other amino acids remain intact [M. Kurata, Y. Kikugawa, T. Kuwae, I. Koyama, and T. Takagi (1980) Chem. Pharm. Bull. 28, 2274-2275; W.S.D. Wong, D.T. Osuga, and R.E. Feeney (1984) Anal. Biochem. 139, 58-67]. Concentrated HCl was used instead of trifluoroacetic acid for analytical purposes. The products were stable to hydrolysis in 6 N HCl, and the reduction did not interfere with hydrolysis and subsequent analyses. Quantitative recovery was achieved with most proteins when they were subjected to acid reduction in ice-cooled concentrated HCl with two incremental additions of pyridine borane. The reaction was terminated after 10 min by dilution with an equal volume of H2O, vacuum sealing, and hydrolyzing at 110 degrees C for 22 h. The yields of the expected values for cytochrome c, catalase, bovine serum albumin, subtilisin BPN', trypsin, chymotrypsin, beta-lactoglobulin, lysozyme, and pepsin were obtained. Ovotransferrin and ovalbumin, however, yielded values for tryptophan lower than literature values. With two different ion-exchange methods, the recoveries of all other amino acids were comparable to those obtained by acid hydrolysis with 6 N HCl. Since the same hydrolysate can be analyzed for both tryptophan and all the other amino acids, the procedure is a more convenient method than those requiring separate determinations. Initial results indicate that the method may be applied to high-performance liquid chromatographic procedures with adaptations of the protocols if necessary.  相似文献

The last step in the biosynthesis of cyanogenic glucosides, the glucosylation of the cyanohydrin intermediate, has been investigated in detail using Triglochin maritima seedlings. The glucosyltransferase activity is not associated with membranes and appears to be a "soluble" enzyme. The cyanohydrin intermediate, which is formed by hydroxylation of 4-hydroxyphenylacetonitrile by a membrane-bound enzyme, is free to equilibrate in the presence of the glucosyltransferase and UDPG, because it can be trapped very efficiently. This indicates that this intermediate is not channeled (unlike some of the other intermediates), although it is probably the most labile of all of them. The glucosyltransferase of T. maritima responsible for the glucosylation of the cyanohydrin was separated from another glucosyltransferase, which used 4-hydroxybenzylalcohol as a substrate, and purified over 200-fold. It catalyzed the glucose transfer from UDPG to only 4-hydroxymandelonitrile and 3,4-dihydroxymandelonitrile, giving rise to the respective cyanogenic glucosides. Although the activities with these two substrates behaved differently in certain respects (e.g., extent of inactivation during purification and difference in activation by higher salt concentrations), most of the data acquired favor the view that only one enzyme in T. maritima is responsible for the glucosylation of both substrates.  相似文献

CH3Hg(II)OH forms complexes at pH 8 with tyrosine and with tyrosine ethyl ester (TEE) that are detected by ultraviolet difference absorption spectra. With Kf defined by CH3HgOH + HB
CH3HgB + H2O, we find log Kf = 3.61 (tyrosine) and 3.36 (TEE). A heavy-atom effect is observed in frozen glasses of the complexes; this indicates a close interaction between Hg and the chromophore. No UV difference spectrum or heavy-atom effect is observed with N-acetyl tyrosine ethyl ester, indicating that complexing at the phenol O does not occur, and suggesting that binding occurs at the amine N. Zero field optically detected magnetic resonance (ODMR) measurements of the CH3Hg(II)-tyrosine triplet state give (D, E) = (0.129, 0.047) or (0.134, 0.041) cm?1 depending upon assignment of transitions. D of tyrosine is relatively unaffected, but E is reduced by CH3Hg(II) complexing. Low-temperature kinetic measurements show that the shortest lived sublevel of the complex is Tz, where z lies along the phenol long axis in tyrosine. A dominant 11.6-msec component in the 77 K decay of the phosphorescence is consistent with the individual sublevel lifetimes obtained by ODMR.  相似文献

The intertidal gastropod, Tegula funebralis (A. Adams) exhibits a shore-level size gradient with mean shell size increasing in a down-shore direction. Snails transferred to zones where they do not usually occur migrated back towards their original zone, thus re-establishing a size gradient and implying differential movement among size classes. Both large (≥2.1 cm shell width) and small (≤ 1.77 cm) snails were photonegative on a horizontal surface and geonegative in the laboratory; there were no statistical differences between size classes. Light, however, inhibited upward, or caused downward, movement of large snails on vertical surfaces. Small snails were unaffected, ranging higher on illuminated vertical surfaces than large snails. Both sizes exhibited similar distributions in the dark. In an experimental chamber providing both emersed and immersed surfaces, T. funebralis established vertical size gradients when the chamber was illuminated from above. It is suggested that light is an important factor in the formation and maintenance of Tegula's shore-level size gradient.In response to water-borne chemicals derived from the sea star Pisaster ochraceus (Brandt), large snails moved up vertical surfaces in greater proportion than small. In response to contact with the predator, large snails moved away faster than small and individuals collected from crevices in the field moved away slower than those collected from open rock faces. Although predation may select for a size gradient in Tegulafunebralis, it is unlikely that responses to predatory sea stars directly and proximally cause or maintain them over the short term.  相似文献

Complexes of purified 40S ribosomal subunits and initiation factor 3 from rabbit reticulocytes were crosslinked using the reversible protein crosslinking reagent, 2-iminothiolane, under conditions shown previously to lead to the formation of dimers between 40S proteins but not higher multimers. The activity of both the 40S subunits and initiation factor 3 was maintained. Protein crosslinked to the factor was purified by sucrose density gradient centrifugation following nuclease digestion of the ribosomal subunit: alternatively, the total protein was extracted from 40S: factor complexes. The protein obtained by either method was analyzed by two-dimensional diagonal polyacrylamide/sodium dodecyl sulfate gel electrophoresis. Ribosomal proteins were found in multimeric complexes of high molecular weight due to their crosslinking to components of eIF3. Identification of the ribosomal proteins appearing below the diagonal was accomplished by elution, radioiodination, two-dimensional polyacrylamide/urea gel electrophoresis, and radioautography. Proteins S2, S3, S3a, S4, S5, S6, S8, S9, S11, S12, S14, S15, S16, S19, S24, S25, and S26 were identified. Because many of the proteins in this group form crosslinked dimers with each other, it was impossible to distinguish proteins directly crosslinked to eIF3 from those crosslinked indirectly through one bridging protein. The results nonetheless imply that the 40S ribosomal proteins identified are at or near the binding site for initiation factor 3.  相似文献

Periodate treatments of apo human serum transferrin (HST), and apo chicken ovotransferrin (COT) were previously reported to cause a rapid loss of Fe+3 binding capacity, with a loss of 3 to 5 tyrosine residues [P. AZARI AND J. L. PHILLIPS (1970) Arch. Biochem. Biophys. 138, 32-38; K. F. GEOGHEGAN, J. L. DALLAS, AND R. E. FEENEY (1980) J. Biol. Chem. 255, 11429-11434]. The effects of periodate and hydrogen peroxide on human lactotransferrin (HLT), HST, and COT have been compared. All three apotransferrins were rapidly inactivated and lost approximately 4 to 5 tyrosine residues by 5 mM periodate treatment; their iron complexes had little or no inactivation and losses of approximately 1 to 2 tyrosine residues. All three iron transferrins were highly resistant to inactivation by 5 mM periodate in bicarbonate, with or without the addition of phosphate, while in phosphate (with ambient carbonate) Fe2HLT was highly resistant, Fe2COT slightly less resistant, and Fe2HST much less resistant. Similar oxidations of methionines to the sulfoxides were found in both the apo and iron forms. After 150 min of 5 mM periodate treatment HST lost approximately 3 (apo 3.1, iron 2.8) of 9, HLT approximately 3 (apo 2.6, iron 2.9) of 6, and COT approximately 7 (apo 7.2, iron 7.2) of 11 methionines per mole of protein. In the presence of 8 M urea HST had essentially all of its methionine residues oxidized by periodate, but only lost part of its activity on renaturation. Treatment of all apo transferrins with 300 mM hydrogen peroxide resulted in little or no losses (less than 10%) in activity. HST lost approximately one-third of its methionines and no tyrosines during the 300 mM hydrogen peroxide treatment. Therefore the essentiality of tyrosines for all three transferrins was confirmed and the nonessentiality of methionines was demonstrated.  相似文献

Purification and characterization of pregastric esterase from calf   总被引:1,自引:0,他引:1  
Calf pregastric esterase (PGE) was purified from calf gullet tissues. The tissue was excised and lyophilized, and lipid materials were extracted with acetone and n-butanol at -20 degrees C. Proteins were extracted from the delipidated tissue with a buffer containing a chaotropic salt (NaSCN) to solubilize hydrophobically bound protein aggregates. Calf PGE precipitated from the crude extract at pH 5.0. The precipitated, solubilized proteins were subjected to anion-exchange chromatography on DEAE-Sephacel, and the enzymatic activity was eluted using a linear gradient from 0.10 to 0.50 M NaCl at pH 8.0. Fractions with high specific activities were then chromatographed twice using gel filtration on Sephadex G-100. The resultant enzyme was shown to be pure upon discontinuous electrophoresis in 12% polyacrylamide containing 0.1% sodium dodecyl sulfate (SDS-PAGE). From SDS-PAGE gel patterns, a molecular weight of 49,000 was determined. The amino acid composition of the enzyme allowed calculation of an "average hydrophobicity" (Bigelow index) of 1150 cal/residue. This indicates that calf PGE is relatively hydrophobic, being similar to proteins such as alpha-lactalbumin and bovine serum albumin in average hydrophobicity.  相似文献

Radiolabeled vanadium as either vanadyl ion or vanadate ion was injected intravenously into adult beagle dogs, and blood samples were collected at various times up to 48 hr post injection. For each sample, the distribution of vanadium between the cells and the plasma was determined, and the plasma was analyzed by electrophoresis to identify specific vanadium-binding proteins. Initially, vanadyl ion left the bloodstream more rapidly than vanadate, but the rates equalized after about 5 hr. A significant fraction of the vanadium in blood was associated with the cellular component following injection of both forms of vanadium. About 77% of the plasma vanadium was eventually bound by the serum iron transport protein transferrin, regardless of the vanadium species initially injected. For both vanadyl and vanadate, about 30 hr were required to reach the maximum degree of transferrin binding.  相似文献

The form and distribution of selenium (Se) in proteins from selected tissues of the rat were studied by measuring 75Se radioactivity in animals provided for 5 months with [75Se]selenite as the main dietary source of Se. Equilibration of the animals to a constant specific activity of 75Se allowed the measurement of 75Se to be used as a specific elemental assay for Se. Skeletal muscle, liver and blood accounted for 73% of the whole-body Se and 95% of the total Se-dependent glutathione peroxidase activity. Over 80% of the whole-body Se was in protein in the form of the selenoamino acid, selenocysteine. All other forms of Se that were measured accounted for less than 3% of the whole-body Se. The Se in protein was distributed in seven subunit sizes and nine chromatographic forms. The Se in glutathione peroxidase accounted for one-third of the whole-body Se. These results show that the main use of dietary Se, as selenite, in rats is for the synthesis of selenocysteine-containing proteins. Furthermore, the presence of two-thirds of the whole-body Se in nonglutathione peroxidase, selenocysteine-containing proteins suggests that there may be other important mammalian selenoenzymes besides glutathione peroxidase.  相似文献
