In the present report a characteristic pattern showed by cytoplasmic filaments (intermediate-sized and actin-like) in the perinuclear area of a freshwater teleost (Pimelodus maculatus) endothelial cells is described for the first time. Thus, many intermediate-sized filaments are directly inserted in the nuclear envelope, but others are connected to one another and to the nucleus through microfilaments. It is suggested that these particular relationships between the nucleus and cytoplasmic filaments are responsible not only for nuclear anchorage, but also for nuclear movements  相似文献

Fusion of lipid bilayers proceeds via a sequence of distinct structural transformations. Its early stage involves a localized, hemifused intermediate in which the proximal but not yet the distal monolayers are connected. Whereas the so-called stalk model most successfully accounts for the properties of the hemifused intermediate, there is still uncertainty about its microscopic structure and energy. We reanalyze fusion stalks using the theory of membrane elasticity. In our calculations, a short (cylindrical micelle-like) tether connects the two proximal monolayers of the hemifused membranes. The shape of the stalk and the length of the tether are calculated such as to minimize the overall free energy and to avoid the formation of voids within the hydrocarbon core. Our free energy expression is based on three internal degrees of freedom of a perturbed lipid layer: thickness, splay, and tilt deformations. Based on exactly the same model, we compare fusion stalks with and without the ability included to form sharp edges at the interfacial region between the hydrocarbon core and the polar environment. Requiring the interface to be smooth everywhere, our detailed calculations recover previous results: the stalk energies are far too high to account for the experimental observation of fusion intermediates. However, if we allow the interface to be nonsmooth, we find a remarkable reduction of the stalk free energy down to more realistic values. The corresponding structure of a nonsmooth stalk exhibits sharp edges at the transition regions between the bilayer and tether parts. In addition to that, a corner is formed at each of the two distal monolayers. We discuss the mechanism how membrane edges reduce the energy of fusion stalks.  相似文献

Abstract 1. Desiccation of needles following oviposition by Neodiprion lecontei in Pinus resinosa caused high egg mortality. Eight-five per cent of pine needles into which sawflies oviposited subsequently desiccated, compared with 2.5% of needles without eggs. No larval emergence occurred from desiccated needles.
2. Within affected shoots, the probability of desiccation increased with the extent of oviposition. In paired comparisons of needles, 41.2% of needle length was occupied by eggs in desiccated needles, compared with 24.0% of needles without desiccation.
3. Both density-dependent and density-independent factors may contribute to desiccation of egg-laden needles. The likelihood that needle vasculature will be severed by ovipositing females probably increases with population density. Drought, which was high during the observations, probably increases the incidence of needle desiccation following mechanical injury caused by oviposition.  相似文献

In this work we investigate the level of theory necessary for reproducing the non-linear variation of the 129Xe nuclear magnetic resonance (NMR) chemical shift with the density of Xe in supercritical conditions. In detail we study how the 129Xe chemical shift depends under supercritical conditions on electron correlation, relativistic and many-body effects. The latter are included using a sequential-QM/MM methodology, in which a classical MD simulation is performed first and the chemical shift is then obtained as an average of quantum calculations of 250 MD snapshots conformations carried out for Xe n clusters (n =?2 ? 8 depending on the density). The analysis of the relativistic effects is made at the level of 4-component Hartree-Fock calculations (4c-HF) and electron correlation effects are considered using second order Møller-Plesset perturbation theory (MP2). To simplify the calculations of the relativistic and electron correlation effects we adopted an additive scheme, where the calculations on the Xe n clusters are carried out at the non-relativistic Hartree-Fock (HF) level, while electron correlation and relativistic corrections are added for all the pairs of Xe atoms in the clusters. Using this approach we obtain very good agreement with the experimental data, showing that the chemical shift of 129Xe in supercritical conditions is very well described by cluster calculations at the HF level, with small contributions from relativistic and electron correlation effects.  相似文献

Seed re-dispersal following initial harvesting by ants may have important implications for the distribution and fate of myrmecochorous seeds. However, the probability of seed re-dispersed by ants and the effect it may have on subsequent survival appear variable, the functional role of diaspore, disperser and seed predator to the fate of discarded seeds remain unclear. To clarify the ecology, we compared the consequences of seed re-dispersal by a keystone seed-dispersing ant (Myrmica ruginodis Nylander) for four sympatric myrmecochorous plants common to the temperate deciduous forests in Qinling Mountains, central China. Plants varied in the probability of re-dispersal and in elaiosome condition. Ants preferred seeds with residual elaiosomes, while rodents only consumed the two larger-seeded species, regardless of the elaiosome presence. The scattered distribution of discarded seeds increased the probability of ant re-harvesting and, to some extent, reduced rodent predation. Thus, difference in the probability of seed re-dispersal and its subsequent effect on seed fate in relation to ants and rodents was attributed primarily to the elaiosome condition, seed size and seed spatial pattern. The results imply that seed re-dispersal could affect the fitness of plants and ultimately influence the plant abundance and distribution pattern. This highlights the necessity to incorporate re-dispersal into myrmecochory to advance our understanding of the benefits of myrmecochory to plants.  相似文献

Abstract Mice were injected with immune complexes containing the recombinant hepatitis B surface antigen (HBsAg) vaccine (S + preS2) bound to different monoclonal antibodies (mAbs), in order to determine whether an enhancement of the response to a human vaccine could be obtained and observed. Enhancement and indifference were observed, as well as a decrease in immunogenicity. N relationship could be established between any effect and affinity or isotypy of the bound mAbs. The preS2 region was rendered more immunogenic when an IgG2a mAb was bound to the S region of the HBsAg. The response to the S region was not modulated, whereas immunogenicity of the preS2 collinear region was decreased by antibody shielding. The mAb which was the most efficient as an enhancer of the antibody response also increased binding of the complexed immunogen to antigen presenting cells. The binding of a human mAb to the sole S region, but not to the preS2 region, should be tested as a potentiating agent of the anti-preS2 human immune response.  相似文献
