To identify genes involved in trifluralin herbicide-induced resistance of melon to Fusarium oxysporum f. sp. melonis , suppression subtractive hybridization (SSH) and cDNA-amplified fragment-length polymorphism (cDNA-AFLP) were used. A total of 123 clones—60 of which have never been isolated from melon—were isolated, sequenced and annotated. A significant proportion (35%) of the total 123 clones exhibited similarity to genes that have been formerly described as stress- or defence-related. Thirty-two selected clones were subjected to a detailed expression analysis, one-third of which were found to be up-regulated in response to trifluralin treatment and/or fusarium inoculation. The putative roles of seven of these clones in stress are discussed. Furthermore, the expression of four stress-related and up-regulated genes was enhanced when the plants were subjected to salinity stress, suggesting that trifluralin induces a general stress response which protects the plant against fusarium wilt.  相似文献

Treatment of mitochondria isolated from Texas male sterile cytoplasmcorn (T mitochondria) with high concentrations of dicyclohexylcarbodiimide(DCCD) (140 nmol DCCD mg–1 mitochondrial protein) completelyand immediately inhibited T mitochondrial swelling by Helminthosporiummaydis Race T toxin (HmT toxin). In order to obtain a specificinteraction between DCCD and the ATPase complex T mitochondriawere incubated with lower DCCD concentrations (1–5 nmolDCCD mg–1 mitochondrial protein) for up to 8 h at 4 °C.After 8 h incubation in the presence of 3.75 nmol DCCD mg–1mitochondrial protein, toxin-induced swelling was decreasedby 69%. Specificity of DCCD action upon the ATPase complex wasconfirmed by (1) SDS gel electrophoresis and fluorographic analysesof proteins from [14C]-DCCD-treated T mitochondria and immunoprecipitatesand (2) physiological experiments showing that DCCD exertednone of its other documented effects. These data suggest thatHmT toxin interacts with the ATPase complex of T mitochondriaeither at or near the DCCD-binding protein within the membranesector of the complex. The sandbar shark, Carcharhinus plumbeus, is a large, cosmopolitan, coastal species. Females are thought to show philopatry to nursery grounds while males potentially migrate long distances, creating an opportunity for male‐mediated gene flow that may lead to discordance in patterns revealed by mitochondrial DNA (mtDNA) and nuclear markers. While this dynamic has been investigated in elasmobranchs over small spatial scales, it has not been examined at a global level. We examined patterns of historical phylogeography and contemporary gene flow by genotyping 329 individuals from nine locations throughout the species’ range at eight nuclear microsatellite markers and sequencing the complete mtDNA control region. Pairwise comparisons often resulted in fixation indices and divergence estimates of greater magnitude using mtDNA sequence data than microsatellite data. In addition, multiple methods of estimation suggested fewer populations based on microsatellite loci than on mtDNA sequence data. Coalescent analyses suggest divergence and restricted migration among Hawaii, Taiwan, eastern and western Australia using mtDNA sequence data and no divergence and high migration rates, between Taiwan and both Australian sites using microsatellite data. Evidence of secondary contact was detected between several localities and appears to be discreet in time rather than continuous. Collectively, these data suggest complex spatial/temporal relationships between shark populations that may feature pulses of female dispersal and more continuous male‐mediated gene flow.  相似文献

Perturbance Analysis of Nuclear Determination in Tetrahymena   总被引:1,自引:0,他引:1  
Mating type frequencies were ascertained among the progeny of crosses of strains A × B, Tetrahymena thermophila under a number of different circumstances. The frequencies are different if the parents are severely starved than if they are well-fed at the time of conjugation; severe starvation of the progeny before the first post-zygotic division has an effect similar to that of starving the parents. Mating type frequencies may also be modified by isolating conjugating pairs into cell extracts before the new macronuclei begin to develop; the changes do not appear to be related in a meaningful way to the mating type of the cells used as a source of the cell extracts. A third means of changing the mating type frequencies involves the exposure of conjugating pairs to CaCl2 solutions. Finally, changed frequency patterns may appear "spontaneously", and reflect either some as yet unsuspected environmental variable, or else an intrinsic metastable state that conditions the probabilities of mating type fixation. With the exception of the starvation effects, the pattern variations seem to fall into two groups. No satisfactory mechanism to account for these results is yet available.  相似文献

Nuclear import and export are crucial processes for any eukaryotic cell, as they govern substrate exchange between the nucleus and the cytoplasm. Proteins involved in the nuclear transport network are generally conserved among eukaryotes, from yeast and fungi to animals and plants. Various pathogens, including some plant viruses, need to enter the host nucleus to gain access to its replication machinery or to integrate their DNA into the host genome; the newly replicated viral genomes then need to exit the nucleus to spread between host cells. To gain the ability to enter and exit the nucleus, these pathogens encode proteins that recognize cellular nuclear transport receptors and utilize the host's nuclear import and export pathways. Here, we review and discuss our current knowledge about the molecular mechanisms by which plant viruses find their way into and out of the host cell nucleus.  相似文献

New specimens of Acomys spinosissimus from northern Malawi and southern Tanzania were subjected to morphological, molecular (cytochrome b mitochondrial gene) and cytogenetical analysis in order to establish the phylogenetic position of these populations among A. spinosissimus, as well as their relationships with other species (A. subspinosus and A. wilsoni). The monophyly of the spinosissimus clade is supported by morphological and molecular phylogenies. Cytogenetics and morphology, but not the cytochrome b phylogeny show the possible existence of two sibling species within spinosissimus. Incongruences are noted between molecular and morphological trees, especially concerning the position of the Transvaal specimens. Two different biogeographic scenarios for the origin and dispersal of A. spinosissimus are discussed in the light of available palaeontological data.  相似文献

Phenolics are aromatic benzene ring compounds with one or more hydroxyl groups produced by plants mainly for protection against stress. The functions of phenolic compounds in plant physiology and interactions with biotic and abiotic environments are difficult to overestimate. Phenolics play important roles in plant development, particularly in lignin and pigment biosynthesis. They also provide structural integrity and scaffolding support to plants. Importantly, phenolic phytoalexins, secreted by wounded or otherwise perturbed plants, repel or kill many microorganisms, and some pathogens can counteract or nullify these defences or even subvert them to their own advantage. In this review, we discuss the roles of phenolics in the interactions of plants with Agrobacterium and Rhizobium.  相似文献

Effects of historic tidal restrictions on salt marsh sediment chemistry   总被引:1,自引:0,他引:1  
The effects of tidalrestrictions by diking on salt marshbiogeochemistry were interpreted by comparingthe hydrology, porewater chemistry and solidphase composition of both seasonally floodedand drained diked marshes with adjacentnatural salt marshes on Cape Cod,Massachusetts. Flooding periods weregreatest in natural and least in drainedmarshes.Differences between the chemistry of thenatural and diked marshes depended upon thedepth of the water table and the supply ofsulfate for anaerobic metabolism. Drainedmarsh sediments were highly acidic (pH <4)with porewaters rich in dissolved Fe; thenatural and diked flooded marshes had pH 6–7.5and Fe orders of magnitude lower. Porewater nutrients, sulfides and alkalinitywere much lower in both flooded and draineddiked marshes than in the natural marsh.Sediments of the drained marsh had subsided90 cm relative to the natural site due toorganic matter decomposition and compaction. However, despite the loss of organic matter,much of the P and N was retained, withNH4 likely protected from nitrificationby low pH and PO4 adsorbed on Fe and Aloxides. Iron, and to a lesser degree sulfur,had also been well retained by the sediment. Despite eight decades of diking, substantialamounts of reduced S, representing potentialacidity, persisted near the top of the watertable.In contrast, the surface of the seasonallyflooded marsh was only 15 cm below thenatural marsh. Accretion since dikingamounted to 25 cm and involved proportionallyless mineral matter.The restoration of seawater flow to bothseasonally flooded and drained diked marsheswill likely extend flooding depth andduration, lower redox, increase cationexchange, and thereby increase NH4,Fe(II), and PO4 mobilization. Increasedporewater nutrients could benefitrecolonizing halophytes but may also degradesurface water quality.  相似文献
