SYNOPSIS. The biochemical effects of some detergents on the ATPase activity of isolated flagella from Euglena gracilis are related to morphologic obliterations induced by those detergents. Enzymic activity can be localized by electron microscopy along the microtubules and also on the paraflagellar rod. The nonionic detergent digitonin solubilizes the enzyme linked to dyneinic arms, whereas the activity linked to residual structures appears enhanced. These results support the hypothesis that the paraflagellar rod may be a structure actively related to the motility of this type of flagellum.  相似文献

SYNOPSIS. The ATPase activity of isolated flagella was studied in Euglena gracilis strain Z in the presence of Mg++ or Ca++. With Mg++, the optimum activity was at pH 7 and with Ca++, at pH 9. The K m values were respectively 6.6 × 10−4 and 3.6 × 10−4. Activity was influenced also by temperature and ionic strength. Results with inhibitors of membrane ATPase suggest the presence of a specific contractile system in the flagella. Our results are compatible with a multicomponent enzymic system containing 2 active ATPases.  相似文献

Abstract Dissociation of active H+ extrusion (?ΔH+) from K+ uptake in pea and maize root segments was attempted by substituting K+ in the incubation medium with lipophilic cations assumed to enter the cell by passive, non-specific, permeation through the lipid component of the plasmalemma. Among the compounds tested, tributylbenzylammonium significantly stimulated ?ΔH+ in the absence of other monovalent cations in the medium. This effect was much more evident when the experiment was carried out in the presence of fusicoccin, which strongly stimulates proton extrusion and monovalent cation uptake, and hyperpolarizes the trans-membrane electric potential in these materials. Also the lipophilic cations tetraphenylphosphonium, dimethyldibenzylammonium and hexylguanidine markedly stimulated FC-promoted ?ΔH+. Octylguanidine at a low concentration induced an early stimulation followed by a strong inhibition of ?ΔH+. A complete lack of additivity was observed between the effects of lipophilic cations and that of K+ on H+ extrusion. Lipophilic cations severely inhibited K+ uptake. These data are interpreted as supporting the view of an electric, rather than a chemical, (namely, involving the same carrier system) nature of the coupling of active H+ extrusion with K+ influx.  相似文献

Mark–release–recapture experiments with Anopheles gambiae s.l. were performed during the wet seasons of 1993 and 1994 in Banambani, Mali. All recaptured mosquitoes were identified to species by PCR analysis and, when possible, by chromosomal analysis to chromosomal form. Two species of the An. gambiae complex were present: An. gambiae s.s. and An. arabiensis ; their ratio differed greatly from one year to the next. Three chromosomal forms of An. gambiae s.s. were found – Bamako, Savanna and Mopti. The drier 1993 was characterized by a high frequency of An. arabiensis and of the Mopti chromosomal forms of An. gambiae s.s. These trends were consistent with large-scale geographical patterns of abundance along a precipitation gradient. We observed no difference in dispersal between the two species, nor among the chromosomal forms of An. gambiae s.s. Therefore, in this situation at least, it is reasonable to group such data on the An. gambiae complex as a whole for analysis. Population size of An. gambiae s.l. females in the village was estimated to be 9000–11 000 in 1993 and 28 000 in 1994. The corresponding numbers were somewhat higher when independently-derived values of daily survival were used. These were consistent with estimates of effective population size obtained from patterns of gene frequency change.  相似文献

Arbuscular mycorrhizas are a widespread symbiosis between soil fungi and plant roots. Flow cytometry, after DNase I partial digestion and DAPI staining, and light and electron microscopy were used to analyse chromatin condensation and nuclear conditions in mycorrhizal and control roots of Allium porrum . The 2C peak, detected by flow cytometry, split into two peaks representing two populations of nuclei, one more resistant and one more susceptible to the enzyme action. The microscopic analyses showed the presence of pyknotic and chromatolytic nuclei, two typical features of senescence. In order to quantify the senescing process, a terminal deoxynucleotidyl transferase assay was performed on extracted nuclei, later analysed by flow cytometry. The numbers of senescing nuclei and their DNA cleavage were higher in control plants. Our results show the existence of senescing nuclei in cortical cells of the bulbous monocotyledon A. porrum and the delaying effect of arbuscular mycorrhizas on senescence.  相似文献

Inbreeding is expected to increase the variability in size and shape within populations. The distinct effects of inbreeding on size and shape suggest that they are governed by different developmental pathways. One unresolved question is whether the non‐allometric shape component is partially unconstrained developmentally and therefore whether shape is evolvable. In the present study, we utilized a mass outbred population of Drosophila melanogaster maintained at standard laboratory conditions. Eight lines with equivalent expected levels of inbreeding (F ≈ 0.67) were obtained by restricting the size of each population to two pairs for nine generations. Nine landmarks were measured on Drosophila wings of the inbreed lines and compared with those of the mass population. Wing landmarks comprise an excellent model system for studying evolution of size and shape. Landmark measurements were analyzed with a Procrustes generalized least squares procedure. To visualize global shape changes among samples, we reconstructed the mean shape and the shape changes related to both the allometric and non‐allometric components. An increased variability in the non‐allometric shape component was found with inbreeding. This indicated that shape was not entirely developmentally constrained, and therefore that shape appears to be evolvable. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 102 , 626–634.  相似文献
