目的探讨接种人巨细胞病毒(Humancytomegalovirus,HCMV)是否引起Wistar大鼠、昆明种小鼠眼组织损伤.方法将HCMVAD169毒株经静脉接种Wistar大鼠和昆明种小鼠,观察动物眼部发病情况,原位杂交检测动物眼组织中的HCMVDNA片段.结果接种病毒后部分动物缓慢出现眼部发病,局部分泌物增多、浑浊甚至失明;经原位杂交于视锥、视杆细胞和角膜内皮细胞中检出HCMVDNA片段.结论HCMV可以感染动物眼组织,并引起动物发生眼病. 相似文献

Morusin is a pure compound isolated from root bark of Morusaustralis (Moraceae). In this study, we demonstrated that morusin significantly inhibited the growth and clonogenicity of human colorectal cancer HT-29 cells. Apoptosis induced by morusin was characterized by accumulation of cells at the sub-G1 phase, fragmentation of DNA, and condensation of chromatin. Morusin also inhibited the phosphorylation of IKK-α, IKK-β and IκB-α, increased expression of IκB-α, and suppressed nuclear translocation of NF-κB and its DNA binding activity. Dephosphorylation of NF-κB upstream regulators PI3K, Akt and PDK1 was also displayed. In addition, activation of caspase-8, change of mitochondrial membrane potential, release of cytochrome c and Smac/DIABLO, and activation of caspase-9 and -3 were observed at the early time point. Downregulation in the expression of Ku70 and XIAP was exhibited afterward. Caspase-8 or wide-ranging caspase inhibitor suppressed morusin-induced apoptosis. Therefore, the antitumor mechanism of morusin in HT-29 cells may be via activation of caspases and inhibition of NF-κB. 相似文献

Berberine (BBR) is one of the main constituents in Rhizoma coptidis and it has widely been used for the treatment of diabetic nephropathy. The aims of the study were to investigate the effects and mechanism of action of berberine on renal damage in diabetic rats. Diabetes and hyperglycaemia were induced in rats by a high-fat diet and intraperitoneal injection of 40 mg/kg streptozotocin (STZ). Rats were randomly divided into 5 groups, such as i) control rats, ii) untreated diabetic rats iii) 250 mg/kg metformin-treated, iv and v) 100 and 200 mg/kg berberine-treated diabetic rats and treated separately for 8 weeks. The fasting blood glucose, insulin, total cholesterol, triglyceride, glycosylated hemoglobin were measured in rats. Kidneys were isolated at the end of the treatment for histology, Western blot analysis and estimation of malonaldehyde (MDA), superoxide dismutase (SOD) and renal advanced glycation endproducts (AGEs). The results revealed that berberine significantly decreased fasting blood glucose, insulin levels, total cholesterol, triglyceride levels, urinary protein excretion, serum creatinine (Scr) and blood urea nitrogen (BUN) in diabetic rats. The histological examinations revealed amelioration of diabetes-induced glomerular pathological changes following treatment with berberine. In addition, the protein expressions of nephrin and podocin were significantly increased. It seems likely that in rats berberine exerts an ameliorative effect on renal damage in diabetes induced by high-fat diet and streptozotocin. The possible mechanisms for the renoprotective effects of berberine may be related to inhibition of glycosylation and improvement of antioxidation that in turn upregulate the expressions of renal nephrin and podocin. 相似文献

山水林田湖草系统保护与修复工程实施的重要目标是维护和提升区域生态系统服务。从乌梁素海流域山水林田湖草的生态现状与功能出发,对乌梁素海流域水土流失、土地沙化等生态敏感性及土壤保持、水源涵养、生物多样性等生态功能重要性进行系统评价,形成基于生态敏感性和生态功能重要性相结合的空间格局评价结果。以维护和提升人类福祉所需的重要生态系统服务为目标,以乌梁素海流域生态敏感性和生态功能重要性相结合的空间格局评价结果为基础,制定了“一中心、二重点、六要素、七工程”的乌梁素海山水林田湖草生态保护与修复体系,并基于此将乌梁素海流域生态保护修复分为6个主要治理区域,形成“四区、一带、一网”的生态安全格局,通过具体工程实施,流域生态环境质量和生态服务能力将取得明显提升,防风固沙能力有效增强,生物多样性持续改善,水环境质量稳定达标,生态系统的稳定性明显加强。通过乌梁素海流域的分析案例为流域山水林田湖草生态保护与修复关键区域的识别提供了定量分析方法,为流域尺度构建生态安全格局、实现山水林田湖草系统保护和修复提供思路和途径。 相似文献

对蝉棒束孢菌子实体(0.75g/kg)重复灌胃SD雄性大鼠90d及恢复28d的早期肾损伤生物标记物肾损伤分子-1(KIM-1)和中性粒细胞明胶酶相关载脂蛋白(NGAL)进行测定,评估蝉棒束孢菌子实体对肾小管上皮细胞的影响;研究不同剂量蝉棒束孢菌子实体(0.25g/kg、0.5g/kg、1.0g/kg)对肾小管上皮细胞增殖和增生能力的影响。给药30、60、90d及恢复28d时,SD大鼠血清中KIM-1浓度与对照组相比均无显著差异(P>0.05),给药30d、60d时,SD大鼠血清中NGAL浓度与对照组相比均无显著差异(P>0.05),给药90d及恢复28d时,SD大鼠血清中NGAL浓度低于对照组(P<0.05),且给药90d组与对照组相比有显著性差异(P<0.01);免疫组化检测增殖细胞核抗原法(PCNA)及四甲基偶氮唑盐微量酶反应比色法(MTT)表明:与对照组相比,蝉棒束孢菌子实体能使肾小管上皮细胞增生能力增强,未导致肾小管上皮细胞凋亡。 相似文献

目的:探讨解剖型锁定钢板治疗Pilon骨折的临床疗效和安全性。方法:选择2009年1月~2012年12月采用解剖型锁定钢板内固定治疗30例Pilon骨折患者为研究对象,所有患者在无明显手术禁忌情况下,均行开放复位钢板内固定术。术后采用Mazur评分系统评价手术效果。结果:本组全部患者均获得随访,骨折平均愈合时间为4.1个月;参照Mazur踝关节功能评分系统:优14例,良13例,可2例,差1例,优良率90%。仅1例开放性Pilon骨折患者发生皮肤感染坏死,局部缺损,经清创、VSD覆盖负压吸引,待新鲜肉芽组织形成后,行带蒂皮瓣移植术,愈合良好。结论:解剖型锁定钢板的解剖形态好,成角稳定,用于治疗Pilon骨折能够恢复关节面的完整性、稳定性,减少并发症的发生,临床效果好,安全性高。 相似文献

The plant hormone abscisic acid (ABA) has been suggested to play a role in fruit development, but supporting genetic evidence has been lacking. Here, we report that ABA promotes strawberry (Fragaria ananassa) fruit ripening. Using a newly established Tobacco rattle virus-induced gene silencing technique in strawberry fruit, the expression of a 9-cis-epoxycarotenoid dioxygenase gene (FaNCED1), which is key to ABA biosynthesis, was down-regulated, resulting in a significant decrease in ABA levels and uncolored fruits. Interestingly, a similar uncolored phenotype was observed in the transgenic RNA interference (RNAi) fruits, in which the expression of a putative ABA receptor gene encoding the magnesium chelatase H subunit (FaCHLH/ABAR) was down-regulated by virus-induced gene silencing. More importantly, the uncolored phenotype of the FaNCED1-down-regulated RNAi fruits could be rescued by exogenous ABA, but the ABA treatment could not reverse the uncolored phenotype of the FaCHLH/ABAR-down-regulated RNAi fruits. We observed that down-regulation of the FaCHLH/ABAR gene in the RNAi fruit altered both ABA levels and sugar content as well as a set of ABA- and/or sugar-responsive genes. Additionally, we showed that exogenous sugars, particularly sucrose, can significantly promote ripening while stimulating ABA accumulation. These data provide evidence that ABA is a signal molecule that promotes strawberry ripening and that the putative ABA receptor, FaCHLH/ABAR, is a positive regulator of ripening in response to ABA. 相似文献

无花果蛋白酶通过8%戊二醛活化载体,共价结合到聚苯乙烯阴离子交换树脂GM201上,固定化作用在pH7.7,酶浓度0.8mg/g树脂,4℃下进行6h。得到的固定化酶表观K_m值(酪蛋白,1.11×10~(-4)mol/L)小于溶液酶K_m值(1.96×10~(-4)mol/L);固定化酶活性在pH6~8保持稳定,溶液酶最适pH为7.2;固定化酶最适温度由溶液酶的50~60℃移至37℃;固定化酶25℃保持7d,重复水解酪蛋白7次后,保留83.3%活性。固定化酶对酪蛋白水解度达47.5%,对大豆球蛋白达11.6%。 相似文献
