A rapid, sensitive and accurate high-performance liquid chromatographic method with UV detection was developed and validated for the quantification of gabapentin in human plasma. Gabapentin was quantified using pre-column derivatization with 1-fluoro-2,4-dinitrobenzene following protein precipitation of plasma with acetonitrile. Amlodipine was used as internal standard. The chromatographic separation was carried out on a Nova-Pak C(18) column using a mixture of 50 mM NaH(2)PO(4) (pH=2.5)-acetonitrile (30:70, v/v) as mobile phase with UV detection at 360 nm. The flow rate was set at 1.5 ml/min. The method was linear over the range of 0.05-5 microg/ml of gabapentin in plasma (r(2)>0.999). The within-day and between-day precision values were in the range of 2-5%. The limit of quantification of the method was 0.05 microg/ml. The method was successfully used to study the pharmacokinetics of gabapentin in healthy volunteers.  相似文献

Soil salinity is a complex issue in which various anions and cations contribute to have a general adverse effect on plant growth. In the present study, effects of salinity from various salts including sodium chloride (NaCl), potassium chloride?+?sodium chloride?+?calcium chloride (KCl?+?NaCl?+?CaCl2), potassium sulfate?+?magnesium nitrate (K2SO4?+?Mg(NO3)2) at two electric conductivities (EC) of 2 and 4 dS m?1 of irrigation water, and a distilled water control were evaluated on coriander plants (Coriandrum sativum L.). At EC?=?2, all salts increased plant yield (shoot fresh weight) than control. Most growth traits including plant height, shoot fresh and dry weight, leaf SPAD value and vitamin C, leaf K, Mg and P concentrations were increased by K2SO4?+?MgNO3, and remained unchanged by KCl?+?NaCl?+?CaCl2 treatment (except reduced plant height). Leaf’s zinc concentration reduced by either treatment. Even sodium chloride at EC?=?2 showed some beneficial effects on leaf chlorophyll index, root fresh weight, leaf’s calcium and phosphorus concentration; however, most traits remained unchanged than control. Treatment of plants with NaCl or KCl?+?NaCl?+?CaCl2 at either EC increased the number of flowered shoots and leaf proline content than control. Most growth and quality traits including leaf minerals and vitamin C content were reduced by NaCl at EC?=?4; however, shoot fresh and dry weights remained unchanged than control. Plant root fresh weight increased by NaCl at EC?=?2 and decreased at EC?=?4 than control. At EC?=?4, shoot dry weight was increased and leaf Ca, P, Zn and Mn were decreased by KCl?+?NaCl?+?CaCl2, whereas shoot dry weight, leaf SPAD value and vitamin C content, leaf Mg and P were increased and leaf Zn was decreased by K2SO4?+?MgNO3 than control. The results indicate that in contrast to sodium chloride, the salinity effects of other salts can not be detrimental on coriander plant growth.  相似文献

Very-long-chain acyl-coenzyme A dehydrogenase (VLCAD) deficiency is a newly identified disease. A 105-bp deletion in the VLCAD cDNA in two patients has been reported, and detailed molecular characterization of this disease has remained to be done. We report here five mutations identified in four patients: a 135-bp deletion encompassing bases 343-477, a C-1837-to-T transition (R613W), 3-bp deletions at the nucleotide positions 388-390 (E130del) and 895-897 (K299del), and an A-1144-to-C transversion (K382Q). Sequencing of genomic DNA amplified by PCR revealed a 135-bp deletion caused by exon skipping due to a 1-bp deletion in a 3' splice site of an intron. In cDNA expression experiments using Chinese hamster ovary (CHO) cells, we found that each of the mRNAs derived from E130del and K299del clones were unstable and that translation products from R613W, E130del, K299del, and K382Q clones were labile. Each of R613W, E130del, K299del, and K382Q proteins expressed in CHO cells appeared abnormal in dimer assembly, as shown in gel-filtration analysis. VLCAD activity was not detected in mutants' transfectants. Thus, we verified that all five mutations identified in these four patients were disease-causing alterations.  相似文献

Factor XIII (XIII) is a heterotetramer consisting of two catalytic A subunits (XIIIA) and two noncatalytic B subunits (XIIIB). We examined the molecular mechanisms of a Y283C mutation which had previously been identified in a patient with XIIIA deficiency. The recombinant Y283C protein was labile when expressed in MEG-01 cells, which can endogenously synthesize XIIIA. We also included two other mutants, G562R and I464stop, previously characterized in a non-XIIIA-producing cell line. All these mutants exhibited decreased thermostability and resistance against proteolytic digestion when compared with the wild-type. Gel-filtration analysis revealed that the mutants were in monomer form, while the wild-type formed a dimer. These results were consistent with the prediction by molecular modeling that the mutant molecules would be misfolded. Although assembly of a heterotetramer with XIIIB was demonstrated for Y283C, its binding ability was 10% that of the wild-type. No complex formation was observed for the G562R or I464stop mutants. The wild-type was stabilized in plasma by complex formation with XIIIB, resulting in an increased resistance against proteolytic digestion. In contrast, the mutants were unstable in plasma even in the presence of XIIIB. Thus, impaired folding, dimer formation, and heterotetramer assembly of the mutant XIIIAs lead to both intra- and extracellular instability, which must be responsible for XIIIA deficiency in the patient.  相似文献

Abstract:  Feeding behaviour of parthenogenetic apterae, post-teneral gynoparae and males of a holocyclic parthenogenetic lineage of Myzus persicae (Sulzer) (Hem., Aphididae) was studied on potato plants, Solanum tuberosum L. (Solanaceae) by DC-electrical penetration graph recording. Differences were observed between morphs during the 7-h recordings, specifically in the first phase of subcuticular probing and in xylem and phloem-related activities. The time required for the first penetration and first cell puncture of the non-vascular tissues was shorter in apterae than in gynoparae and males. Apterae more often showed phloem activities and sustained sap ingestion (100%) than winged morphs ( ca. 25%). In addition, phloem ingestion was much longer in apterae than in winged morphs, 3458 s and 25–70 s respectively. In contrast, winged morphs spent 14–21% of the available time ingesting sap from xylem while in apterae xylem sap uptake was only for 2% of the time. Furthermore, the data showed that males perform in general all the feeding behaviours that other conspecific morphs exhibit. The differences in feeding behaviour among morphs of M. persicae are discussed in relation to host specialization and to their possible involvement in non-persistent transmission of viruses.  相似文献

A sensitive stereoselective HPLC method was developed for determination of mefloquine (MFQ) enantiomers in plasma, urine and whole blood. The assay involved liquid-liquid extraction of MFQ from biological fluids with a mixture of hexane and isopropanol in the presence of sodium hydroxide and derivatization of the residue by (+)-(S)-naphthylethylisocyanate (NEIC) as chiral derivatizing reagent. Separation of the resulting diastereomers was performed on a silica normal-phase column using chloroform-hexane-methanol (25:74:1) as the mobile phase with a flow-rate of 1 ml/min. Using 200 μl of plasma or whole blood, the limit of determination was 0.2 μg/ml with UV detection for both enantiomers. The limit of determination in 500 μl of urine was 0.08 μg/ml with UV detection.  相似文献

The present study investigates the possible role of exogenous nitric oxide (NO) supplementation as sodium nitroprusside (SNP), on increasing phytoextraction and phytoremediation ability of arsenic hyperaccumulator, Isatis cappadocica. Arsenate (500, 1000 and 1500 µM) alone or in combination with 200 µM SNP was given to hydroponically grown plants for 14 days. The highest level of arsenate (1500 µM) reduced the plant growth and chlorophyll content, while SNP alleviated these inhibitory effects. The application of SNP significantly increased the As concentration in the root (from 1004 to 1943 mg/kg) and shoots (from 1304 to 1859 mg/kg) compared with As-stressed plants. However, SNP treatment did not affect translocation factor value significantly in the As-stressed plants which shows enhancement of both As uptake and translocation under SNP application. This is the first study demonstrating the favorable effects of SNP on As tolerance, uptake, and accumulation of highly valuable As hyperaccumulator, I. cappadocica.  相似文献

Covalent cross-linking of fibrin chains is required for stable blood clot formation, which is catalyzed by coagulation factor XIII (FXIII), a proenzyme of plasma transglutaminase consisting of catalytic A (FXIII-A) and non-catalytic B subunits (FXIII-B). Herein, we demonstrate that FXIII-B accelerates fibrin cross-linking. Depletion of FXIII-B from normal plasma supplemented with a physiological level of recombinant FXIII-A resulted in delayed fibrin cross-linking, reduced incorporation of FXIII-A into fibrin clots, and impaired activation peptide cleavage by thrombin; the addition of recombinant FXIII-B restored normal fibrin cross-linking, FXIII-A incorporation into fibrin clots, and activation peptide cleavage by thrombin. Immunoprecipitation with an anti-fibrinogen antibody revealed an interaction between the FXIII heterotetramer and fibrinogen mediated by FXIII-B and not FXIII-A. FXIII-B probably binds the γ-chain of fibrinogen with its D-domain, which is near the fibrin polymerization pockets, and dissociates from fibrin during or after cross-linking between γ-chains. Thus, FXIII-B plays important roles in the formation of a ternary complex between proenzyme FXIII, prosubstrate fibrinogen, and activator thrombin. Accordingly, congenital or acquired FXIII-B deficiency may result in increased bleeding tendency through impaired fibrin stabilization due to decreased FXIII-A activation by thrombin and secondary FXIII-A deficiency arising from enhanced circulatory clearance.  相似文献
