The Dipole Model and Phase Transitions in Biological Membranes

Assuming the dipole model for a membrane, approximate calculations are made which employ a dipole-dipole interaction energy. The calculations are based upon the assumption of cooperative coupling of membrane polar molecules and make use of the Bragg-Williams approximation. A theoretical estimate is made of the critical temperature at which phase changes might occur in certain biological membranes. Proposals are presented which explain how the dipole transition might relate to the sometimes observed thermal phase transitions in biological membranes.     

Nusinersen treatment and cerebrospinal fluid neurofilaments: An explorative study on Spinal Muscular Atrophy type 3 patients

The antisense oligonucleotide Nusinersen has been recently licensed to treat spinal muscular atrophy (SMA). Since SMA type 3 is characterized by variable phenotype and milder progression, biomarkers of early treatment response are urgently needed. We investigated the cerebrospinal fluid (CSF) concentration of neurofilaments in SMA type 3 patients treated with Nusinersen as a potential biomarker of treatment efficacy. The concentration of phosphorylated neurofilaments heavy chain (pNfH) and light chain (NfL) in the CSF of SMA type 3 patients was evaluated before and after six months since the first Nusinersen administration, performed with commercially available enzyme-linked immunosorbent assay (ELISA) kits. Clinical evaluation of SMA patients was performed with standardized motor function scales. Baseline neurofilament levels in patients were comparable to controls, but significantly decreased after six months of treatment, while motor functions were only marginally ameliorated. No significant correlation was observed between the change in motor functions and that of neurofilaments over time. The reduction of neurofilament levels suggests a possible early biochemical effect of treatment on axonal degeneration, which may precede changes in motor performance. Our study mandates further investigations to assess neurofilaments as a marker of treatment response.     

Lipido‐sterolic extract of Serenoa repens (LSESr,Permixon®) treatment affects human prostate cancer cell membrane organization

The molecular mechanism by which the lipido‐sterolic extract of Serenoa repens (LSESr, Permixon®) affects prostate cells remains to be fully elucidated. In androgen‐independent PC3 prostate cancer cells, the LSESr‐induced effects on proliferation and apoptosis were evaluated by counting cells and using a FACScan cytofluorimeter. PC3 cells were stained with JC‐1 dye to detect mitochondrial membrane potential. Cell membrane lipid composition was evaluated by thin layer chromatography and gas chromatographic analysis. Akt phosphorylation was analyzed by Western blotting and cellular ultrastructure through electron microscopy. LSESr (12.5 and 25 µg/ml) administration exerted a biphasic action by both inhibiting proliferation and stimulating apoptosis. After 1 h, it caused a marked reduction in the mitochondrial potential, decreased cholesterol content and modified phospholipid composition. A decrease in phosphatidylinositol‐4,5‐bisphosphate (PIP2) level was coupled with reduced Akt phosphorylation. After 24 h, all of these effects were restored to pre‐treatment conditions; however, the saturated (SFA)/unsaturated fatty acid (UFA) ratio increased, mainly due to a significant decrease in ω6 content. The reduction in cholesterol content could be responsible for both membrane raft disruption and redistribution of signaling complexes, allowing for a decrease of PIP2 levels, reduction of Akt phosphorylation and apoptosis induction. The decrease in ω6 content appears to be responsible for the prolonged and more consistent increase in the apoptosis rate and inhibition of proliferation observed after 2–3 days of LSESr treatment. In conclusion, LSESr administration results in complex changes in cell membrane organization and fluidity of prostate cancer cells that have progressed to hormone‐independent status. J. Cell. DOME/GALT type adenocarcimoma of the colon: a case report,literature review and a unified phenotypic categorization

Several types of colorectal cancers are associated with a prominent lymphoid component, which is considered a positive prognostic factor.We report a case of a dome-type carcinoma of the cecum in a 57 year old female.The sessile, non-polypoid lesion histologically consisted of a tubulovillous adenoma with low-grade dysplasia.The submucosal invasive component showed low-grade architectural features that included cystically dilated glands containing eosinohilic debris. Immunohistochemical studies displayed retention of the four mistmach repair proteins, consistent with a stable phenotype. After 3 years, the patient remains free of recurrence.A literature review highlighted striking similarities between dome-type carcinoma and the gut-associated lymphoid tissue carcinoma, the two sharing an intimate association with the gut associated lymphoid tissue.The two variants might therefore be grouped into a unified category.     

Synthesis and characterization of two novel cobalt (II) phosphine complexes: crystal structures of [CoCl3(Cy2PCH2PCy2H)] and [Co(NO3)2(Cy2PCH2PCy2O)]. Cy = cyclohexyl, C6H11

The complexes [(Cy₂PCH₂PCy₂H)CoCl₃] (1) and [(Cy₂PCH₂PCy₂O)Co(NO₃)₂] (2), Cy = cyclohexyl (C₆H₁₁), have been prepared and characterized by EPR, UV-Vis, microanalysis and X-ray crystallography. The reaction CoCl₂ · 6H₂O and dcpm, dcpm = bis(dicyclohexylphosphino)methane, was found to form the monomeric, four coordinate, thermochromic and paramagnetic complex [(Cy₂PCH₂PCy₂H)CoCl₃] (1). Of particular interest is the formation of a zwitterion, or inner salt, in which the dangling phosphine adds a hydrogen atom, giving the phosphorus a +1 formal charge. The molecule adopts a pseudo-tetrahedral geometry around the central cobalt atom to which the cyclohexyl groups bind in an equatorial fashion to the phosphine. The reaction of Co(NO₃)₂ · 6H₂O and dcpm in a toluene/methanol/methylene chloride mixture yields the pseudo-octahedral complex [(Cy₂PCH₂PCy₂O)Co(NO₃)₂] (2). The cobalt is in the +2 oxidation state with one of the phosphorus atoms again having a +1 formal charge. The complex adopts a pseudo-octahedral geometry around the central cobalt atom with the cyclohexyl groups binding in an equatorial fashion to the phosphine similar to [(Cy₂PCH₂PCy₂H)CoCl₃].
