We monitored the radioligand-binding characteristics of thyrotropin-releasing hormone (TRH) receptors, functional activity of Gq/11α proteins, and functional status of the whole signaling cascade in HEK293 expressing high levels of TRH receptors and G11α. Our analyses indicated that disruption of plasma membrane microdomains by cholesterol depletion did not markedly influence the binding parameters of TRH receptors, but it altered efficacy of signal transduction. The functional coupling between TRH receptor and Gq/11α was assessed by agonist-stimulated [35S]GTPγS binding, and results of these measurements pointed out to significantly lower potency of TRH to mediate G protein activation in the plasma membrane fraction isolated from cholesterol-depleted cells; there was a shift in sensitivity by one order of magnitude to the higher concentrations. A markedly lower sensitivity to stimulation with TRH was also observed in our experiments dealing with determination of hormone-induced Ca2+ response. These data suggest that the intact structure of plasma membranes is an important optimum signal transduction initiated by TRH receptors and mediated by Gq/11α proteins.  相似文献

Aphidophagous and coccidophagous ladybirds, similar to their prey, show marked differences in their pace of life (Dixon, 2000), in particular in their rate of development, with all stages of aphidophagous species developing much faster than those of coccidophagous species. Two hypotheses are proposed to account for the large difference in the pace of life of these two groups. These are that differences in the rate of development are a result of differences in lower temperature thresholds for development or the quality of their respective prey as food (Dixon et al., 2011). Analysis of published results on the rates of development of the eggs of ladybirds indicates that the inverse relationships between the number of day‐degrees required for development (K) and the lower temperature threshold for development (tdmin) of these two groups are significantly different. In particular, the respective tdmin overlap and K of the aphidophagous and coccidophagous species with a similar tdmin are, on average, 38 and 117 day‐degrees (Do). The relationship between the rate of development (R) and temperature (T) for aphids reared on poor‐ or high‐quality foods indicates that, although the value of tdmin of a species depends on food quality, K does not, showing that it is unlikely that K is governed by food quality. Thus, there is little support for differences in either the tdmin or food quality governing the difference in the pace of life of these two groups of ladybirds. The results indicate that the physiological mechanism that may govern the difference in the pace of life between these two groups is the number of day‐degrees (K) needed to complete their development. The possible evolutionary reason for this is discussed.  相似文献

1. Gall‐forming insects are a guild of endophages that exhibit a high level of fidelity to their host plants, however, their level of host specificity is seldom explicitly tested. 2. Gall‐forming insect taxa from 32 species of woody tropical plants with resolved phylogenetic relationships were collected and reared, representing 15 families from all the major clades of angiosperms, at three lowland rainforest locations in Madang, Papua New Guinea (PNG). 3. More than 8800 galled plant parts were collected from 78 gall morphospecies at an average of 2.4 per host plant. Total species richness at the sampling sites was estimated to be 83–89. All but one morphospecies were monophagous resulting in an effective specialisation of 0.98. 4. Specific leaf weight, foliar nitrogen, the presence of latex, and the successional preference of plant species all gave a phylogenetic signal, but only plant successional preference influenced the species richness of galls on analysis of phylogenetically independent contrasts. Gall species were distributed randomly among host plant species and showed no preference for any particular plant lineage. Furthermore, most gall‐forming taxa were evenly dispersed across the host plant phylogeny. 5. In the tropical rainforests of New Guinea, gall‐forming insects are ubiquitous but occur in species‐poor assemblages. Local species richness is closely tied to the diversity of angiosperms owing to very high host specificity. 6. Finally, galler species richness data from the literature across habitats and latitudes were compared and suggest that tropical rainforests may be richer in galls than previously acknowledged.  相似文献

1. Different groups of specialised herbivores often exhibit highly variable responses to host plant traits and phylogeny. Gall‐forming insects and mites on willows are highly adapted to their hosts and represent one of the richest communities of gallers associated with a single genus of host plants. 2. The present study evaluated the effects of host plant secondary metabolites (salicylates, flavonoids, condensed tannins), physical traits (trichome density), nutrient content (N:C) and phylogeny on the abundance and richness of gall‐forming arthropods associated with eight willow species and Populus tremula. 3. Galler abundance was affected by N:C rather than by willow defensive traits or phylogeny, suggesting that gallers respond differently to host plant traits than to less specialised guilds, such as leaf‐chewing insects. None of the studied defensive traits had a significant effect on gall abundance. Gall morphospecies richness was correlated with the host phylogeny, mainly with the nodes representing the inner division of the willow subgenus Vetrix. This suggests that the radiation of some willow taxa could have been important for the speciation of gallers associated with willows. 4. In conclusion, it is shown that whereas willow traits, such as nutrient content, appeared to affect abundances of gallers, it is probably willow radiation that drives galler speciation.  相似文献

Insect herbivores were sampled from the foliage of 15 species of Ficus (Moraceae) in rainforest and coastal habitats in the Madang area, Papua New Guinea. The collection included 13 193 individuals representing 349 species of leaf-chewing insects and 44 900 individuals representing 430 species of sap-sucking insects. Despite a high sampling intensity, the species accumulation curve did not reach an asymptote. This pattern was attributed to the highly aggregated distribution of insects on individual host trees. The number of insect species collected on a particular Ficus species ranged from 34 to 129 for leaf-chewing and from 51 to 219 for sap-sucking insects. Two Ficus species growing on the seashore sustained less speciose insect communities than their counterparts growing in forest. For the forest figs, significant predictors of insect species richness included leaf palatability and leaf production for leaf-chewing insects (40% of the variance explained), and tree density and leaf expansion for sap-sucking insects (75%). The high faunal overlap among Ficus communities and the importance of local resources for insect herbivores suggest that highly specialized interactions between insect herbivores and Ficus in Papua New Guinea have not been conserved in evolutionary time. This is at variance with the dogma of old, extremely specialized and conservative interactions between insect herbivores and their hosts, providing numerous ecological niches in the floristically rich tropics.  相似文献

Abstract.  1. Bark and ambrosia beetles are crucial for woody biomass decomposition in tropical forests worldwide. Despite that, quantitative data on their host specificity are scarce.
2. Bark and ambrosia beetles (Scolytinae and Platypodinae) were reared from 13 species of tropical trees representing 11 families from all major lineages of dicotyledonous plants. Standardised samples of beetle-infested twigs, branches, trunks, and roots were taken from three individuals of each tree species growing in a lowland tropical rainforest in Papua New Guinea.
3. A total of 81 742 beetles from 74 species were reared, 67 of them identified. Local species richness of bark and ambrosia beetles was estimated at 80–92 species.
4. Ambrosia beetles were broad generalists as 95% of species did not show any preference for a particular host species or clade. Similarity of ambrosia beetle communities from different tree species was not correlated with phylogenetic distances between tree species. Similarity of ambrosia beetle communities from individual conspecific trees was not higher than that from heterospecific trees and different parts of the trees hosted similar ambrosia beetle communities, as only a few species preferred particular tree parts.
5. In contrast, phloeophagous bark beetles showed strict specificity to host plant genus or family. However, this guild was poor in species (12 species) and restricted to only three plant families (Moraceae, Myristicaceae, Sapindaceae).
6. Local diversity of both bark and ambrosia beetles is not driven by the local diversity of trees in tropical forests, since ambrosia beetles display no host specificity and bark beetles are species poor and restricted to a few plant families.  相似文献

SYNOPSIS A new species of the family Colpodidae, genus Colpoda O. F. Müller, Colpoda spiralis sp. n. is described. This organism has been found only in treehole fluid. Its species status is based on the possession of a unique and prominent preoral protuberance, multimicronuclearity, and the preference for a treehole habitat. Its range is known to include the tri-state area of Arizona, New Mexico and Utah, and the mixed-deciduous forests of Maryland. This species prefers treehole fluids with a pH > 7 and is naturally limited to tree species which possess these alkaline treeholes. It encysts as a treehole dries and excysts when the treehole is again filled by fluids from the tree, or rainwater.  相似文献
