Emerald ash borer (Agrilus planipennis Fairmaire) (Coleoptera: Buprestidae) is a major stem borer of ash (Fraxinus spp.). It is univoltine in Tianjin, while it is semivoltine in Heilongjiang Province, and both univoltine and semivoltine in Changchun, Jilin Province, where the majority is univoltine. The longevity of emerald ash borer adults is 17.2 ± 4.6 days (n = 45), eggs 9.0 5:1.1 days (n = 103), univoltine larvae 308 days, semivoltine larvae 673 days, and pupae 61.2 ± 1.6 days (n = 45). It takes about 100 days from the time larvae bore into the phloem to when they complete the pupal cell. In a 10-year-old velvet ash (Fraxinus velutina Tort.) plantation in Tianjin, emerald ash borer preferred to oviposit on the regions of boles from 50-150 cm above ground, accounting for 76.7% of the total girdling. Girdling on the south side of the tree boles accounted for 43.40% of the total girdling. The emerald ash borer population density is higher at the edge of the plantation compared with the center.  相似文献

In an effort to improve the understanding of electron transfer mechanisms at the microbe–mineral interface, Shewanella oneidensis MR-1 mutants with in-frame deletions of outer-membrane cytochromes (OMCs), MtrC and OmcA, were characterized for the ability to reduce ferrihydrite (FH) using a suite of microscopic, spectroscopic, and biochemical techniques. Analysis of purified recombinant proteins demonstrated that both cytochromes undergo rapid electron exchange with FH in vitro with MtrC displaying faster transfer rates than OmcA. Immunomicroscopy with cytochrome-specific antibodies revealed that MtrC co-localizes with iron solids on the cell surface while OmcA exhibits a more diffuse distribution over the cell surface. After 3-day incubation of MR-1 with FH, pronounced reductive transformation mineral products were visible by electron microscopy. Upon further incubation, the predominant phases identified were ferrous phosphates including vivianite [Fe3(PO4)2·8H2O] and a switzerite-like phase [Mn3,Fe3(PO4)2·7H2O] that were heavily colonized by MR-1 cells with surface-exposed outer-membrane cytochromes. In the absence of both MtrC and OmcA, the cells ability to reduce FH was significantly hindered and no mineral transformation products were detected. Collectively, these results highlight the importance of the outer-membrane cytochromes in the reductive transformation of FH and support a role for direct electron transfer from the OMCs at the cell surface to the mineral.  相似文献

斑翅肩花蝽Tetraphleps galchanoides Ghauri是铁杉球蚜Adelges tsugae Annand (hemlock woolly adelgid) 的重要天敌昆虫。在云南省兰坪县天生桥林区通过实验室和林间的饲养观察与测定、林间线路调查等方法, 研究了斑翅肩花蝽的生物学特性、生境及食性选择,并描述各虫态形态特征。斑翅肩花蝽在该林区一年发生2代, 无世代重叠, 寿命长。第1代(5月下旬至10月下旬)卵期13.8±1.6 d,若虫期97.6±7.4 d,成虫期55.2±4.7 d;第2代(11月中旬至翌年4月下旬)卵期11.3±1.1 d,若虫期105.7±8.5 d,成虫期60.4±5.3 d。若虫共5龄,以5龄若虫在云南铁杉Tsuga dumosa枝条树皮裂缝下或枯枝落叶层内越冬。雌雄成虫性比8.5∶1,雌雄成虫飞行缓慢,其飞行活动主要受到交尾、产卵、捕食铁杉球蚜补充营养的影响,多在树冠下层活动。该林区的斑翅肩花蝽最适生境选择: 海拔为2 851~2 980 m,云南铁杉密度为13.3~15.5株/100 m、郁闭度为0.61~0.70。以寄主铁杉球蚜及附近常见的冷杉球蚜Aphrastasia pectinatae (Cholodkovsky)、华山松球蚜Pineus armandicola Zhang和落叶松球蚜Adelges laricis Vall为食物进行选择性试验,结果表明,斑翅肩花蝽对这4种球蚜的选择性存在显著差异,最喜好捕食铁杉球蚜,可成为生物防治铁杉球蚜的主要天敌之一。  相似文献

Abstract. Midgut protease activity in Glossina morsitans centralis and G. m. morsitans , at 48h post bloodmeal averaged 1.8IU of trypsin-like activity. These two tsetse subspecies differ in their susceptibility to trypanosome infection. Except for low levels in flies fed on waterbuck blood (0.7IU), activity did not differ in flies fed a variety of host bloods (goat, pig, cow, buffalo, eland) and trypanosome species ( Trypanosoma congolense, T.brucei, T.simiae ). Protease activity was also not correlated with infection rates, despite large differences in infection rates among experiments. Nevertheless, addition of 0.06M D(+)-glucosamine to parasitaemic blood resulted in a three-fold reduction in protease activity, coincident with a large increase in infection rate. This effect did not occur when parasites or D(+)-glucosamine were added alone to the bloodmeal, suggesting that the effect was due to metabolism of D(+)-glucosamine by parasites.  相似文献

A collection of 121 isolates of Rhizobium leguminosarum biovar (bv.) trifolii was obtained from root nodules of Trifolium subterraneum L. (subclover) plants growing in an established pasture. The collection consisted of a single isolate from each of 18 plants sampled from seven microplots. The following year, a further 28 and 27 isolates were collected from the first and seventh sampling points, respectively. Analysis of restriction fragment length polymorphisms (RFLPs) of both chromosomal and Sym (symbiotic) plasmid DNA and multilocus enzyme electrophoresis (MLEE) were used to assess the diversity, genetic relationships and structure of this population. Symbiotic effectiveness tests were used to examine the symbiotic phenotype of each isolate collected in the first year. Analysis of RFLPs of the first year isolates revealed 13 chromosomal types and 25 Sym plasmid types. Similar Sym plasmid types were grouped into 14 families containing 1–6 members. No new chromosomal types and six new Sym plasmid types were detected in the second year. The symbiotic effectiveness of the first year isolates of the same Sym plasmid type was similar. Significant differences in symbiotic effectiveness were detected between different Sym plasmid types in the same plasmid family. Representative isolates of each chromosomal type Sym plasmid type identified in the first year were analysed using multilocus enzyme electrophoresis. Mean genetic diversity per locus was high (0.559). Enzyme electrophoresis revealed 17 electrophoretic types (ETs). Ouster analysis of the enzyme data revealed large genetic diversity amongst the ETs. Strong linkage disequilibrium was observed for the population as a whole, i.e. clonal population structure, but significantly less disequilibrium was observed among a cluster of ETs suggesting that recombination occurred between ETs within the cluster. Our results revealed that a population of naturally occurring isolates of Rhizobium leguminosarum bv. trifolii can be genetically diverse and support the possibility that recombination plays a role in generating new genotypes.  相似文献

African Buffalo Serum Contains Novel Trypanocidal Protein   总被引:2,自引:0,他引:2  
ABSTRACT. The high ability of African buffalo, as compared to domestic cattle, to control infections with Trypanosoma brucei brucei IL Tat 1.4 organisms did not correlate with the timing or magnitude of parasite surface coat-specific antibody responses and may have resulted from the constitutive presence in buffalo blood of a novel trypanocidal factor. Buffalo plasma and serum contained material that killed bloodstream stage T. b. brucei, T. b. rhodesiense, T. b. gambiense, T. evansi, T. congolense , and T. vivax organisms during four h of incubation at 37° C in vitro. Serum from eland was also trypanocidal whereas serum from oryx, waterbuck, yellow-back duiker, cattle, horse, sheep, goat, mouse, rat, and rabbit was not trypanocidal. The buffalo serum trypanocidal material was not lipoprotein, or IgG, and had the following properties: 1) a density of < 1.24 g/ml determined by flotation ultracentrifugation; 2) insolubility in 50% saturated ammonium sulphate; 3) non-reactivity with anti-bovine IgM, and anti-bovine IgG; 4) non-reactivity with protein G, and protein A; 5) a relative molecular mass of 152 kDa determined by chromatography on Sephacryl S 300, and of 133 kDa determined by chromatography of the 50% SAS cut of IgG-depleted buffalo serum on Superose 12; 6) no associated cholesterol; and 7) inactivation by digestion with proteinase K that was immobilized on agarose.  相似文献
