The direct binding of porcine pancreatic phospholipase A2 and its zymogen to 1,2-bis(heptanylcarbamoyl)-rac-glycerol 3-sulfate was studied by optical detection of triplet-state magnetic resonance spectroscopy in zero applied magnetic field. The zero-field splittings of the single Trp3 residue undergo significant changes upon binding of phospholipase A2 to lipid. Shifts in zero-field splittings, characterized mainly by a reduction of the E parameter from 1.215 to 1.144 GHz, point to large changes in the Trp3 local environment which accompany the complexing of phospholipase A2 with lipid. This may be attributed to Stark effects caused by the binding of a charged group near Trp3 in the enzyme-lipid complex. The cofactor, Ca2+, which is strongly bound to the enzyme active site, has an influence on the bonding, as reflected by smaller zero-field splitting shifts. A relatively small change in the Trp environment was observed for the interaction of the zymogen with lipid.  相似文献

The foliage leaf epidermis of 35 species representing 12 key genera of woody bamboos of the Asian tropics was investigated using light and scanning electron microscopy. The results indicated that papillae forms and distributional patterns around the stomatal apparatus of the abaxial foliage leaf epidermis were usually constant and were of great taxonomic significance at the specific and generic levels. However, papillae characters were not suitable for dividing subtribes within woody bamboos of the Asian tropics. On the basis of papillae characters, Schizostachyum s.s. and Cephalostachyum were confirmed, but their delimitations should be modified. The transfer of Leptocanna chinensis and Schizostachyum sanguineum into Cephalostachyum was supported, and Cephalostachyum virgatum and C .  pergracile were confirmed to be members of Schizostachyum s.s. The subtribe Racemobambosinae did not obtain support and Racemobambos appeared to be better placed in subtribe Bambusinae. Neomicrocalamus was supported as a close relative and better treated as a synonym of Racemobambos . The nucleus of a blastula cell from Tilapia (Oreochromis nilotica, family Cichlidae, order Perciformes) was transplanted into an enucleated egg of Loach (Paramisgurnus dabryanus, family Cobitidae, order Cypriniformes). From among 3747 nucleo-cytoplasmic hybrid (NCH) eggs two NCH larval fish (0.05%) were obtained; one died on the 6th day and the other died on the 12th day after the operation. Morphological examinations showed that both NCH larval fish had developed normally with an opened mouth except they could not take food after complete utilization of their egg yolk on the 5th day of development. The possible mechanisms for obtaining such inter-order NCH larval fish are discussed. This is the first report indicating that inter-order NCH larval fish can be obtained in spite of their evolutionary divergence.  相似文献
