The intermediate filament protein nestin is characterized by its specific expression during the development of neuronal and myogenic tissues. We identify nestin as a novel in vivo target for cdk5 and p35 kinase, a critical signaling determinant in development. Two cdk5-specific phosphorylation sites on nestin, Thr-1495 and Thr-316, were established, the latter of which was used as a marker for cdk5-specific phosphorylation in vivo. Ectopic expression of cdk5 and p35 in central nervous system progenitor cells and in myogenic precursor cells induced elevated phosphorylation and reorganization of nestin. The kinetics of nestin expression corresponded to elevated expression and activation of cdk5 during differentiation of myoblast cell cultures and during regeneration of skeletal muscle. In the myoblasts, a disassembly-linked phosphorylation of Thr-316 indicated active phosphorylation of nestin by cdk5. Moreover, cdk5 occurred in physical association with nestin. Inhibition of cdk5 activity-either by transfection with dominant-negative cdk5 or by using a specific cdk5 inhibitor-blocked myoblast differentiation and phosphorylation of nestin at Thr-316, and this inhibition markedly disturbed the organization of nestin. Interestingly, the interaction between p35, the cdk5 activator, and nestin appeared to be regulated by cdk5. In differentiating myoblasts, p35 was not complexed with nestin phosphorylated at Thr-316, and inhibition of cdk5 activity during differentiation induced a marked association of p35 with nestin. These results demonstrate that there is a continuous turnover of cdk5 and p35 activity on a scaffold formed by nestin. This association is likely to affect the organization and operation of both cdk5 and nestin during development.  相似文献

We developed a method for studying the synchrony of behaviour based on calculations of overdispersion of a binomial process. The lying behaviour of cows was investigated under two different housing units inside the same barn. The first unit housed 30 cows undergoing conventional milking and the second unit housed 27 cows undergoing automatic milking. The lying behaviour of the cows was observed over 3 weeks in 12 periods of 6 h each. Every 5 min, we counted the number of cows lying down in the cubicles. As external cues, like feeding and conventional milking, can promote synchrony in dairy cows, we expected that cows conventionally milked would show more behavioural synchrony than automatically milked cows. Cows lied down synchronously in both units (overdispersion 1.67, P < 0.01). Lying synchrony tended to be slightly bigger in automatically than in conventionally milked cows (overdispersion 1.76 v. 1.58, P = 0.09), although the proportion of cows lying down was on average greater in conventionally than in automatically milked cows (60.7% v. 45.6%). This suggests that synchronized lying behaviour in cows is a constant phenomenon that depends on social facilitation rather than on external cues. The overdispersion index appears to be a useful tool for studying the synchrony of animal behaviour when observations are made at the group level.  相似文献

This study investigated the influence of stockperson's behaviour and housing conditions on calves' behavioural reactions to people, and behavioural and physiological reactions to handling and short transport. Sixty-four Finnish Ayrshire male calves were used; half of them were housed in individual pens, the other half were housed in group pens of two calves. In both housing conditions half of the calves received minimal contact from the stockperson, while the other half were stroked on their necks and shoulders for 90s a day, after milk meals. The effects of housing and contact with the stockperson on the responses of calves to people, either entering or approaching the pen, were studied. Furthermore, calves' behavioural and physiological (cortisol, heart rate) reactions to being loaded onto a truck, transported for 30min and unloaded were observed. When a person entered the home pen, calves housed by pairs took significantly more time to interact and interacted less frequently with the person than individually housed calves did (p<0.01). Calves that received additional contact interacted for longer time with the unfamiliar person than calves with minimal contact (p=0.02). When a person approached the front of the calves' pens, less withdrawal responses were shown by calves that had received additional contact (p<0.05) than those that had received minimal contact. When the calves were loaded onto the truck, it took more time and effort to load pair housed calves than individually housed calves (p<0.01) and less effort to load calves that had received additional contact (p<0.01) compared to those that had received minimal contact. During loading additional contact calves had lower heart rates (p<0.05) than those that had received minimal contact, while during transport pair housed calves had lower heart rates compared to individually housed ones (p<0.05). For all the observations performed, no interactions were found between housing conditions and human contact.It is concluded that, compared to calves housed individually, calves housed in pairs are less ready to approach humans and less easy to handle. Providing calves with regular positive contacts makes them less fearful of people and improves handling. Due to the greater difficulty in handling calves housed in groups, it is concluded that these animals need to have regular contact with humans.  相似文献

Vimentin is the major intermediate filament (IF) protein of mesenchymal cells. It shows dynamically altered expression patterns during different developmental stages and high sequence homology throughout all vertebrates, suggesting that the protein is physiologically important. Still, until recently, the real tasks of vimentin have been elusive, primarily because the vimentin-deficient mice were originally characterized as having a very mild phenotype. Recent studies have revealed several key functions for vimentin that were not obvious at first sight. Vimentin emerges as an organizer of a number of critical proteins involved in attachment, migration, and cell signaling. The highly dynamic and complex phosphorylation of vimentin seems to be a likely regulator mechanism for these functions. The implicated novel vimentin functions have broad ramifications into many different aspects of cell physiology, cellular interactions, and organ homeostasis.  相似文献

DCs express intrinsic cellular defense mechanisms to specifically inhibit HIV-1 replication. Thus, DCs are productively infected only at very low levels with HIV-1, and this non-permissiveness of DCs is suggested to go along with viral evasion. We now illustrate that complement-opsonized HIV-1 (HIV-C) efficiently bypasses SAMHD1 restriction and productively infects DCs including BDCA-1 DCs. Efficient DC infection by HIV-C was also observed using single-cycle HIV-C, and correlated with a remarkable elevated SAMHD1 T592 phosphorylation but not SAMHD1 degradation. If SAMHD1 phosphorylation was blocked using a CDK2-inhibitor HIV-C-induced DC infection was also significantly abrogated. Additionally, we found a higher maturation and co-stimulatory potential, aberrant type I interferon expression and signaling as well as a stronger induction of cellular immune responses in HIV-C-treated DCs. Collectively, our data highlight a novel protective mechanism mediated by complement opsonization of HIV to effectively promote DC immune functions, which might be in the future exploited to tackle HIV infection.  相似文献
