In the course of study on the mechanism of the tartaric acid formation from 5-ketogluconic acid, a new intermediary substance with mauve color to Abdel-Akhel and Smith’s reagent was isolated from intact cell culture liquid. The chemical structure of this substance was determined as 1,2-dihydroxyethyl hydrogen L(+) tartrate from the results of hydrolysis experiments and from the identifications of the constituents of the molecule, and named “pretaric acid.” Tartaric acid was evidently produced from pretaric acid by intact cell culture. Clearly, then, pretaric acid appears to be an intermediate in the formation of tartaric acid from 5-ketogluconic acid. The authors assumed that in the formation of pretaric acid from 5-ketogluconic acid, a Baeyer-Villiger type oxidation occurred.  相似文献

Changes of the resting potential of Valonia cell in sea wateragainst a 10-fold increase of the external concentrations ofK$, Na$ and Cl were 1±1, 6.2±0.1 and 38.9±4mV, respectively. The potassium conductance was smaller than7 µ/cm2, while the Na and Cl conductances were 45 and281 µ/cm2, respectively, in normal sea water. The positivevacuolar potential could be explained by these ionic conductances.On the other hand, the membrane became more sensitive to K$,if the cell was incubated for about 30 min in K-rich (100 mM)sea water. Summary The current-voltage curve (I–V curve) of theChara membrane was obtained by applying a slow ramp hyper- and depolarization by use of voltage clamp. By inhibiting the electrogenic pump with 50m DCCD (dicyclohexylcarbodiimide), theI–V curve approached a steadyI–V curve within two hours, which gave thei d -V curve of the passive diffusion channel. Thei p -V curve of the electrogenic pump channel was obtained by subtracting the latter from the former. The sigmoidali p -V curve could be simulated satisfactorily with a simple reaction kinetic model which assumes a stoichiometric ratio of 2. The emf of the pump (E p ) is given as the voltage at which the pump current changes its sign. The conductance of the pump (g p ) can be calculated as the chord conductance from thei p -V curve, which is highly voltage dependent having a peak at a definite voltage. The changes of emf and conductance during excitation were determined by use of the current clamp (I=0). Since theE p andg p (V) are known, the changes, during excitation, of emf (E d ) and conductance (g d ) of the passive diffusion channel can be calculated. The marked increase of the membrane conductance and the large depolarization during the action potential are caused by the marked increase of the conductance of the passive diffusion channel and the large depolarization of its emf. The conductance of the electrogenic pump decreases to about half at the peak of action potential, while the pump current increases almost to a saturated level.  相似文献

We examined a role of Ca2+ in the activation of the two majorion channels, i.e., Cl and K+ channels at the excitationof the characean plasmalemma. The current-voltage relation (I-Vcurve) of the Chara membrane was compared under the ramp voltageclamp condition before and after external application of 20µMof La3+ (a Ca2+ channel blocker). The transient inward currentcomponent, which is carried mainly by the efflux of Cl,disappeared almost completely in about 30 min with La3+ treatment.On the other hand, no effect was observed on the late largeoutward current, which is mainly carried by the efflux of K+in a large depolarization region (less negative than –50mV). These results suggest that the Cl channel in theChara plasmalemma is activated by Ca2+ influx, while the K+channel is simply activated by depolarization. Effects of hydrodynamic shear on the shape of the endothelial surface are examined based on evaluations of the rate of entropy generation at the cell surface. A linear solution of the flow over a sinusoidally varying endothelial surface is used to evaluate the entropy generation rate on the cell surface for which measured cell dimensions are available. Both the local rate of entropy generation (equivalent to the rate of energy dissipation by viscous shear) at the peak of a cell and the total entropy generation rate over the cell surface are minimized under conditions of a constant cell surface area and a constant cell peak height; which yields horizontal cell dimensions that are close to those obtained experimentally.  相似文献

The membrane potential of Nitella internode shows a spontaneousrhythmicity. The period depends largely upon temperature andthe length of the cell. Moreover, the period is reversibly prolongedat low temperature and also by treatment with cytochalasin B.We concluded that this bioelectrical rhythm is closely relatedto cytoplasmic streaming. The purpose of the present investigation is to obtain the superior mutants from the tartrate producing strain, Gluconobacter suboxydans 2026Y2 previously isolated from nature. Some mutant strains obtained by treatment with N-methyl-N′-nitro-N-nitrosoguanidine were found to accumulate L(+) tartaric acid in culture broth with much higher yield than in the case of the wild strain.

The high tartrate productivity of the mutants was followed by the low accumulation of 2-ketogluconic acid. The mutants having high assimilability of 5-ketogluconate showed high tartrate productivity.

The culture conditions for tartaric acid production by a mutant, Gl. suboxydans N-3874, were investigated. As a result, the amount of tartaric acid accumulated in culture broth reached to a level of 14.6g/liter in the medium containing 5% glucose and 0.3% corn steep liquor.  相似文献

Summary The steady N shapeI/V curves were obtained by applying slow ramp hyper- and depolarization pulses toChara cells under the voltage-clamp condition. Application of calcium channel blocker, 20 m La3+, to theChara membrane caused, in about 30 min, a marked reduction of the transient inward current and later almost complete blocking of the pump current, while the steady outward current remained almost unaffected. Removal of external Ca2+ with 0.5mm EGTA caused similar results. Application of calmodulin antagonists, 10 m TFP or 20 m W-7, also gave very similar results, i.e., the decrease of the transient inward current and of H+-pump activity. These results suggest that not only the excitatory mechanisms but also the H+-pump activity ofChara membrane are regulated by calmodulin within a comparatively narrow range of internal Ca2+ level.  相似文献

Summary The sigmoidal current-voltage curve (i p -V curve) of the electrogenic H+-pump of theChara membrane was simulated satisfactorily with a simple reaction kinetic model which assumed consecutive changes in state of H+-ATPase. Four rate constants, i.e., forward and backward ones in voltage-dependent and-independent steps could be evaluated from the data. The emf of the pump (E p ), the voltage at which the pump current changes its sign, varies only slightly with temperature. However, the pump current (i p ) is highly temperature dependent, and there-fore the conductance (g p ) of the pump, calculated as the chord conductance from thei p-V curve, is also highly voltage dependent having a peak at a level somewhat less negative than the resting potential. In contrast tog p , the conductance (i p ) of the passive channel does not change appreciably with temperature. Arrhenius plots ofg p and also of the rate constants showed a clear bend at about 19°C. Great temperature dependence of the kinetic parameters offers useful information on the pumping mechanism of theChara membrane.  相似文献
