The organic non-crystalline medium of 5,6-dichloro-2-[[5,6-dichloro-1-ethyl-3-(4-sulfobutyl)-benzimidazol-2-ylidene]-propenyl]-1-ethyl-3-(4-sulfobutyl)-benzimidazolium hydroxide (TDBC) is emerging as possible alternative plasmonic material for noble metal in visible region. In this paper, a novel long-range surface exciton-polariton (LRSEP) sensor based on TDBC film covered with graphene is reported. To enhance the imaging sensitivity, the thickness of TDBC film and the number of graphene layers are optimized. The result shows that the optimized imaging sensitivity is enhanced to 3243 RIU−1 when ns = 1.34. Compared with the traditional noble metal film-based sensor, the proposed LRSEP sensor demonstrates that the imaging sensitivity has been greatly improved. This is the first study of the TDBC film-based LRSEP sensor, which we hope to support the potential development of chemical sensing and bio-sensing.

Sleep adaptation in an unfamiliar environment, the so-called “first-night effect”, is known to occur in healthy individuals. To avoid the confounding effects of the “first-night effect”, the first-night sleep data are not used in most of sleep studies. In the present study, we examined changes of sleep adaptation in hospitalized patients with depression. Polysomnographic recordings were obtained for two consecutive nights from 14 patients, and sleep parameters were compared between both nights. Total sleep time, sleep latency, awakening times, movement awakening time, sleep efficiency, sleep architecture, rapid eye movement (REM) sleep latency, REM intensity, REM density, REM time, REM cycles, and other indicators showed no significant difference (p > 0.05) between the first and second nights. To conclude, hospitalized patients with depression have relatively less change in sleep adaptation, thus, the data from their first night do not need to be discarded.  相似文献

BAX is an important proapoptotic protein of the BCL-2 family, and its stability is essential for the regulation of the mitochondrial apoptotic pathway. A previous study revealed that BAX could undergo degradation through the ubiquitin-proteasome pathway. In this study, we identified two lysine sites, K21 and K123, that were critical ubiquitin-binding sites in BAX. Mutation of these two sites prolonged the half-life of BAX and also affected its proapoptotic ability. Intriguingly, we found that ABT-737, a BCL-2 inhibitor, significantly enhanced TRAIL-induced BAX degradation in HCT116 cells and increased TRAIL-induced apoptosis in the HCT116 only with the BAX K21R/K123R mutant, not other BAX mutants. In addition, overexpression of PARKIN, an E3 ubiquitin ligase targeting BAX, dramatically decreased BAX protein level when only treated with ABT-737 in HCT116 cells. Therefore, we speculated that BAX activation is essential for its ubiquitin-dependent degradation.  相似文献

KIAA1377 has been found to be linked with lymph node metastasis in esophageal squamous cell carcinoma (SCC) in our previous study; however, the regulation of KIAA1377 remains far from understood. Herein, to understand the regulation of KIAA1377 from the angle of microRNA (miRNA)–messenger RNA (mRNA) modulation in the setting of SCC cells, the basal level of KIAA1377 was determined by quantitative real‐time polymerase chain reaction (qRT‐PCR) and western blot analysis in KYSE‐150 and HeLa cells; biological roles of KIAA1377 contributing in the proliferation, migration, and invasion were evaluated using 3‐(4,5‐dimethyl‐2‐thiazolyl)‐2,5‐diphenyl‐2H‐tetrazolium bromide (MTT), wound‐healing and Transwell assays, respectively, after KIAA1377 was knocked out mediated by the CRISPR‐Cas9 system. Bioinformatic prediction revealed that let‐7b‐5p was a putative miRNA regulating KIAA1377, which was ensuingly validated by the luciferase reporter assay; after which, variation of KIAA1377 expression was further verified by qRT‐PCR and western blot analysis. Moreover, the biological roles of let‐7b‐5p in proliferation, migration, and invasion of KYSE‐150 and HeLa cells were also evaluated. It was exhibited that KIAA1377 was able to promote the proliferation and motility of both KYSE‐150 and HeLa cells, which can be reverted by re‐expression of let‐7b‐5p. The luciferase reporter assay verified that let‐7b‐5p can diametrically target KIAA1377. Collectively, our data demonstrated that let‐7b‐5p can directly but negatively regulate KIAA1377 in SCC cell lines, Ecal109, and HeLa cells.  相似文献

Recent studies have demonstrated pleiotropic roles of pyruvate kinase isoenzyme type M2 (PKM2) in tumor progression. However, the precise mechanisms underlying the effects of PKM2 on esophageal squamous cell carcinoma (ESCC) metastasis and transforming growth factor β1 (TGF-β1)-induced epithelial-mesenchymal transition (EMT) remain to be established. In this study, we observed upregulation of PKM2 in ESCC tissues that was markedly associated with lymph node metastasis and poor prognosis. High PKM2 expression in tumor tissues frequently coincided with the high pSTAT3Tyr705 expression and low E-cadherin expression. Furthermore, altered PKM2 expression was significantly associated with proliferation, migration, and invasion of ESCC cells, in addition to expression patterns of EMT markers (Snail, E-cadherin, and vimentin) and pSTAT3Tyr705/STAT3 ratio. Overexpression of STAT3 significantly attenuated the effects of PKM2 knockdown on cell proliferation and motility as well as expression of pSTAT3 Tyr705 and EMT markers. Consistently, stable short hairpin RNA (shRNA)-mediated silencing of PKM2 reversed the effects of TGF-β1 treatment, specifically, upregulation of PKM2, phosphorylation of STAT3 at Tyr705, and increased EMT, migration, and invasion. We propose that PKM2 regulates cell proliferation, migration, and invasion via phosphorylation of STAT3 through TGF-β1-induced EMT. Our findings collectively provide mechanistic insights into the tumor-promoting role of PKM2, supporting its prognostic value and the therapeutic utility of PKM2 inhibitors as potential antitumor agents in ESCC.  相似文献

A novel ligand‐assisted assembly approach is demonstrated for the synthesis of thermally stable and large‐pore ordered mesoporous titanium dioxide with a highly crystalline framework by using diblock copolymer poly(ethylene oxide)‐b‐polystyrene (PEO‐b‐PS) as a template and titanium isopropoxide (TIPO) as a precursor. Small‐angle X‐ray scattering, X‐ray diffraction (XRD), transmission electron microscopy (TEM), high‐resolution scanning electron microscopy, and N2‐sorption measurements indicate that the obtained TiO2 materials possess an ordered primary cubic mesostructure with large, uniform pore diameters of about 16.0 nm, and high Brunauer–Emmett–Teller surface areas of ~112 m2 g?1, as well as high thermal stability (~700 °C). High resolution TEM and wide‐angle XRD measurements clearly illustrate the high crystallinity of the mesoporous titania with an anatase structure in the pore walls. It is worth mentioning that, in this process, in addition to tetrahydrofuran as a solvent, acetylacetone was employed as a coordination agent to avoid rapid hydrolysis of the titanium precursor. Additionally, stepped evaporation and heating processes were adopted to control the condensation rate and facilitate the assembly of the ordered mesostructure, and ensure the formation of fully polycrystalline anatase titania frameworks without collapse of the mesostructure. By employing the obtained mesoporous and crystallized TiO2 as the photoanode in a dye‐sensitized solar cell, a high power‐conversion efficiency (5.45%) can be achieved in combination with the N719 dye, which shows that this mesoprous titania is a great potential candidate as a catalyst support for photonic‐conversion applications.  相似文献

粤东三个中型水库富营养化特征分析   总被引:5,自引:0,他引:5  
于2000年的丰水期和枯水期,对粤东三个中型水库:合水水库、河溪水库、沙田水库进行了富营养化特征进行了调查与分析。结果表明:三个水库多数采样点处于中营养阶段;总氮在0.16~1.58mg·L~(-1)范围内,总磷为0.034~0.15mg·L~(-1)叶绿素则为0.732~6.57mg·m~(-3);浮游植物大部分是中污性至寡污性的种类,以蓝藻、绿藻种类占优势,常见种是微囊藻(Microcystis sp.)、颗粒直链藻(Melosira granulata)、梅尼小环藻(Cyclotella meneghiniana)。合水水库和河溪水库的富营养化水平较高,TSI指数在40左右,丰水期的富营养化水平高于枯水期;而沙田水库的富营养化水平相对要低,TSI指数在30左右,丰水期的富营养化水平要低千枯水期。说明这三个水库受流域的影响程度不同。  相似文献
