Hydroxy radicals induce cross-linking between phenylalanine (Phe) and 2-deoxyribose (dR) via formation of corresponding free radical intermediates. The cross-linked products were separated and identified by capillary gas chromatography-mass spectrometry. When phenylalanine and 2-deoxyribose radicals were generated in a 1:1 ratio, the predominant interaction was between Phe and dR radicals while the Phe-Phe and dR-dR cross-links were less abundant. The newly discovered cross-link between 2-deoxyribose and phenylalanine may serve as a model for radiation or free radical induced cross-linking between DNA and proteins and in general between sugar moieties and amino acids.  相似文献

OH radicals generated by ionizing radiation in aqueous solutions of thymine (T) and phenylalanine (Phe) induce crosslinking between thymine and phenylalanine. The crosslinked products were isolated and characterized by capillary gas chromatography-mass spectrometry. They are formed via OH radical adducts to thymine and phenylalanine and the reaction between dissimilar radicals is greatly favoured (T-Phe:Phe-Phe:T-T = 0.46:0.14:0.05). The reaction mechanism presented may serve as a model for radiation or any free radical-induced crosslinks between DNA and proteins.  相似文献

Regression analysis of the spontaneous spike activity of neurons in Helix pomatia was carried out with the aim to establish the statistical parameters of this activity under constant experimental conditions and during longer time intervals. The activity of 38 randomly chosen neurons in visceral and parietal ganglia, penetrated by microelectrodes and activated either endogenously by pacemaker potentials or by synaptic inputs, was recorded during time intervals lasting from 20 min to 3 h. The main results of the statistical analyses are presented in the table where the parameters of both cell types are listed. The validity of the regression analysis applied here is discussed from the point of the possibility it offers for carrying on the data processing quickly and without applying complex calculating means. The results are also considered regarding the current interest of our research group.  相似文献

The antioxidant capacity of 5-hydroxy-tryptophan. 5-hydroxy-indole. and DOPA (3,4-dihydroxy-phenyI-alanine) was tested in the Fe-induced lipid peroxidation of liver microsomes of normal- and vitamin E-deficient rats, using ascorbate as a reductant. Lipid peroxidation was monitored as low-level chemilu-minescence, indicative of generation of electronically-excited states arising from the recombination of secondary lipid peroxyl radicals.  相似文献

Endogenous oxidative processes are shown to generate hydrogen peroxide and .OH radicals, which react in vivo to form a variety of products. Thymidine glycol (Tg) and 8-hydroxydeoxyguanosine (8-dRG-OH) are such products. They result from the excision repair of DNA and are excreted in urine. Both products can be used as biomarkers in the dosimetry of oxidative damage to DNA. Since oxidative processes and accumulation of their effects contribute to carcinogenesis, the proposed rate-of-damage hypothesis provides a rationale for using these biomarkers in early diagnostics and in the assessment of carcinogenic and anticarcinogenic properties of diets, foods, and food components, as well as certain exogenous toxicants and agents. Approaches for measurement of urinary biomarkers of DNA damage are reviewed.  相似文献

Articular cartilage injuries are a common source of joint pain and dysfunction. We hypothesized that pulsed electromagnetic fields (PEMFs) would improve growth and healing of tissue-engineered cartilage grafts in a direction-dependent manner. PEMF stimulation of engineered cartilage constructs was first evaluated in vitro using passaged adult canine chondrocytes embedded in an agarose hydrogel scaffold. PEMF coils oriented parallel to the articular surface induced superior repair stiffness compared to both perpendicular PEMF (p = .026) and control (p = .012). This was correlated with increased glycosaminoglycan deposition in both parallel and perpendicular PEMF orientations compared to control (p = .010 and .028, respectively). Following in vitro optimization, the potential clinical translation of PEMF was evaluated in a preliminary in vivo preclinical adult canine model. Engineered osteochondral constructs (∅ 6 mm × 6 mm thick, devitalized bone base) were cultured to maturity and implanted into focal defects created in the stifle (knee) joint. To assess expedited early repair, animals were assessed after a 3-month recovery period, with microfracture repairs serving as an additional clinical control. In vivo, PEMF led to a greater likelihood of normal chondrocyte (odds ratio [OR]: 2.5, p = .051) and proteoglycan (OR: 5.0, p = .013) histological scores in engineered constructs. Interestingly, engineered constructs outperformed microfracture in clinical scoring, regardless of PEMF treatment (p < .05). Overall, the studies provided evidence that PEMF stimulation enhanced engineered cartilage growth and repair, demonstrating a potential low-cost, low-risk, noninvasive treatment modality for expediting early cartilage repair.  相似文献
