N-ras is one of the transforming genes in human hepatic cancer cells.It has been found that N-ras was overexpressed at the mRNA and protein level in hepatoma cells.In order to explore the biological roles of N-ras in human hepatic carcinogenesis and the potential application in control of cancer cell growth,a preudotype retrovirus containing antisense sequence of human N-ras was constructed and packaged.A recombinant retrovirus vector containing antisense or sense sequences of N-ras cDNA was constructed by pZIP-NeoSV(X)1.The pseudotype virus was packaged ang rescued by transfection and infection in PA317 and ψ 2 helper cells.It has been demonstrated that the pseudotype retrovirus containing antisense N-ras sequence did inhibit the growth of human PLC/PRF/5 hepatoma cells accompanied with inhibition of p21 expression,while the retrovirus containing sense sequence had none.The pseudotype virus had no effect on human diploid fibroblasts.  相似文献

IL-15及IL-15受体 (IL-15R)阳性细胞在成人T淋巴细胞性白血病 (ATL)、多发性骨髓瘤及炎症性自身免疫性疾病病理过程中起着重要作用。为研制特异的可消除IL-15受体阳性异常细胞的新型导向药物 ,将人IL-15拮抗剂 (IL-15M)基因片段与人工改造的绿脓杆菌外毒素 (PE)变异体 (PEΔ293)基因按正确的阅读框架融合 ,定向克隆在pET16b表达载体T7启动子的下游 ,得到质粒pET-IL15M-PEΔ293。从大肠杆菌相应重组菌株中通过Ni2+ NTA亲和层析纯化出融合蛋白IL15M-PEΔ293。IL15M-PEΔ293对IL-15R阳性红白血病细胞系K562及其多药耐药细胞系K562 AO2 均具有杀伤作用 ,对IL-15R阴性细胞系Jur kat则没有明显的杀伤作用 ,而且过量的重组IL-15可以完全阻断融合蛋白对K562的细胞毒效应 ,说明融合蛋白的细胞毒作用具有靶向性。这些结果提示本文构建的融合蛋白在与IL-15IL-15R异常表达相关的疾病甚至耐药性肿瘤的治疗中具有潜在的应用价值。  相似文献

This study characterized the attachment of chondrocytes to RGD-functionalized alginate by examining the effect of substrate stiffness on cell attachment and morphology. Bovine chondrocytes were added to wells coated with 2% alginate or RGD-alginate. The alginate was crosslinked with divalent cations ranging from 1.25 to 62.5 mmol/g alginate. Attachment to RGD-alginate was 10-20 times higher than attachment to unmodified alginate and was significantly inhibited by antibodies to integrin subunits α3 and β1, cytochalasin-D, and soluble RGD peptide. The equilibrium level and rate of attachment increased with crosslink density and substrate stiffness. Substrate stiffness also regulated chondrocyte morphology, which changed from a rounded shape with nebulous actin on weaker substrates to a predominantly flat morphology with actin stress fibers on stiffer substrates. The dependence of attachment on integrins and substrate stiffness suggests that chondrocyte integrins may play a role in sensing the mechanical properties of the matrices to which they are attached.  相似文献

为获取紫背天葵(Cynura bicolor DC.)叶片中花青素种类及其合成调控基因等信息,该试验以紫背天葵叶背面紫色以及经处理叶背面几乎全绿(对照)的叶片为材料,进行转录组测序分析,同时进行6个相关差异表达基因的qRT-PCR分析和6种花青素苷元的HPLC检测,以揭示紫背天葵特有的花青素苷元及其合成调控关键基因信息。结果表明:(1)在紫背天葵中共获得14个花青素苷元及32个花青素合成调控基因信息,其中表达量差异显著下调的4个基因为Pg(c11692)、Cy(c42112)、ANS(c38551)和3GT(c9064),表达量差异显著上调的2个基因是D FR(c35961)和3GT(c20283)。(2)qRT-PCR检测结果显示,上述6个基因在2种紫背天葵叶中的表达趋势(上调或下调)与转录组测序结果完全一致,但转录组测序检测到的表达趋势差异倍数比qRT-PCR检测结果更加明显。(3)HPLC分析显示,紫背天葵叶中均未检测到Dp、Pt、Pn及Mv等4类花青素苷元,但紫背天葵叶中富含Cy花青素苷元,且背面紫色的叶中Cy类花青素苷元含量(62.21 mg/kg)显著高于绿色叶对照(6.86 mg/kg);背面紫色和全绿叶中的Pg花青素苷元含量均低于0.43 mg/kg。研究推测,Cy和Pg花青素苷元在绿叶紫背天葵(对照)中含量显著降低可能是因为存在1个ANS和1个3GT正调控以及1个DFR和1个3GT负调控所致。  相似文献

The peroxisome proliferator-activated receptor gamma (PPARgamma) Pro12Ala polymorphism affects plasma lipids, but to what extent alcohol intake interferes with this association remains unknown. We randomly recruited 251 nuclear families (433 parents and 493 offspring) in the framework of the European Project on Genes in Hypertension study and genotyped 926 participants in whom all serum lipid variables and information on alcohol consumption were available for PPARgamma2 Pro12Ala. Genotype-phenotype relations were assessed using generalized estimating equations (GEE) and a quantitative transmission disequilibrium test (QTDT). The Ala12 allele was more frequent in Novosibirsk (0.17) than in Cracow (0.12) and Mirano (0.11) (P < 0.01). Using GEE (P = 0.03) or QTDT (P = 0.007), Italian offspring carrying the Ala12 allele had higher serum HDL cholesterol than noncarriers. HDL cholesterol levels were on average 0.086 mmol/l (P = 0.001) higher in drinkers than in nondrinkers. Compared with Pro12 homozygotes, Ala12 allele carriers consuming alcohol had higher serum total and HDL cholesterol, with the opposite trend occurring in nondrinkers. This genotype-alcohol interaction was independent of the type of alcoholic beverage and more pronounced in moderate than in heavy drinkers. We conclude that alcohol intake modulates the relation between the PPARgamma2 Pro12Ala and HDL cholesterol level and that, therefore, the Pro12Ala polymorphism, pending confirmation of our findings, might affect cardiovascular prognosis.  相似文献

Caiçaras are inhabitants of the Atlantic Forest coast in SE Brazil. We studied the uses of plants by five Caiçara communities and compared medicinal plant citations by informants in coastal and island communities. We use diversity indices to evaluate the use of plants and to compare communities. There is a high diversity of plants used in the Atlantic Forest coasts: we found 276 species used for food, medicine and construction. Caiçaras rely on folk medicine, and medicinal plants were especially cited in interviews. Following predictions of island biogeography theory, we found a lower diversity of medicinal plants cited in islands compared to continental communities.  相似文献
