Blossom blight caused by Botrytis cinerea is one among the most devastating diseases that cause complete post-harvest loss in flower crops. The present study focuses on the development of effective bioformulation towards suppression of blossom blight and plant growth promotion in rose. Bacillus amyloliquefaciens (VB2) and Bacillus subtilis (AP) effectively inhibited mycelial growth of B. cinerea in vitro. Genome screening of VB2 and AP revealed the presence of antimicrobial peptide genes including, ituD, ipa14, bacA, bacD, srfA, sfP, spaC, spaS responsible for the biosynthesis of antibiotics such as iturin, bacilysin, bacillomycin, surfactin and subtilin. Further, the presence of volatile antifungal compounds in the bacterial secretome was identified through gas chromatography–mass spectrometry (GC/MS) analysis. Upon treatment, AP accelerated the metabolite profile of the plants and a rise in peak area of antifungal compounds such as, pentadecanoic acid, n-hexadecanoic acid, octadecanoic acid (stearic acid) and tetradecanoic acid was observed. In vitro, VB2 produced maximum indole acetic acid (9.17 µg/ml) and gibberellic acid (8.20 µg/ml) in nutrient broth. Under field conditions, foliar spray of VB2 at 0.5% (5 ml/l), four times at weekly interval suppressed blossom blight incidence (64% reduction over control) and also promoted yield. Future research towards development of an effective bioformulation with extended shelf life will aid in the management of various fungal, bacterial and viral diseases in different crop plants.
New phthalimide derivatives were easily prepared through condensation of phthalic anhydride and selected amines with variable yields (70–90%). All compounds (3a–l) were evaluated against Mycobacterium tuberculosis H37Rv using Alamar Blue susceptibility. The compounds 3c, 3i, and 3l have the minimum inhibitory concentrations (MICs) of 3.9, 7.8, and 5.0 μg/mL, respectively, and could be considered new lead compounds in the treatment of tuberculosis and multi-drug resistant tuberculosis. 相似文献
Two species of microsporidia distinguished by the shape of their spores were found to infect cells of various tissues of Sciara ocellaris. Although the infection affects profoundly the development of the infected cells, the interaction between the infective agents and the host cells are often well-balanced and the infected cells may survive longer than the uninfected of the same tissue. The infected cells, including their nuclei and chromosomes, increase greatly. The general reaction of the chromosomes of cells of S. ocellaris infected by microsporidia is the increase of their volume by an increase in the polyteny and the accumulation of chromosomal products between their chromonemata. The cells of the fat bodies, on the other hand, have peculiar types of reactions. Some show an increase in polyteny, frequently showing asynapsis of the entire or parts of the chromosome. Other cells show increased chromosome polyteny associated with different degrees of polyploidy. Still other infected cells develop a new type of chromosome morphology called brachy-polytene which may or may not be associated with polyploidy. Special emphasis must be given to the fact that in Diptera, which have polytene chromosomes, the relationships of the infection and the host cell may in many cases be studied thoroughly, starting with the reaction of the genes to the infective agent. It was shown that in the infected cells of the salivary gland of S. ocellaris the pattern of puffs is greatly changed; hence, this change may be the probable cause of their change in development. Infected cells of the salivary gland with enlarged and active polytene chromosomes were found in adult flies, a situation which never occurs in non-infected animals. We have compared in the same cell type the surface distribution and partition in freeze-fractured plasma membranes of Sindbis virus glycoproteins in three different situations: (i) in permanently transformed cells that express the glycoproteins as the only viral product; (ii) in cells in which prebound viruses were forced to fuse with the plasma membrane by low pH treatment; (iii) in virus-infected cells. We report here that the viral proteins expressed on the surface of transfected cells show a uniform and unclustered distribution; conversely, in Sindbis virus-infected cells they appear clustered, regionally distributed, and always associated with budding viruses (i.e., interacting with the nucleocapsid on the cytosolic side of the membrane). Furthermore, the viral proteins expressed on transfected cells or implanted by low pH-mediated fusion partition during freeze-fracture with the exoplasmic faces of the cell plasma membranes, whereas an opposite partition is observed in infected cells. These results strongly suggest that in infected cells the clustering and the partition with the protoplasmic faces of the plasma membrane depend only on the strong "anchorage" of the glycoproteins to the nucleocapsid. 相似文献
The effect of time delay in specific growth rate () on the periodic operation of bioreactors with input multiplicities is theoretically analyzed for productivity improvement. A periodic rectangular pulse is applied either in feed substrate concentration (Sf) or in dilution rate (D). Periodic operation under feed substrate concentration cycling gives improvement in productivity at lower value of ¯Sf of the two steady-state multiplicities of Sf only when the time delay in is larger. Whereas the larger value of ¯Sf gives improvement in average productivity for all values of time delay. Dilution rate (D) cycling gives an improvement in average productivity particularly for larger time delay in . Treatment of sea urchin eggs for 10 min prior to fertilization with the kinase inhibitor 6DMAP (6-dimethylaminopurine) reversibly inhibits swelling and loss of conical morphology of the male pronucleus. Male pronuclei inhibited with 1 mM 6DMAP for 25 min undergo phosphorylation of Sp H1 and Sp H2B histones as fully as do control nuclei. Therefore, Sp histone kinase, whose target sequences resemble those of the M-phase histone kinase, is not inhibited by 6DMAP, and Sp histone phosphorylation, although it may be necessary, is not sufficient for chromatin decondensation. 相似文献
Hawaiian stilts (Himantopus mexicanus knudseni) are an endangered subspecies of the Black-necked stilt endemic to the Hawaiian Islands. Despite long-term study, the main drivers of Hawaiian stilt population dynamics are poorly understood. We tested for density dependence using two sources of evidence: a 30-year time series of annual estimated range-wide abundance, and two 15+ year time series of reproductive success. Using separate methods with independent data, sources allowed us to make up for the potentially positive bias of one approach with the more conservative nature of the second. We compared nonlinear density-dependent and density-independent population model fits to our time-series data, using both frequentist and Bayesian state-space approaches. Across both approaches, density-dependent models best fit observed population dynamics, with lower AICc and cross-validation statistics compared to density-independent models. Among density-dependent models, a conditional model in which density-independent dynamics occur below a population size threshold (~850–1,000 birds), and then density-dependent dynamics occur above that threshold, performed best across Bayesian and frequentist model comparisons, with the Ricker model ranked next or equivalently. Our analysis of reproduction data revealed a strong negative effect of local adult density on nest success (proportion of nests hatching at least one chick) at Kealia National Wildlife Refuge on Maui, where few alternative breeding habitats are available, but no such effect at another site where many nearby alternative wetlands are available. These congruent results across independent datasets and analytical approaches support the hypothesis that Hawaiian stilts exhibit density dependence across their range. 相似文献
